    Notes: The local immune reaction of progressive chronic periodontal disease may be particularly influenced by macrophages and macrophage-derived factors. Among these substances the prostaglandins and lysosomal enzymes may play an important pathogenetic role. Parallel immunohistochemical and radioimmunological studies were done to investigate the relationships of the immune-competent cells and the inflammatory mediators in gingival tissues. The radioimmunological analysis revealed that prostaglandin E increases markedly in the established gingival lesions. Immunohistochemically prostaglandin E was mainly localized within macrophage-like cells. Cytoplasmic lysozyme could be detected in these cells, too. On the other hand, the B-cell response is the prominent feature in established chronic periodontal disease. However, there is apparently a disturbed B-cell reaction as indicated by the irregular IgG-subclass pattern and by the production of mainly monomeric IgA. The possible interactions of macrophages and especially B-cells via prostaglandin E-mediated mechanisms are discussed.

    Notes: Six oral papillomas and 7 oral leukoplakias were studied with genus-specific antibodies against detergent-disrupted papillomaviruses. Indirect immunofluorescence staining was applied to frozen sections. Distinct nuclear staining of superficial keratinocytes was seen in 5 of 6 oral papillomas, 2 of 5 homogenous leukoplakias, and in 2 cases of nodular leukoplakia, one of these showing transition into an invasive carcinoma. Papillomavirus antibodies offer a new way to detect and localize papillomaviruses in epithelial hyperplasias. The relevance of the immunomorphological identification of papillomavirus-associated antigens is discussed with special reference to the molecular hybridization technique.

    Notes: Biopsies were taken at 8 and 12 weeks and at 6 months from oral jejunal mucosal transplants in 5 patients who had undergone extensive surgery for tumors of the floor of the mouth. Microvaseular anastomosed jejunal mucosa had been used for the primary reconstruction. Histological findings included flattening of the jejunal relief, atrophy and erosion of the epithelium with reduction of jejunal excretion and marginal overlapping of oral mucosa. Immunohistocheniical labelling of the cytoskeleton revealed thai jejunal epithelium does not undergo rapid metaplasia in the course of adaptation to oral mucosa. These findings indicate that the jejunal mucosa does not completely change its architecture and does not adapt to the requirements of the oral cavity after a period of 6 months.

    Notes: Monoclonal antibodies which react against T-cell subpopulations and Langerhans' cells were used to examine the phenotype of immunocompetent cells in oral lichen planus. Immunomorphologically, the stromal infiltrate of this condition considerably resembled the delayed type of immune reaction. Although all subpopulations of immunocompetent cells were present in the stromal infiltrate, Langerhans' cells (OKT-6 positive, HLA-DR positive) and suppressor/cytotoxic T-lymphocytes (OKT-3 positive, OKT-4 negative, OKT-8 positive, HLA-DR positive) predominated. Our immunohistological findings support the suggestion that aggregations of T-lymphocytes are responsible for the cytotoxic processes which occur within the squamous epithelium in oral lichen planus.

    Notes: Summary Cytoskeleton of eucaryotic cells consists of three filament systems. Micro tubules, intermediate filaments, microfilaments. Among these filament systems intermediate filaments are of special interest, because they vary dependent from the tissue of origin. It is the keratin filament which is characteristic of epithelious cells. In contrast Vimentin-filaments occur only in mesenchymal cells (Langerhans cells and lymphocytes). With regard to the differentiation of epithelial tumors (basaliomas, carcinomas) it is particularly important, that keratin filaments of different molecular weight are demonstrated in these neoplasias. These different keratin filaments can be immunhistologically identified by means of specific antibodies. We want to present two female patients who suffered from metachronisly developed basaloid tumors at different regions of the face epidermis. In the case of these patients we want to demonstrate the value of immunhistological investigations of the intermediate filament system for clinical research and praxis.

    Notes: Summary Skin biopsies of patients with small and large plaque parapsoriasis, premycotic lesions and mycosis fungoides in different stages were examined. Special attention was paid to the relationships between Langerhans cells (LC) and the neighbouring keratinocytes and lymphocytes. At the contact areas of LC and keratinocytes as well as LC and lymphocytes, particular cell membrane phenomena were observed. Aggregations of Langerhans granules and fusions of granules with LC plasma membranes were found exclusively at LC-keratinocyte interfaces. At LC-lymphocyte contact zones cell membrane appositions were seen. In all cases investigated, virus-like particles were mainly ound in LC and indeterminate cells (IDC). In 3 cases lymphocytes also contained these particles. It was of particular interest that virus-like particles were observed in skin specimens of all diseases investigated. Discrimination of these particles from other cellular organelles - especially lysosomes- was difficult, however. The significance of our findings, particularly regarding to the supposed virus aetiology of cutaneous T cell lymphomas, is discussed.

