    Notes: Abstract The expression of p53 was studied immunohistochemically in combination with the DNA ploidy pattern by gland isolation in 97 alcohol-fixed gastric lesions. A polyclonal antibody, CM-1, was applied to the paraffin-embedded sections in this study. Overexpression of the p53 protein was found in 73.2% of 41 well or moderately differentiated gastric carcinomas and 52.2% of 23 cases with poor differentiation (P〈0.05). Immunoreactivity of p53 was also detected in isolated cancerous glands. No p53 immunoreactivity was detected in benign gastric lesions including adenomas, hyperplastic polyps and regions of intestinal metaplasia. In addition, flow cytometric DNA analysis was performed on isolated glandular epithelium adjacent to the portions used for immunostaining. DNA aneuploidy (DA) was detected in 85.7% of the well or moderately differentiated carcinomas and 42.9% of those with poor differentiation (P〈0.05). There was a positive correlation between DA, p53 positivity and the presence of regional lymph node metastasis, but not with other clinicopathological variables. In spite of the limited applicability of this method to poorly differentiated gastric cancer, we found that immunostaining and flow cytometry in combination with the gland isolation method facilitates analysis of gastric carcinogenesis.

    Notes: Summary The morphology and function of peripheral motor nerves of WBN/Kob rats of the newly identified strain with late onset and long-lasting diabetes were studied in comparison with those of nondiabetic age-matched rats. The most conspicuous and frequent change seen by electron microscopy in the diabetic rats was myelinopathy, initiated by vesicle formation in granular material accumulated in the myelin lamella. Initial changes developed into myelin blebbing or distention, demyelination and remyelination. A decreased population of nerve fibers in diabetic rats of advanced age suggested that the final stage of these changes was neuronal loss resulting from complete destruction of the neurons. The myelinopathy was consistently more frequent and conspicuous in diabetic rats than in age-matched nondiabetic rats, but the difference was not significant in rats of over 28 months old, because of the age-dependent increase of the change in older rats. The conduction velocity was significantly less in diabetic rats than in age-matched nondiabetic animals at 20 months but not at 28 months of age, because at the older age nondiabetic rats also showed a reduced conduction velocity. These findings showed that male WBN/Kob rats develop diabetic motor neuropathy, but that the essential changes in diabetic neuropathy are indistinguishable morphologically and functionally from those in age-dependent neuropathy.

    Description / Table of Contents: Zusammenfassung Das Genom des Hepatitis-C-Virus (HCV) wurde im Speichel von 76 chronischen HCV-Trägern (Durchschnittsalter knapp 60 Jahre), die einen hohen c-100-Serum-Titer und Core-Antikörper der zweiten Generation aufweisen und in einer ländlichen Stadt in Japan leben, gesucht. In 27 Proben (27 Fälle, 36%) wurde das HCV-RNA-Genom nachgewiesen durch reverse Transcriptase-Polymerase-Kettenreaktion mit beiden Primer-Sätzen, die zwei Regionen des HCV-Genomes einschließen: die 5′-nichtkodierende Region und die die mutmaßliche Hülle umgebende Region (E1). Die Transaminasen-Werte zur Zeit der Probenahme waren bei Patienten mit HCV-RNA im Speichel höher als bei denjenigen ohne feststellbare HCV-RNA (p=0.04 für Alaninaminotransferase, p=0.04 für Aspartataminotransferase; Wilcoxon-Test). Zwar war die Häufigkeit der Positivität bei 5′-nichtkodierenden Primern höher (14/59 gegenüber 15/68), aber die Differenz war nicht signifikant. Unsere Daten zeigen, daß die Schwere und die Dauer der Leberfunktionsstörung die Nachweisbarkeit des HCV-Genoms im Speichel beeinflussen, was ein strittiger Punkt unter den Forschern war.
    Notes: Summary The hepatitis C virus (HCV) genome was sought in the saliva of 76 chronic HCV carriers (mean age nearly 60 years) in a rural Japanese twon, who had high serum titers of c-100 and anti-core second generation antibodies. In 27 samples (27 cases, 36%), the HCV-RNA genome was detected by the reverse transcriptase — polymerase chain reaction with either of two sets of primers covering two regions of the HCV genome: the 5'noncoding region and the region encompassing the putative envelope (E1). Transaminase values at the time of sampling were higher in the patients with than in those without detectable HCV RNA in saliva (p=0.04 for alanine aminotransferase, p=0.04 for aspartate aminotransferase; Wilcoxon test). The prevalence of the positivity was higher by 5'noncoding primers (14/59 vs. 15/68). Our data show that the severity and duration of hepatic dysfunction influence the detectability of the HCV genome in the saliva. This has been a controversial point among investigators.

    Notes: Abstract Cell culture models of calcium phosphate renal stone formation were established using the MDCK cell line. Renal microliths were detected within pseudocysts in three-dimensional soft agar cultures, and were also observed in the basal region of cells lining the cell sheet, and immediately beneath domes or blisters in monolayers and collagen gel cultures. Light and scanning electron microscopy indicated that these microliths had a similar lamellated and spherical appearance to those in humans. These microliths were first detected microscopically after 21 days of culture, and were found to be composed of calcium phosphate by X-ray and microinfrared spectroscopic analyses. These culture models may provide a powerful new tool to study the pathogenesis of renal stone diseases and/or calcium phosphate stone formation in humans and animals.
