    Notes: We report a patient who had raised serum and amniotic fluid alpha-fetoprotein levels at 16 weeks gestation. Ultrasonography revealed an apparently normal fetus and amniography displayed a soft tissue tumor in front of the fetus. The tumor turned out to be a placental haemangioma, when an apparently healthy girl weighing 2·88 kg was born at term.

    Notes: Mouse interleukin 4 (IL-4) has been shown to act on B cells as an induction factor for Ig class switch. We studied the characteristics of IL-4-regulated Ig isotype production in lipopolysaccharide (LPS)-stimulated splenic B-cell cultures with emphasis on the comparison between the IgG1 and IgE responses. The results show that the kinetics for the appearance of IgG1 and IgE isotypes are similar, but that the dose of IL-4 required for the induction of an IgE response is 3–10 times higher than that for an IgG1 response. No requirement for T cells was found for the induction of either isotype. Pre-incubation of cells for 24 h with IL-4 alone was sufficient 10 induce an IgG1 response when cells were recultured with LPS from days 1 to 6. However, the simultaneous presence of both IL-4 and LPS for at least 24 h was required for a detectable IgE response. For an optimal IgE response. IL-4 needed to be present for more than 72 h in LPS-activated cultures. The possible reasons for the different regulation of IgG1 and IgE responses are discussed.

    Notes: The presence of a mitogenic lectin during the cytotoxicity assay enables the detection of non-specific cytotoxicity (lectin-dependent cell-mediated cytotoxicity (LDCC). Human mononuclear cells cultured at 37°C in vitro became cytotoxic after 1 day, as detected by the LDCC. In some individuals, the presence of adherent cells during culture inhibited the development of cytotoxicity. Activation by concanavalin A (Con A) resulted in enhanced cytotoxicity after a few days in culture. There was no clear relationship between the lectin doses required for optimal induction of cytotoxicity and [3H]|thymidine uptake. High Con A concentrations that induced a proliferate response often inhibited cytotoxicity in mononuclear cells depleted ot adherent cells.

    Notes: Recombinant protein A(SpA) produced in Escherichia coli or Bacillus subtilis bacteria did not induce activation of human peripheral mononuclear cells, whereas SpA preparations obtained from naturally occurring Staphylococcus aureus bacteria as well as recombinant SpA from Staphylococcus xylosus were potent mitogens. Further purification of SpA from S. aureus showed that the mitogenic material was concentrated in the side fractions containing more basic molecules. Some staphylococcal enterotoxins are mitogenic for human cells and in order to test whether contaminating enterotoxins would be responsible for the mitogenic effect of SpA preparations, rabbit antibodies were produced against enterotoxin A and B. These antibodies inhibited activation of human cells induced by the enterotoxins used for immunization but did not affect the activation induced by SpA preparation. The addition of selected human sera to in vitro cultures resulted in an inhibition of the response induced by low doses of SpA. There was no clear relationship between these effects and the content of IgG antibodies against staphylococcal enterotoxins A, B, and C1 in the sera. Thus, we conclude that the mitogenicity of SpA preparations is caused by contaminating molecules, probably not enterotoxins A, B, or C1.

    Notes: Intact IgG and fragments F(ab')2γ, Fabγ and Fcγ from a rabbit anti-protein-A serum (RapA) and corresponding preparations from normal rabbit IgG (NRG) were tested for their inhibitory effect on the binding of protein-A-reactive 125I-IgE and 125I-Fcγ, respectively, to protein-A–Sepharose. Intact IgG, F(ab')2γ and Fabγ of RapA inhibited the binding of protein-A-reactive 125I-IgE, whereas only intact IgG and Fcγ fragments from both RapA and NRG inhibited the binding of 125Fcγ to protein A-Sepharose. Further, the functional relationship between RapA and human polyclonal IgG was studied in a nephelometric test system. Intat IgG or fragments of IgG from human polyclonal IgG and rabbit anti-protein-A were found to affect the precipitation between human IgG and protein A in a similar way. Thus F(ab')2γ fragments and intact IgG enhanced the precipitation, whereas Fabγ and Fcγ fragments inhibited the precipitation. Protein A and an Fc-binding fragment of protein A (fragment B) were tested for their abilities to link different radiolabelled immunoglobulin preparations expressing the alternative and the classical protein-A reactivity to immobilized Fc fragments. All proteins expressing the alternative reactivity were efficiently bound both by fragment B and by protein A, indicating that fragment B, in addition to its classical Fc-binding activity, also expresses the alternative protein-A reactivity.