    Notes: Während das Plattenepithelkarzinom der Vulva eine konstante Inzidenz aufweist, lässt sich für die vulväre intraepitheliale Neoplasie (VIN) eine steigende Häufigkeit nachweisen. Diese gründet sich auf die wachsende Infektionsrate mit den in einem Teil der Fälle pathogenetisch bedeutsamen Humanen Papillomaviren (HPV). Hinsichtlich der klinischen Therapieplanung sowie der prognostischen Einschätzung ist eine histopathologische Differenzierung der präneoplastischen und neoplastischen Erkrankungen der Vulva unerlässlich. Gleiches gilt für maligne Erkrankungen der Vagina, wobei hier insbesondere primär vaginale Tumoren von metastatischen Absiedelungen zu unterscheiden sind. Für den klinisch-onkologisch tätigen Arzt ist aus diesen Gründen die Kenntnis histopathologischer Kriterien zur Interpretation der ihm übermittelten Befunde hilfreich.

    Notes: Summary The migratory and proliferative capacity of oral epithelium was studied after induction of subepithelial suction blisters in palatal rat mucosa. FITC-coupled lectins and their affinities to epithelial cells at the wound margin were analyzed by immunfluorescence microscopy. Immunocytochemical distribution of 67 K-keratin polypeptides in different epithelial layers were studied using specific antisera against keratin proteins. By means of3H-thymidine-autoradiography the proliferation of epithelial basal cells and their migration at the wound edge was quantified by evaluation of the labelling index. Double labelling was performed, combining the autoradiographic and the keratin staining technique. It was found that the subepithelial suction blister showed rapid repair (epithelial regeneration after 3–4 days). Peanut Agglutinin (PNA) showed a selective affinity to the basal 2–3 layers of oral rat mucosa. No modifications of lectin affinities were found in epithelial wound repair. PNA-positive keratinocytes were demonstrated in the migratory epithelial tongue. The selective presence of 67 K-keratin (67 K) in the suprabasal epithelial cell layers was interpreted as an indication of the differentiation process in rat oral mucosa. The3H-thymidine-labelling index of basal cells increased significantly, beginning at the wound edge after 24 h. The peak of the labelling index curve was located in the vicinity of the wound after 96 h. Double labelling technique revealed 67 K-negative and3H-thymidine-positive basal cells as well as 67 K-positive and3H-thymidine-negative cells at the migration front. These observations support findings that migrating epithelial cells in oral wound healing derive both from undifferentiated (str. basale) and from differentiated (inferior str. spinosum) cell compartments.

    Notes: Summary Leu-M1 antigen is a monocyte/granulocyte-related marker known to be consistently expressed in the Reed-Sternberg cells of patients with Hodgkin's disease. Recently, however, the presence of Leu-M1 has also been noted in tumour cells of a variety of non-haematopoietic neoplasms, most of them adenocarcinomas. The biological significance of this aberrant reaction has not been clarified. We have been able to demonstrate marked epithelial Leu-M1 immunoreactivity (〉15% tumour cells positively stained) in 24 out of 76 (32%) papillary carcinomas of the thyroid gland (PC). This phenomenon was more frequently observed among PCs at an advanced stage of disease (pT4 vs. pT1–3 and M1 vs. M0 p〈0.05). The degree of epithelial Leu-M1 positivity was also shown to be significantly correlated to the clinical course of PC. Irrespective of other morphological and clinical features, death resulting from cancer occurred 17 times more frequently among PCs with marked Leu-M1 positivity (8/24) when compared with tumours with only slight or absent immunoreactivity (1/52) (p〈0.00005). These findings suggest that Leu-M1 immunostaining provides significant prognostic information for patients with papillary carcinoma of the thyroid gland.

    Notes: Summary Autopsy specimens of normal human thymus, from cases of accidental involution, follicular hyperplasia, thymomas and a teratoma were investigated by immunocytochemistry using specific immune sera to small and large keratins. Keratin antisera represent a “marker” of both Hassall's corpuscles (HC) and so-called epithelial reticular cells. There were no apparent differences in keratin polypeptides distribution between cortical and medullary thymic epithelial cells. In accidental involution, the epithelial framework became prominent: epithelial cortical borders and epithelial perivascular sheaths appeared often to be discontinous structures. The central and occasionally cystic spaces of HC did not react with keratin antisera. In follicular hyperplasia, almost solid epithelial aggregates were seen which were located around germinal centers. In thymic tumours, neoplastic epithelial cells displayed a marked immunorectivity with keratin antisera. Immune sera against keratin filaments represent an interesting tool in thymus research and in the diagnostic pathology of thymic tumours.
