    Beschreibung / Inhaltsverzeichnis: Zusammenfassung 24 Std nach i.v. Injektion von 100 mg/kg eines Pentosanschwefelsäureesters (SP 54) sind in der Nierenrinde von Ratten schwere degenerative Veränderungen der proximalen Tubuluszellen zu beobachten. Das Tubulusepithel ist flachkubisch umgewandelt, das Tubuluslumen ist angefüllt mit ausgestoßenen Mitochondrien und anderen Zelltrümmern. Am stärksten betroffen ist die Pars contorta des proximalen Tubulus. Weniger stark veränderte Tubuluszellen sind angefüllt mit Cytosomen, die eine spezifische Feinstruktur haben oder gehäuft gegenüber Kontrollen Cytoplasmaprotrusionen in das Tubuluslumen aufweisen. Die akute Tubulusdegeneration ist ein spezifischer Effekt von SP 54. Heparin bewirkt nur eine leichte Schwellung des Tubulusepithels, die häufiger auftritt als bei Kontrolltieren.
    Notizen: Summary 24 hrs after the intravenous injection of 100 mg/kg of a Pentosansulfuricacidester (SP 54) severe degenerative alterations are observed in the proximal tubular cells in rat kidney cortex. The tubular epithelium has changed to a single layer of elongated cubic cells. The tubular lumen is filled with cellular debris and extruded mitochondria. The greatest change is seen in the pars contorta of the proximal tubule. Less degenerated tubular cells have a great number of cytosomes with a specific substructure or have cytoplasmatic protrusions into the tubular lumen. This acute tubular degeneration is a specific event after the administration of SP 54 and is not seen after an equimolar dosis of Heparin. Heparin induces only a slight swelling of the tubular epithelium, more frequent seen as in the control animals.

    Notizen: Abstract Urinary excretion of glomerular basement membrane (GBM)-related peptides was analysed in 72 patients with a variety of renal diseases by immunoblotting using polyclonal antibodies against either collagenase or pepsin digests of human GBM. The specificity of the antibodies was verified by elution of antibodies bound to urinary GBM-related peptides on nitrocellulose blots and demonstration of reactivity of the eluted antibodies with the respective GBM digests. Furthermore, six mice immunized with urinary GBM-related peptides all developed focal linear deposits of mouse IgG along their GBM, linear and mesangial deposits of C3 in the glomeruli and serum antibodies reactive with human GBM. Monoclonal antibodies against urinary GBM-related peptides of one of the mice reacted with different peptides of the non-collagenous and collagenous domains of type IV collagen, the major structural protein of GBM. In the majority of the 75 patients' urines tested, excretion of GBM-related peptides with molecular weights of 33, 50, 80 and 150 kilodaltons (kD) was detectable. Patients with a diminished glomerular filtration rate (GFR) demonstrated excretion of the 33 kD peptide more frequently (91%) and never of the 80 kD peptide as compared with patients with normal GFR (33 kD [42%] 80 kD [87%]). The pattern of urinary GBM-related peptides was not specific for the underlying renal disease as in Alport's syndrome.

    Notizen: Summary Deposition of complement factors, immunoglobulins and infiltrating cells was evaluated by immunohistochemical staining in 30 temporal artery biopsy specimens from patients suffering from temporal arteritis and/or polymyalgia rheumatica and in controls. In the temporal arteritis group infiltrating cells, classic complement, alternative complement and lytic complex activation were detected. In specimens from patients suffering from only polymyalgia rheumatica there was unexpected evidence of classic complement and lytic complex activation. We conclude that immuno-histochemistry provides support for the concept of temporal arteritis and polymyalgia being based on the same pathological process.