    Notes: The peptic fragments F(ab′)2 and Fc″ of human polyclonal IgE were tested by the SpA-IgE radioimmunoassay for interaction with protein A from Staphylococcus aureus. The Fab″2ɛ fragment but nut the Fc″ɛ fragment retained the ability to react with protein A. IgG preparations from different species were tested for their capacity to inhibit the reaction between 125I-IgE and protein A. IgG preparations from man and the dog, pig, guinea-pig and rhesus monkey were all potent inhibitors whereas those from the rabbit, rat, sheep, horse, cow, goat and mouse were very poor inhibitors. The inhibition of the binding of 125I-IgE to protein A by Fab′2γ fragments, but not by Feγ fragments, suggests a novel type of interaction of protein A with immunoglobulins.

    Notes: Serum IgA and IgE levels were studied in the postnatal period in 21 infants having a paternal heredity of atopic disease. Three different sampling techniques were used, aspirated cord blood (CB), gravity-collected cord blood, and capillary collected blood at 4–5 days of age. Significant differences among the three sampling techniques were recorded for IgA (P〈0.01), but not for IgE. The IgA levels decreased from birth to 4–5 days of age in 90% (19/21) of the infants (P〈0.01). The corresponding decrease in IgE levels was 20%. This postnatal difference in the frequency of decreasing/increasing IgA and IgE levels was significant (P〈0.05). An analysis of CB IgA to detect maternal contamination of CB was found to be of questionable value, since only 50% (2/4) of the cases with an elevated CB-IgA level could be considered contaminated. The results of this study further emphasize that aspiration of CB and capillary collection of blood at 4–5 days of age are the best sampling techniques to avoid contamination. The general finding that paternal heredity had no significant influence on infant IgE contrasts with the strong influence of maternal heredity. Further studies will show whether the explanation lies in genetic or transplacental factors, or in both kinds of factors.

    Notes: A previous study revealed an increased occurrence of work-related respiratory complaints among Swedish woodwork teachers. For determination of whether an IgE-mediated mechanism was the cause of the symptoms, 127 woodwork teachers and 111 reference subjects (other school personnel) in Stockholm gave serum for analysis of total IgE, Phadiatop, and RAST to extract of sawdust from five commonly used Scandinavian woods (pine, birch, juniper, alder, and linden). The total serum-IgE level was similar in the woodwork teachers (geometric mean 35, range 〈2–1700 kU/l) and the reference group (34, 〈2–860 kU/l), and the frequency of positive Phadiatop was equal (29.9% and 31.5%, respectively). RAST was positive only in three cases, two reference subjects (birch and pine) and one woodwork teacher (alder). Hence, no correlation between exposure and sensitization to wood dust was found, and the results do not support the assumption that the complaints related to exposure to dusts from the examined Scandinavian woods are IgE mediated.

    Notes: The natural humoral immune response to the dust mite Lepidoglyphus destructor was assessed by comparing the IgG1 and IgG4 responses elicited in allergic (n= 44) and healthy (n= 16) individuals in a farming population chronically exposed to this allergen. With the aid of an immunoblotting technique and ELISA, the sera were analyzed for anti-L. destructor antibodies. While the majority of sera from the allergy group displayed several bands for both IgG1 and IgG4, the nonatopic healthy group was negative as analyzed in immunoblotting. When they were analyzed in ELISA, there was a significantly higher response in the allergy group than in the healthy group for IgG4, but not for IgG1. Taken together, these results imply that the immune system of individuals spared from allergic reactions to L. destructor not only lacks IgE antibodies but also seems largely to “ignore” these allergens/antigens despite exposure.

    Notes: ELISA (enzyme-linked immunosorbent assay) inhibition with a monoclonal antibody (mAb) (42B6) to Lepidoglyphus destructor was used to detect and quantify the storage-mite allergens in 30 dust samples collected from barns. Regarding the mite fauna, microscopic inspection of the barn dust and mite counts showed that L. destructor infested all 30 barns investigated (range 430–195400 mites/g dust). In 29/30 barns, L. destructor constituted more than 70% of the Astigmata species. Acarus siro was found in 26 samples, the highest value being 16 155 nites/g. No Dermatophagoides species were found. As to mites of the suborder of Prostigmata, species belonging to seven different families were detected. Besides the predominant L. destructor, allergens derived from other storage mites such as Glycyphagus domesticus, A. siro, and Tyrophagus putrescentiue have previously been assessed by this ELISA method. The correlation between number of mites and concentrations of mite antigen as measured by ELISA was assessed by linear regression (r2= 0.83). Thus, inhibition of mAb 42B6 in ELISA would seem to offer a simple and reliable method to detect levels of dust-mite species belonging to the Acaridae and Glycyphagidae families.
