    Notes: Summary The bone resorptive factor and amphipathic antigen (AcA) previously identified by us in preparations fromActinomyces viscosus have been partially purified, characterized chemically, and compared. They elute at the same location on chromatography with Ac 22. The fatty acid composition of AcA and the bone resorptive factor is the same. Some differences in carbohydrate composition are observed. TheActinomyces factor does not affect calcium influx or cyclic AMP in isolated bone cells. Therefore it is concluded that AcA stimulates resorption either by gaining entrance into bone cells or by way of a yet undetermined second messenger.

    Notes: Abstract Changes in biomass and chemical composition, and the reproductive phenology ofPorphyra columbina Mont. were monitored at three sites in southern New Zealand over two growing seasons. Both temporal and spatial variations were found. Seasonal changes in biomass and chemical components were correlated with seawater nitrate concentrations and temperature. The summer decline in biomass was a result of the onset of unsuitable environmental conditions and the release of reproductive tissue. Under more suitable conditions, the decline in biomass was delayed. There was an inverse relationship between vegetative growth and reproduction. Reproductive plants first appeared in August at a time of increasing temperature, irradiance and daylength. Only larger plants which were mainly found in subsites low on the shore became reproductive. Plants sampled from high subsites had a shorter growth season, were generally smaller, had lower nitrogen and pigment content and were non-reproductive.

    Notes: Abstract Tissue samples ofMacrocystis pyrifera from 2 sites in southern New Zealand and harvested over a period of 12 months were analysed for alginate content and composition (M:G ratio). Plants were divided into three frond classes of different length and each frond was further separated into age categories of blades and stipes (viz young, mature and old blades; mature and old stipes). Within each size class, younger blades had higher alginate content than older blades. Stipes did not show such variation with age. Alginate from younger blades and stipes had higher proportions of mannuronic acid residues than those from old blades and stipes. The range of M:G ratios for age categories of either blades or stipes from longer fronds was greater than those for smaller fronds. Alginate content and M:G ratios of stipes were always higher than for blades. The difference between M:G ratios of blades and stipes was greater for smaller fronds than for longer ones. Differences between collection sites and seasonal trends are also discussed.

    Notes: We have applied a technique of interactive continuous selection (ICS) to the isolation of streptomycin-resistant mutants of the streptomycin-producing organism, Streptomyces griseus. A series of mutants, each with a different colonial morphology and expressing successively greater resistance to streptomycin, was isolated during the course of selection. Takeover of the mutants has been correlated with changes in on-line estimates of streptomycin concentration such that these estimates may be used as a real-time measure of the genetic state of the cell population. When grown in the medium employed for ICS, mutants expressed increased antibiotic production titers; the best mutant produced 10 to 20 times more streptomycin than the parent strain. Absolute improvements in the maximum specific growth rate and intrinsic resistance to streptomycin did not account for the observed growth advantage of all mutants. Rather, each mutant exhibited relative increases in specific growth rate at increasing concentrations of streptomycin. © 1996 John Wiley & Sons, Inc.

    Notes: Analysis of mutant human fibroblasts deficient in a cell surface receptor for low density lipoproteins (LDL) has led to the delineation of an important, hitherto unrecognized, regulatory process for cholesterol metabolism. In normal cells, binding of LDL to this receptor regulates cholesterol metabolism by two mechanisms: (a) suppression of cholesterol synthesis and (b) facilitation of the rate of proteolytic degradation of the lipoprotein. In cells from homozygotes with the autosomal dominant disorder Familial Hypercholesterolemia, a nearly total reduction in LDL receptors results in two secondary abnormalities: (a) overproduction of cholesterol due to an inability of LDL to suppress the activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase, the rate-controlling enzyme in cholesterol biosynthesis, and (b) impairment in the rate of proteolytic degradation of LDL. Cells from heterozygotes possess about 50 per cent of the normal number of LDL receptors; this leads to a concentrationdependent defect in regulation, so that attainment of rates of cholesterol synthesis and LDL degradation equal to that in normal cells requires a two to three-fold higher concentration of extracellular LDL in the heterozygote. The identification of this genetic regulatory defect in fibroblasts of heterozygotes with Familial Hypercholesterolemia makes available an in vitro system for studying the molecular mechanism by which a dominant mutation affects gene expression in mammalian cells.

