    Notes: Malignant atrophic papulosis (Degos disease) is a rare disorder characterized by a vasculopathy of unknown origin. 1 The cutaneous manifestations comprise erythematous papules, which heal to leave scars with a pathognomonic central porcelain-white atrophic area and a peripheral telangiectatic rim. There is usually involvement of the gastrointestinal tract but other organ systems can also be affected, the central nervous system being involved in 20% of cases. 2 It is frequently fatal within 2 or 3 years from onset of systemic involvement, the cause of death usually being intestinal perforation. 3 Our patient is of interest as she has survived an unusually long time despite florid cutaneous and neurological manifestations.

    Notes: Expression of the genes le20, ni3212, le25 and lcyp2 was quantified in relation to soil and plant water status for pot-grown tomato (Lycopersicon esculentum Mill. cv. Ailsa Craig) in a replicated trial in a partially environmentally controlled glasshouse. Leaf water potential, relative water content, stomatal conductance and gravimetric soil water content were measured at 0800,1200 and 1600 h on three days during the imposition of three different watering regimes. Paired leaf samples were analysed for mRNA. Plant-to-plant variability was quantified and partitioned. le20 mRNA was found in all leaves and responded both to short-term diurnal fluctuations in water status and to more severe stress, though with reduced sensitivity to the greatest stresses. An overnight decline in le20 mRNA was independent of soil or plant water status. The pattern of expression for ni3212 was similar though ni3212 mRNA was much more abundant and the overnight drop in expression was not significant. le25 mRNA was only detected where leaf water potential was below −0.9 MPa and increased linearly with further decline in leaf water potential. lcyp2 was constitutively expressed with a diurnal pattern characterized by a sharp depression at 1600 h but with expression being enhanced by increasing water deficit at all times. The control and significance of the four genes are discussed in relation to the proposed function of their respective gene products.

    Notes: The abscisic acid (ABA)-deficient tomato mutant notabilis (not) is believed to be a null mutation in the gene LeNCED1, encoding a 9-cis-epoxycarotenoid dioxygenase involved in ABA biosynthesis. We have sequenced and analysed a 19 kb genomic clone containing LeNCED1 and 5.4 kb of its promoter. This clone was transferred to not homozygotes and several non-wilty transformed plants were obtained. The basal ABA content, water relations, shoot and root growth, adventitious rooting, ethylene evolution and ability to accumulate ABA under water stress are described for two of these lines, notcomp.13 and notcomp.1. Partial complementation was observed for most parameters measured for notcomp.1. Full complementation was observed in notcomp.13 for all parameters measured in whole plants under well-watered and water-stressed conditions. These data provide further evidence that LeNCED1 is the wild-type allele of the not mutant gene. However, notcomp.13 was unable to accumulate the wild-type levels of ABA in rapidly dehydrated leaves, indicating that it too was only partially complemented. Since LeNCED1 is an environmentally regulated gene encoding a rate-limiting enzyme, precise levels and patterns of gene expression may be needed to fully recreate wild-type phenotype. The utility of partially complemented lines to study the role of ABA in plant responses to stress conditions, and in promoter analysis, is discussed.

    Notes: As a first step towards transferring a tetracycline (Tc)-inducible gene expression system to tomato, we have transformed tomato plants with the Tn10-encoded tet repressor gene (tetR). Homozygous transformed plants with high expression of tetR mRNA show a deleterious phenotype, having reduced shoot dry weights and leaf chlorophyll content, an even more marked reduction in root dry weight and leaf size, and altered photosynthetic physiology. It appears that TetR protein exerts its toxicity only when expressed beyond a threshold level and by interacting with a process that is non-limiting under slow growth conditions. The deleterious phenotype was almost completely reversed by the application of 1 mg dm−3 Tc to plants grown in sand. The possiblity is discussed that TetR causes these symptoms by binding to a specific DNA sequence functioning as a Tet operator.The effect of Tc on growth and physiology in wild-type plants grown in sand or rockwool is described. Tc at 0.1 mg cm−3 had no effect. Tc at 1 mg dm−3 caused a small reduction in root growth, while 5 and 20 mg dm−3 Tc caused large reductions in growth and photosynthetic parameters.

