    Notes: Summary The effect of D-mannose and D-glucose on bacteriuria due to Escherichia coli with mannose-sensitive adhesins was investigated in adult male Sprague-Dawley rats undergoing diuresis. Inocula of 105, 107, or 108 bacteria in 0.1 ml of normal saline or 2.5% or 10% D-mannose or D-glucose were injected intravesically and urine was cultured 1, 3, 5, 7 and 9 days later. The levels of bacteriuria on days 1 and 5 were significantly lower in rats inoculated with 105 E. coli and 10% D-mannose than in controls (p〈0.05 and 0.01 respectively) and the percentages of rats with 〈 100 bacteria/ml were higher on days 1 and 3 (p=0.05 and 0.02 respectively). Bacteriuria was significantly lower in rats inoculated with 107 bacteria and 10% D-mannose than in controls on days 5 and 7 (p〈0.01 for each day) and the percentage of rats with 〈100 bacteria/ml was higher on day 7 (p=0.01). D-glucose reduced bacteriuria significantly only with a concentration of 10% after instillation of 105 E. coli (p〈0.05, day 1). The results indicate that D-mannose and D-glucose can significantly reduce bacteriuria within 1 day and that their efficacy is dependent upon the concentration of both saccharide and bacteria.

    Notes: Summary Previous studies have demonstrated that after intravesical inoculation of Escherichia coli, rats drinking 5% glucose-water remained bacteriuric for up to 21 days while rats drinking tap water became abacteriuric within a few days. To facilitate accurate monitoring of bacteriuria, we created a ventral bladder hernia for percutaneous aspiration of urine. After intravesical inoculation with 108 E. coli, rats with ventral bladder hernias demonstrated clearance of bacteria at a rate comparable to that observed in rats with intrapelvic bladders (p〈0.01). Of rats drinking tap water, 6 of 7 (85%) with intrapelvic bladders and 8 of 9 (89%) with ventral hernias had less than 10 colony forming units per ml of urine within 9 days of inoculation. Of rats drinking 5% glucose-water, 4 of 5 (80%) with intrapelvic bladders and 6 of 8 (75%) with ventral bladder hernias had 〉105 colony forming units per ml of urine 9 days after inoculation. The results suggest that this technique does not alter the antibacterial response of control or polyuric rats to E. coli inoculated intravesically.

    Notes: Summary The anther culture technique is useful for the recovery of haploids which when doubled provide homozygosity. Additionally, beneficial as well as deleterious genetic and epigenetic changes are promoted by the in vitro procedures. The majority of plants recovered from anther calli of the cultivar ‘Calrose 76’ were similar to the starting cultivar but plants regenerated from microspore calli had a wider range of responses than controls for several characteristics. Plants with larger seeds, higher levels of seed protein, shorter stature and more highly tillered than the starting cultivar were obtained from selfed anther-derived plants. The data also support the concept that in vitro procedures including anther culture of specific cultivars of rice promoted the recovery of phenotypes with increased seed storage proteins.

    Notes: Summary Frequency of callus formation in wheat (Triticum aestivum L. em. Thell) anthers cultured in vitro and the frequency of subsequent plantlet formation from such calli were examined in a diallel population produced from five inbred spring wheat cultivars. Two of the five cultivars were believed to possess relatively high frequencies of response and the other three relatively low response frequencies, based on previous studies. General and specific combining abilities were estimated and found to be highly significant for both traits. Reciprocal effects were also estimated and were highly significant for both traits. Of the 25 entries, the largest mean callus formation frequency was observed on anthers of ‘Kitt’ x ‘Olaf’, while the largest mean plantlet formation frequency was observed using anthers of the cultivar, ‘Fielder’. No significant correlation was observed between the two traits. Heritability estimates in the range of 0.6–0.7 suggested, however, that both traits were highly heritable, so that rapid gain from selection for these traits should be possible. Current limitations due to genetic variation in responses therefore may not constitute a major obstacle to application of in vitro techniques by wheat breeders.

