    Description / Table of Contents: Zusammenfassung Es konnte gezeigt werden, daß sich sowohl die mitochondriale als auch die nucleäre DNS des Hamstermelanoms A Mel 3 mit radioaktivem Thymidin markieren läßt. Nach in vivo-Etikettierung mit 3H-Thymidin wurde zu verschiedenen Zeitpunkten die spezifische Radioaktivität der DNS an isolierten Mitochondrien und Zellkernen bestimmt. Aus den gemessenen Werten lassen sich sog. Halbwertzeiten berechnen, die in den Tumorfraktionen erheblich kürzer als in entsprechenden Leberfraktionen sind. In vivo appliziertes Proflavin verändert sowohl die “Halbwertzeiten” als auch den mitochondrialen DNS-Gehalt. Die Befunde werden diskutiert.
    Notes: Summary Mitochondrial and nuclear DNA were labelled by radioactive thymidine in the A Mel 3 hamster melanoma. After various time intervals the specific activity of 3H-Thymidine pulse labelled DNA was determined in mitochondria and nuclei after isolation. From the results socalled half value times can be calculated. They were significantly shorter in the tumor fractions than in corresponding liver fractions. Injections of proflavine into the tumor bearing animals changes the half value times as well as the amount of mitochondrial DNA. The results are discussed.

    Description / Table of Contents: Zusammenfassung 1. Die Ultrastruktur des experimentellen Hamstermelanoms A Mel 3 von Fortner wird beschrieben. Wesentlich sind die Zell- und Kernpolymorphie sowie Zeichen einer hohen Stoffwechselaktivität: große Nucleolen und Kernporen, Reichtum an Ribosomen und Mitochondrien. 2. In allen untersuchten Tumoren wurden Virus-artige Partikel von ca. 100mµ Durchmesser gefunden, die aus einem elektronendichten Nucleoid und einer umgebenden Hüllmembran bestehen. Sie liegen einzeln oder gruppenweise vorwiegend im endoplasmatischen Reticulum, teilweise auch frei im Cytoplasma, möglicherweise auch in Mitochondrien. 3. Die ultrastrukturellen Befunde stehen in guter Korrelation zu den von uns früher mitgeteilten biochemischen Charakteristica dieses Tumors.
    Notes: Summary 1. The ultrastructure of Fortner's experimental hamster melanoma A Mel 3 is described. Most characteristic are the polymorphy of cells and nuclei, and morphological signs of high metabolic activity: nucleoli, nuclear pores, and the abundance of ribosomes and mitochondria. 2. In all tumors investigated so far, virus-like particles of about 100 mµ in diameter have been detected. These virus-like particles consist of an electron dense nucleoid and a surrounding membrane. They are found singly or in groups, mainly within the endoplasmic reticulum, sometimes freely in the cytoplasm, and possibly also in mitochondria. 3. These ultrastructural findings are in good correlation with the biochemical characteristics of this tumor which we have described elsewhere.

Für die Freisetzung des Enzyms ist die Einwirkung des nicht-ionischen Detergens Nonidet P-40 auf die Partikel erforderlich.
    Notes: Summary The amelanotic hamster melanoma A Mel 3 contains a particleassociated RNA-directed DNA polymerase (reverse transcriptase) which can be liberated from the particles by the non-ionic detergent Nonidet P-40. In addition, the presence of a single-stranded high molecular weight RNA (70S RNA) has been detected simultaneously with the presence of the reverse transcriptase activity.

    Notes: Summary Human meningiomas show characteristic chromosome aberrations very similar to those reported in human cell cultures experimentally transformed by Simian virus 40 (SV 40). Three out of eight meningiomas tested showed SV 40 specific tumor antigen but no virus capsid antigen. Experiments to rescue virus were successfully performed. Electron microscopic studies of these cell cultures revealed cytoplasmic inclusions containing virus particles which corresponded in size and morphology to Papova viruses.

    Notes: Summary Two of the main biochemical features characteristic for oncogenic RNA (oncorna) viruses have been detected in human glioblastoma. In all tumors tested so far a RNA instructed DNA polymerase (“reverse transcriptase”) activity was present which exhibited the criteria specific for oncorna viruses. It was stimulable by the synthetic polynucleotide poly-rA: oligo-dT, it was almost insensitive to actinomycin D but very sensitive to ethidiumbromide. Using the simultaneous detection test, a high molecular weight RNA species was found in the subcellular fraction with the highest reverse transcriptase activity. As revealed by electron microscopy, this subcellular fraction contains particles of a morphology similar to oncogenic RNA viruses of the C-type.