    Notes: Field experiments in Gloucestershire, UK, in the 1990–91, 1991–92, 1993–94 and 1994–95 growing seasons explored the merits of grazing in spring a traditional tall wheat (Triticum aestivum) variety, Maris Widgeon, with more modern shorter varieties. In the first 2 years, defoliation was achieved by mowing at 7 cm in March and/or April. In the second 2 years, varieties sown at two sowing dates were grazed by sheep at a stocking rate of 42 × 50 kg sheep ha−1 for 3 or 4 days in March. Defoliation reduced crop height and interception of photosynthetically active radiation (PAR). In 1991–92, mowing significantly reduced grain yield of some of the shorter varieties but not of Maris Widgeon. This interaction was related to the amount of PAR intercepted. In this year, mowing improved the establishment of undersown white clover (Trifolium repens) and perennial ryegrass (Lolium perenne), but in subsequent years the conditions were much drier and undersowing failed. In the last two experiments, grazing in March did not significantly reduce grain yield of any variety. The quality of the forage eaten by the sheep had a modified acid-detergent fibre (MADF) content of less than 300 g kg−1 dry matter (DM) and a crude protein (CP) content of more than 200 g kg−1 DM in both seasons. Yield of DM and calculated metabolizable energy (ME) of different varieties removed by the sheep interacted strongly with sowing date. September-sown Maris Widgeon provided ≈ 0·7 and 0·3 t DM ha−1 (or 7·8 and 3·4 GJ ME ha−1) in March 1994 and March 1995 respectively. However, the shorter wheat varieties, Hereward and Genesis, only provided 0·3 and 0·1 t DM ha−1 when sown at the same time in the 2 years. At later sowing dates all of the varieties only provided about 0·1 t DM ha−1 when sown in October 1993, or 0·01 t DM ha−1 when sown in November 1994. Sheep grazing reduced total weed biomass in June, and reduced the emergence of weed seedlings from soil samples collected after the wheat harvest. Effects of defoliation on foliar infection by Septoria tritici were inconsistent.

    Notes: Abstract The unique sensitivity of magnetic resonance imaging (MRI) in detecting disease activity in multiple sclerosis (MS) and the objective nature of the information obtained suggest that MRI will be a useful and reliable way of monitoring treatment trials. There is a need to develop an appropriate database which would provide a standardised means of assessment, not only of MRI, but also of essential clinical information. As part of the program of Concerted Action in Multiple Sclerosis, funded by the Commission of the European Community (CEC), we have developed a database for recording serial brain MRI results. The database consists of core, entry and follow-up sections. Both entry and follow-up parts are subdivided into clinical, MR system and MRI data. We expect that the use of this database will maximise efficiency of MRI monitoring in MS treatment trials, particularly in multicentre studies.

    Notes: Abstract MRI readily detects the lesions of multiple sclerosis (MS) in the brain and spinal cord. Conventional MRI sequences do not, however, permit distinction between the various pathological characteristics (oedema, demyelination, axonal loss and gliosis) of lesions in MS. Magnetisation transfer (MT) imaging may be more specific in distinguishing the pathologies responsible for disability in MS, namely demyelination and axonal loss, and therefore may have a potential role in monitoring treatment. We have applied MT imaging to the cervical spinal cord to see if it is feasible to measure MT ratios (MTR) in this region where pathological changes may result in considerable disability. We studied 12 patients with MS and 12 age- and sex-matched normal controls using a sagittal T2-weighted fast spin-echo sequence with and without an MT pulse. The median value for cervical cord mean MTR measurement in normal controls was 19.30 % units (interquartile range 19.05–19.55), whereas values were significantly lower in MS patients (median = 17.95 % units, interquartile range 17.25–19.00, P = 0.0004). There was a low intrarater variability for repeated mean MTR measurements. We conclude that it is possible to measure MTR in the cervical spinal cord, that a significant reduction occurs in patients with MS, and that there may be a role for this measure in future MS treatment trials.

    Notes: Abstract We report assessment of a new three-dimensional fast spin echo (3D FSE) sequence in ten patients with clinically definite multiple sclerosis, comparing it with standard 2D FSE, and in ten normal controls. We saw 29 focal lesions on the 2D images and 53 on the 3D FSE imgages (P = 0.05); none were seen in controls. Lesion length was significantly smaller on the 3D FSE than on to the 2D FSE images (3D: 1.36; 2D 2,0; P = 0.03). This may relate in part to separation into several lesions on the 3D images of confluent abnormal signal seen on 2D and in part to detection of small lesions missed by the thicker 2D FSE slices (3 mm compared to 1.5 mm). The 3D FSE sequence looks promising in improving spinal cord imaging.

    Notes: Summary In a fatal case of central pontine myelinolysis (CPM) a low field strength (0.08 Tesla) magnetic resonance image revealed reduction of image intensity in the pons with sparing of two central symmetrical areas in the ventral portion. The latter correlated with preservation of centrally located groups of longitudinal myelinated nerve fibres shown at autopsy. Although such sparing is well recognised in pathological studies of CPM it has never previously been demonstrated in life.
