    Notes: Two examples of IgM χ-monolypic cold agglutinins occurring in patients with non-Hodgkin lymphomas, reacted with erythrocyte autoantigens, which were protease-resistant but were inactivated by neuraminidase. The cold agglutinins were inhibited by the trisaccharide sialyllactose [NcuNAc (α,2→3)Gal (β,1→+4)Glc], which is not related to oligosaccharides known to inhibit anti-I/-i cold agglutinins. Anti-Pr cold agglutinins are not inhibited by sialyllactose, although N-acetylneuramimc acid (NeuNAc) is an essential component not only of Gd but also of Pr determinants. Gd determinants are not bound lo erythrocyte membrane glycoproteins, but are apparently bound to membrane glycolipids (gangliosidcs).

    Notes: The variable-region subgroup determined by amino acid sequence analysis of heavy and light chains of two monoclonal cold agglutinins with the new anti-Gd specificity is reported. Both proteins belong to the VHI 11 subgroup of heavy chains; one light chain falls into the VKI subgroup, the other has a blocked N-terminus which so far has not been observed in human kappa chains. The comparison of anti-Gd with anti-l/-i or anti-Pr cold agglutinins indicates that anti-Gd differs from other cold agglutinins with respect to variable-region subgroup. The data extend previous findings on the restriction of certain antibodies to distinct variable-region subgroups.

    Description / Table of Contents: Zusammenfassung Bei der Lues-Serodiagnostik von Blutspenden ergab die Einführung des TPHA-Tests einen Anstieg positiver Testergebnisse von 0,16 % auf 0,64 % bei Erstspendern. Infolge der hohen Spezifität reduzierte der TPHA-Test die Zahl der fraglichen oder unspezifischen Reaktionsausfälle. Das wichtigste Testergebnis „TPHA- und CMT reagierend“ signalisiert eine behandlungsbedürftige Lues. Da diese Zahl in den Jahren 1967–1978 mit 0,06 bis 0,07 % unverändert geblieben ist, ergibt sich bei Blutspendern kein Anhalt für eine Zunahme der Lues. Dagegen gelang es uns, durch die Anwendung des TPHA-Tests eine bei Blutspenderuntersuchungen bisher unerkannt gebliebene Gruppe mit einer dreifach größeren Häufigkeit als die bisherigen seroreaktiven Lues-Fälle ausfindig zu machen. Fälle mit Reaktionsmuster „TPHA-Test reaktiv, CMT nicht reagierend“ weisen auf eine früher durchgemachte Lues hin. Die Anwendung des TPHA-Tests bei der Blutspenderuntersuchung vermindert die Risiken bei der Bluttransfusion und verbessert die durch das Gesetz geforderte Bekämpfung der Geschlechtskrankheiten.
    Notes: Summary The introduction of the TPHA test in syphilis screening of blood units donated at the Heidelberg University blood bank resulted in an increase of positive test results from 0.16 percent to 0.64 percent at the first donation. Due to the high specificity of the TPHA and (FTA-Abs) test the number of doubtful and unspecific reaction results was significantly reduced. The result “TPHA and CMT reactive” indicates syphilis to be treated. From 1967–1978 this figure (0.06 %–0.07 %) has remained unaltered, indicating that the incidence of syphilis among blood donors has not increased. Using the TPHA test it was possible to detect blood donors with a history of syphilis, a group which could not be identified in the past. It is characterized by the reaction pattern “TPHA reactive, CMT not reactive” and is three times as frequent among our donors as the seroreactive group identified by CMT only. If fresh or unsufficiently treated cases of syphilis can be excluded, these results indicate that the individual has had a syphilis sometimes in the past. Application of the TPHA test in blood donor care reduces the risks of blood transfusion and improves the control of venereal diseases as required by law.

    Notes: Summary A human monoclonal IgM-K cold agglutinin Ad, reacting with immunodominant neuraminyl groups on red cell membrane sialoglycoproteins, as typical for anti-Pr CA, was studied for its subspecificity. Inhibition experiments with chemically modified red cell sialoglycoproteins and agglutination studies with animal red cells showed that the CA Ad did not fit into the subspecificities anti-Pr1h,-Pr1d,-Pr2,-Pr3. Cold agglutinin Ad presents a new specificity within human monoclonal antibodies against immunodominant neuraminyl groups.

    Description / Table of Contents: Summary A simple method to prepare Gc-globulin from native human serum is described. The serum is separated on hydroxylapatit columns usingSörensen buffers. The Gc fraction contains part of the albumin which can be eliminated by means of rechromatography, leaving traces of albumin only. The output of Gc protein is almost quantitative. Furthermore, observations on the fundamental behaviour of the serum proteins during the hydroxylapatit chromatography with special reference to the α2-globulins are reported.
    Notes: Zusammenfassung Ein einfaches Verfahren zur Gc-Globulindarstellung aus nativem menschlichen Serum wird beschrieben. Die Auftrennung des Serums erfolgt an Hydroxylapatit-Säulen unter Verwendung vonSörensen-Puffern. Die Gc-Fraktion enthält noch einen Teil des Albumins, der durch Rechromatographie bis auf sehr geringe Reste eliminiert wird. Die Gc-Proteinausbeute ist nahezu quantitativ. Weiterhin werden Beobachtungen über das prinzipielle Verhalten der Serum-Proteine, insbesondere der des α2-Bereiches, bei der Hydroxylapatit-Chromatographie mitgeteilt.
