A New Indolizine Synthesis with 2-Acylpyridines, I. Azomethines Derived from 1-Aminoindolizines and their DecompositionThe 2-acylpyridines 1, 2, and 3 react with the aromatic aldehydes 4, and ammonium acetate in glacial acetic acid to yield the azomethines 5, 6, and 7 of 1-aminoindolizines. - The structure of these products is shown by cleavage with perchloric acid as well as with acetic anhydride and water. The reaction with perchloric acid leads to salts of 1-hydroxyindolizines. Depending on the substitution they have the structure of enols 19 or ketones 20. Schotten-Baumann acylations of both 19 and 20 yield the 1-benzoyloxyindolizines 22 or 23. - With acetic anhydride and water the azomethines yield 1-acetaminoindolizines 26 or 27 via the immediate acylation of the cleavage products, the 1-aminoindolizines 24 or 25. The 1-acet-aminoindolizines 26 or 27 can also be prepared from 1-acetylindolizines 28 by the Schmidt reaction.

Aus 2-Acyl-pyridinen 1, 2, 3 und aromatischen Aldehyden 4 entstehen beim Erhitzen mit Ammoniumacetat in Eisessig Azomethine 5, 6, 7 von 1-Amino-indolizinen, deren Struktur durch Spaltung mit Perchlorsäure sowie mit Acetanhydrid und Wasser bewiesen wird. - Mit Perchlorsäure bilden sich aus ihnen Salze von 1-Hydroxy-indolizinen, die je nach ihrer Substitution in der Enolform 19 oder Ketoform 20 erhalten werden. Unabhängig von diesem Strukturunterschied erhält man aus ihnen durch Schotten-Baumann-Reaktion die 1-Benzoyl-oxy-indolizine 22 bzw. 23, - Mit Acetanhydrid und Wasser werden die Azomethine ebenfalls gespalten. Hier werden jedoch die intermediär auftretenden 1-Amino-indolizine 24, bzw. 25 nicht wie bei der Spaltung mit Perchlorsäure hydrolysiert, sondern zu den 1-Acetamino-indolizinen 26 bzw. 27 acetyliert, die man auch aus den anderweitig leicht zugänglichen 1-Acetyl-indolizinen 28 durch die Schmidt-Reaktion darstellen kann.
