    Notes: We report a 49-year-old white woman having asymptomatic hyperkeratotic comedone-like lesions on her legs, arms and trunk. Her sister is similarly affected, but less severely. The clinical and histopathological features indicated a diagnosis of familial dyskeratotic comedones, a rare autosomal dominant condition.

    Notes: Background:  To understand the typical growth behavior of basal cell carcinoma (BCC) we searched for the correlation between proliferation and apoptosis and progression of BCC.Methods: Expression of Bcl-2, Bax, and Ki-67 was immunohistochemically investigated in both normal skin and BCC cells, as well as in the epidermis overlying BCC.Results:  The results showed that in normal epidermis, Bcl-2 was homogeneously expressed in the basal cell compartment, whereas Ki-67 expression was largely restricted to the parabasal layer, the layer just above the basal cell layer, and exhibited a more scattered staining pattern. Bax was occasionally expressed in the basal layer and widely in the suprabasal compartment. Strikingly, the apparently normal epidermis overlying BCC showed an increased Bcl-2 staining. In BCC, cells stained homogeneously for Bcl-2, whereas Bax and Ki-67 showed scattered staining patterns. Simultaneous expression was seen for Bcl-2 and Bax in 80 ± 7% of the tumor cells, and co-expression of Bcl-2 and Ki-67 in 20 ± 7% of the tumor cells. The cells expressing Bcl-2 and Ki-67, but lacking expression of Bax, the progressive fraction, comprised on average 7 ± 3% of the tumor cell population.Conclusion:  These results suggest that this small progressive fraction of tumor cells, in combination with the relatively high percentage of cells still prone to apoptosis, can explain the indolent growth behavior of BCC.

    Notes: Summary The trends in the incidence and the sites of primary basa cell carcinoma (BCC) and squamous cell carcinoma (SCC) of the skin were examined in a defined population of 650,000 persons in the SE Netherlands during the period 1975–88. The data was obtained from the Cancer Registry in Eindhoven and was from the dermatological and surgical clinics of 12 community hospitals. The incidence rates for BCC during this period rose from 42 to 53/100,000 person years for males and 24 to 38/100,000 person years for females. The rate of incidence for SCC varried with an increase mainly among males. In about 80% of the cases BCC and SCC occurred on the head and neck. Allowing for some detection bias a rise in the true incidence of BCC and SCC was observed, with an increase in SCC only since 1982. There was a marked decline in the incidence of SCC on the lips of males. These rates and trends fit into a pattern observed in other parts of Europe.

    Notes: Summary In previous experiments Ciamexone, derivative of 2-cyan-aziridine, was able to influence T-cell-mediated regulatory mechanisms but seemed to have no or only little effect on T cell effector mechanisms. On the basis of these observations Ciamexone seems to be a highly selective immunosuppressive agent. In order to evaluate further possible mechanisms of Ciamexone it was the aim of our investigation to study its influence on interferon γ (IFNγ) production in phytohaemogglutinin (PHA)-stimulated T lymphocytes of 15 tumour patients and 12 healthy reference subjects. The IFNγ concentration of the cell supernatant was measured using an enzyme-linked immunosorbent assay. When the cells were stimulated with PHA at 7.5 µg/ml the IFNγ concentration rose to significantly different values in the reference group (1.0 ng/ml) as compared to the tumour patients (0.4 ng/ml) (P 〈0.05). An addition of Ciamexone (at any of the concentrations administered) to PHA stimulation of peripheral blood mononuclear cells (PBMC) showed no influence on the IFNγ release in either test group. The influence of hydrocortisone on the stimulation of PBMC with PHA resulted in a dose-dependent suppression of IFNγ production in both test groups, again with significant differerences between them. The IFNγ concentration was 0.95 ng/ml in the reference group and 0.2 ng/ml in the tumour patients when 0.01 µg/ml hydrocortisone was added (P 〈0.05). At 10 µg/ml hydrocortisone suppressed IFNγ production completely in both groups. Our results corroborate those investigations that showed no influence of the compound on T cell effector mechanisms. The attenuation of humoral immunophenomena, however, suggest a very specific point of action within the immune system by Ciamexone.

    Notes: Summary Human tumor cells from a squamous cell carcinoma of the lung and from a malignant melanoma and human bone marrow progenitor cells were cultured in a methylcellulose monolayer system. To obtain tumor material for repeated experiments with human tumor specimens, human xenografts grown in the nude mouse system were used. The cultures were incubated continuously with adriamycin, actinomycin-D, bleomycin, cisplatinum, and melphalan. Dose response curves for bone marrow and tumor cell colonies were established. A comparison of bone marrow and tumor cell colony dose response curves might be useful for the assessment of the drug sensitivity of tumor cells independent of in vivo pharmacologic data and duration of drug exposure.

    Notes: Summary A methylcellulose monolayer system is described that facilitates the “in vitro” cultures of human spontaneous tumor cells. The tumor samples were disaggregated mechanically and cultured in 0.9% methylcellulose with 30% fetal calf serum in Iscove's modified Dulbecco's medium. Eighty-five individual tumor samples with different histological types were seeded, 45 gave rise to tumor cell colonies. The plating efficiency ranged from 0.02% to 0.22%. Cytologic and cytochemical staining from aspirated cells revealed the same morphology of cells as the cells in the suspension. A flattening of colonies, as it has been described by others using methylcellulose monolayer, was not observed.

    Notes: Summary Bone marrow specimens from 27 patients with small cell carcinoma of the lung (17 with limited and 10 with extensive disease) were plated in a culture system that supports the growth of multilineage haemopoietic progenitors CFU-GEMM. In five patients (three with extensive and two limited disease) atypical colonies could be observed that were not identifiable as haemopoietic colonies. Cytological staining and cytochemical examination as well as electronic micrographs suggest that these colonies are derived from metastatic carcinoma cells. The histological examination of marrow cells from three out of these five patients revealed no bone marrow involvement. Additional studies might provide further evidence whether bone marrow cultures are a useful probe in order to monitor bone marrow involvement in patients with small cell carcinoma of the lung.

    Description / Table of Contents: Abstract. Bilateral linear frontoparietal scleroderma en coup de sabre is an extremely rare form of localized linear scleroderma. In this paper the case of a 35-year-old woman with bilateral linear temporoparietal en coup de sabre is presented. The manifestations that can be associated with linear scleroderma and the serologic abnormalities it envolves are briefly discussed.
    Notes: Zusammenfassung. Die bilaterale lineare Sklerodermie en coup de sabre ist eine extrem seltene Form der lokalisierten bandförmigen Sklerodermie. Der Fall einer 35jährigen Patientin mit bilateraler linearer temporoparietaler Sklerodermie en coup de sabre wird vorgestellt. Die möglicherweise auftretenden Begleitsymptome und serologischen Befunde bei linearer Sklerodermie werden kurz erläutert.
