    Notes: Summary Effects of the organic calcium antagonists verapamil and flunarizine on pentylenetetrazol induced paroxysmal depolarizations were tested in organotypic neocortical explants taken from neonatal rats. In these in vitro experiments the papaverin derivative verapamil depressed, and finally abolished, epileptic discharges in all cases. The piperazine derivative flunarizine, however, which is known to suppress epileptic discharges in hippocampal CA3 neurons (Bingmann and Speckmann 1986), showed no significant antiepileptic effects in the explanted neocortical neurons. Thus, the present findings may indicate that the suppressive action of flunarizine on the generation of paroxysmal depolarizations is restricted to distinct populations of neurons.

    Notes: Summary Chronic blockade of spontaneous nerve impulses by means of tetrodotoxin leads to abnormally diffuse afferent projections into spinal cord cross-sections cultured for two to six weeks in vitro. In addition, even untreated explants which show a low level of spontaneous cord discharges failed to develop the normal degree of dorsal pathway selectivity. It is therefore concluded that centrally generated neuronal activity may play an important role in eliminating ‘exuberant’ connections which, during early development, are transiently present in this part of the nervous system.

    Notes: Summary Cultured spinal cord explants in which little spontaneous bioelectric activity was present showed, when monitored using sensory ganglion-evoked monosynaptic action potentials, diffuse innervation by ingrowing afferent fibers at 3–4 weeks in vitro. In contrast, highly active cultures of the same age showed a strong tendency for functional sensory connections to be made within the dorsal half of the cord. Regional specificity was present in mature cultures (4–5 weeks in vitro), however, even when their spontaneous activity level was low. The results support earlier results using tetrodotoxin, and make it appear likely that centrally generated neuronal discharges can influence the topography of afferent terminals within the developing spinal cord.

    Notes: Summary Following transganglionic transport of cobaltic lysine applied to the cut end of cutaneous nerves, the central terminal arbours were filled to such an extent that it was possible to visualize the dermatomal projection upon the dorsal horn in unoperated and in skin-rotated frogs. Sensory nerves of the trunk skin terminate in an oval-shaped area which probably corresponds to laminae 1–3 (or 1–4) in the mammalian spinal cord. One type of terminating collateral had a restricted terminal field either in the medial, or in the lateral, part of the oval-shaped area. Another type ended with a large terminal arbour covering almost the total extent of the oval shaped area. These terminal arbours were found for both the dorsal and the ventral cutaneous nerves. Dorsal cutaneous nerves projected preferentially to the lateral part, while ventral cutaneous nerves projected to the medial part, but both nerves sent a significant contigent of collaterals to the whole extent of the oval-shaped area. A rearrangement of the central projection patterns of the dorsal cutaneous nerves, consistently mediating misdirected responses, was not observed in skin-roated frogs. Collaterals of trunk skin nerves terminated exclusively in the thoracic segments; they did not enter the limb-innervating (brachial and lumbosacral) segments of the spinal cord. The results corroborate the crossed pattern of sensory input in the dermatomal projection. The large, extensively overlapping, areas of termination of individual skin nerves suggest that wiping responses mediated by these nerves should be regarded as complex behaviour released from a preprogrammed centre rather than a reflex arc in which the response specificity would be determined by wiring specificity.

    Notes: Abstract The presence and time-course of β-adrenoceptor density in cultured explants of neocortex obtained from 6-day-old rat pups were investigated using a [125I]ICYP binding assay. A delayed, but more pronounced, increase in the receptor expression was observed as compared to the situation previously described in vivo. These changes only occurred for the β1-subtype of the receptor, whereas the β2-subtype binding remained constant up to 3 weeks in vitro. The delay of β1-adrenoceptor expression may be due to the incomplete presence of the proper maturational input, and the late enhancement of receptor expression to upregulation related to the absence in vitro of noradrenergic input. Decreased β-adrenoceptor levels could be induced by chronic treatment of the β-agonist isoproterenol (1 μM) introduced either for 3 or 13 days. Again, changes in density were found only for the β1-adrenoceptor binding sites. There is no reduction of receptor density following return to control conditions for 10 days after a 3-day treatment with isoproterenol, demonstrating the ability of this model to attain its final receptor density notwithstanding the developmental insult.

    Notes: Summary Recent investigations have suggested specific differences in back and belly skin in anurans which appear to influence the quality of reflex responses obtained from various areas of the animals body. The present investigation represents a comparative morphological study of back and belly skin in control and skin-graftedRana pipiens, with special regard to the neuroepithelial relationships. A distinct difference in pigmentation of back and belly skin was observed. Intra-epithelial Merkel cells were present in all skin samples studied. The origins of the numerous unclassifiable cells in the Merkel cell region are discussed in relation to a presumed coordinating function of the Merkel cell during epithelial differentiation. Epitheliomesenchymal interactions were observed in the richly innvervated dermal regions. Two types of morphologically different intra-epithelial nerve endings were observed. These observations are discussed in relation to earlier observations on vertebrate skin and in relation to misdirected reflex responses obtained in skin-grafted anurans.