    Notes: Specific binding of leukemia-inhibitory factor (LIF) to osteoblasts, but not multinucleated osteoclasts, was demonstrated by receptor autoradiography by *using cells isolated from newborn rat long bones. The clonal rat osteogenic sarcoma cells, UMR 106-06, which have several phenotypic properties of osteoblasts, expressed 300 LIF receptors per cell, with an apparent KD of 60 pM. Treatment of calvarial osteoblasts or UMR 106-01 cells with LIF resulted in a dose-dependent inhibition of plasminogen activator (PA) activity. Both calvarial osteoblasts and osteogenic sarcoma cells were shown by Western blotting and reverse fibrin autography to produce plasminogen activator inhibitor-1 (PAI-1), the production of which was increased by LIF treatment. Northern blot analysis revealed that LIF treatment resulted in a rapid (peak 1 hour), dose-dependent increase in mRNA for PAI-1. LIF treatment of the preosteoblast cell line, UMR 201, enhanced the alkaline phosphatase response of these cells to retinoic acid. Each of the osteoblast-like cell types (calvarial osteoblasts, UMR 106-06, and UMR 201) was shown to produce LIF by bioassay and, by using the polymerase chain reaction (PCR), was shown to express low levels of mRNA for LIF. These data establish that cells of the osteoblast lineage are targets for LIF action. The reported anabolic effects of this cytokine on bone formation in vivo could be related to inhibition of protease activity. LIF may be an important paracrine modulator in bone, or perhaps an autocrine one, based on the evidence for its production by osteoblasts and osteoblast-like cells.

    Notes: Severn Sound is a heavily used recreational and beating area in the southeast corner of Georgian Bay, Lake Huron, Canada. Because of the concern over the possible release of tributyltin species (TBT) from antifouling paints on boat hulls and marinas, surveys were carried out in 1989 and 1992 to determine the presence of this species and its degradation products dibutyltin (DBT) and monobutyltin (MBT) in this area. Many fish (pike and young-of-the-year spottail shiners) and sediment samples collected in 1989 contained detectable levels of TBT. A maximum concentration of TBT was recorded in northern pike in the spring to be 240 ng Sn g-1. Maximum levels occurred in marinas during the beginning of the boating season and significantly reduced during the summer and early autumn, although the maximum value of TBT in sediment (392 ng Sn g-1) was observed in the summer of 1989. The seasonal variation of TBT levels was further substantiated in the subsequent 1992 study, in which sediments from three areas in a marina were sampled at monthly intervals from May to October. TBT levels were much higher in May and then generally decreased with time. Mussels (Elliptio complanta) caged in the marina for three months also contained TBT. DBT was frequently detected in the sediments but less frequently in fish and mussels. MBT was generally below detection limits. Plants (macrophytes and cladophora) contained very small amounts of butyltin compounds.

    Notes: A cut or crush injury to a peripheral nerve results in the degeneration of that portion of the axon isolated from the cell body. The rapid degeneration of this distal segment was for many years believed to be a process intrinsic to the nerve. It was believed that Schwann cells both phagocytosed degenerating axons and myelin sheaths and also provided growth factors to promote regeneration of the damaged axons. In recent years, it has become apparent that the degenerating distal segment is invaded by monocytes from the blood. We will review the evidence that these recruited macrophages play a role in both degeneration and regeneration of peripheral nerve axons after injury and consider whether the slow degeneration and poor monocyte recruitment in the central nervous system may contribute to the poor regeneration there.