    Notes: Summary The effects of long-term tissue culture on mitochondrial DNAs were examined using rice (Oryza sativa) cell suspension cultures. Mitochondrial DNAs were isolated from P. I. 353705 (an indica subspecies of rice similar to ‘Asam 5’), its anther-culture-derived line BL2 (an 8-year-old cell suspension culture), and five other cell lines (A1, A7, A11, A13, and A23), also derived from BL2 and independently selected for resistance to the lysine analog, S-(2-amino)-ethyl-L-cysteine. Mitochondrial DNAs of the rice lines were digested with ten restriction endonucleases (BamHI, BglII, EcoRI, EcoRV, HindIII, PstI, PvuII, SalI, SmaI, and XhoI), electrophoresed, and transferred to nylon membranes. Southern blots were hybridized with one rice and five maize probes containing mitochondrial genes. The restriction patterns of ten Southern blots and hybridization patterns of 60 endonuclease/probe combinations were analyzed. DNAs from all sources produced unique restriction patterns when digested with HindIII or BglII; with the other endonucleases an array of similarities and differences was observed. Lines BL2 and A11 showed unique patterns with all restriction endonucleases tested. No hybridization pattern differences were observed among the lines when probes containing apt9 and atpA were used. However, extensive hybridization pattern differences were observed with coxI, coxII, rrn18-rrn5, and atp6 probes. Both restriction and hybridization patterns revealed variation due to tissue culture effect. Coxll was most efficient in revealing the uniqueness of BL2. Among the analog selected lines A11 was most divergent, and probes rrn18-rrn5 and atp6 were most efficient in revealing its distinctiveness. Unique mitochondrial genomic organizations were found to be associated with long-term tissue culture.

    Notes: Summary Under greenhouse conditions the dark green wild type (su/su) tobacco grows 2–3 times faster than the yellow mutant (Su/su) and contains five-fold more chlorophyll. On a fresh weight basis, however, both genotypes contain similar amounts of RuBPCase and fraction 11 protein in approximately equal proportion and have similar levels of 70s and 80s ribosomes. When seedlings are cultured on agar medium supplemented with sucrose and equal concentrations of IAA and kinetin or kinetin alone, a drastic reduction of RuBPCase and free 70s ribosomes, but not of chlorophyll content, were observed. Moreover, albino (Su/Su) seedlings developed on supplemented media still contain appreciable amounts of RuBPCase and free 70s ribosomes although chlorophyll levels are extremely low indicating no correlation between RuBPCase and chlorophyll content. RuBPCase crystallized from both wild type and yellow mutant plants seem to have identical composition and structure when examined by isoelectric focusing, amino acid analysis or peptide mapping techniques. The slow-growing yellow mutant is apparently deficient only in chlorophyll of the light harvesting chlorophyll-protein complex but with no alteration of the protein moiety or chlorophyll a/b ratio.

    Notes: Summary Chloroplast (ct) and mitochondrial (mt) DNAs were isolated from two subspecies of rice (Oryza sativa), japonica (Calrose 76) and indica (PI353705) and compared by restriction endonuclease fragment pattern analysis. Similarly, PI353705 (A5) mtDNA was also compared with the mtDNA of its long term tissue cultured line, BL2. Variation in the ctDNA of the 2 subspecies was detected with two (AvaI and BglI) of the 11 restriction endonucleases tested, whereas their mtDNAs showed considerable variation when restricted by PstI, BamHI, HindIII and XhoI endonucleases. Thus, the chloroplast DNA was more highly conserved than the mtDNA in the subspecies comparisons. Only minor variation was observed between the restriction endonuclease patterns of the mtDNAs of BL2 and A5. Southern blots of mtDNA were hybridized with heterologous probes from maize and spinach organelle genes. Differences were found in the hybridization patterns of the two subspecies for six of the eight (mitochondrial and chloroplast) probes tested. Two of the seven (mitochondrial) probes (coxII and 26S rRNA) detected tissue culture generated variation in mtDNA. The relative values of restriction endonuclease and hybridization patterns for studying phylogenetic and genetic relationships in rice are discussed.

    Notes: In recent years air quality and atmospheric investigations have become very important topics. This results from a new legal basis for environmental protection which generates the need for the accurate measurement of ambient conditions. Despite the importance of these investigations, detailed reviews characterizing the existing analytical GC systems are scarce. The author has attempted to remedy this unsatisfactory situation by presenting a comprative review of the principal commercially available systems as well as some homemade set-ups. No single method will be optimal for all purposes. The system of choice will depend on the particular application and on the quality and composition of the sample. This review does not attempt to cover all existing systems, but some exotic or lesser known systems have also been included.
