    Description / Table of Contents: Zusammenfassung 5-Aminoacridin-hydrochlorid (0.4g gelöst in 50%igem Äthanol) erwies sich nach einer Vorfixierung mit Methanol-Formol (9:1 V/V) als ein hervorragendes Fixativum für die Erhaltung der wasserlöslichen Granula der menschlichen Blutbasophilen. Die Granula waren in allen Präparaten gut erhalten und wiesen keine Zeichen einer Diffusion auf. Im Gegensatz zu den anderen spezifischen Fixativa erzeugt 5-Aminoacridin-hydrochlorid in Kernspurenemulsionen keine chemographischen Effekte. Die Substanz eignet sich damit besonders für die autoradiographische Untersuchung von Blutbasophilen des Menschen.
    Notes: Summary 5-aminoacridine hydrochloride (0.4% in 50% aqueous ethanol) proved to be a useful fixative for the preservation of human blood basophils pretreated with methanol-formaldehyde (9:1 V/V). Leaching of the water soluble basophil granules or other artificial cell alterations common to fixatives in current use were not observed. The organic compound used did not induce any chemographic artifacts in nuclear tracers, in contrast to most of the other fixatives hitherto applied. 5-aminoacridine hydrochloride is therefore particularly suitable for autoradiography of the human blood basophils.

    Notes: Abstract The pharmacokinetics of free and different liposomal formulations of hexadecylphosphocholine (HPC) was investigated in tumor-bearing (human mammary tumor MaTu) and tumor-free mice after intravenous and intraperitoneal administration. The levels of HPC were evaluated at different times in serum, normal tissues, and tumor. The purpose was to test the hypothesis that the enhanced therapeutic efficacy of sterically stabilized HPC liposomes in comparison to conventional vesicles and free HPC is due to its pharmacokinetics. Conventional non-compartmental pharmacokinetic analysis and an elaborate three- and four-compartmental model were used for explaining the experimental data. The serum levels of HPC obtained with sterically stabilized liposomes were only consistently higher in comparison to conventional vesicles and free HPC in the first 4 h. In the xenografted MaTu carcinoma, the differences of the HPC content between the different groups are unexpectedly low and do not reflect the high therapeutic activity [5] of sterically stabilized HPC liposomes. Detailed analysis shows that the liposomally encapsulated drug displays a modified pharmacokinetic behavior, which may also involve lymphatic absorption of the liposomal drug.

    Notes: Abstract New sterically stabilized liposomes derived from the antitumoragent hexadecylphosphocholine with reduced uptake by the mononuclearphagocyte system and improved antitumor activities were developedand tested. The bilayer of such sterically stabilizedliposomes consists of hexadecylphosphocholine, cholesterol and polyethylene glycol-linkedphosphoethanolamine. The measurement of carbon clearance in miceshows that these stabilized liposomes, in contrast toconventional alkylphosphocholine liposomes, are not largely engulfed bythe mononuclear phagocyte system. Their therapeutic activity onexperimental human breast carcinomas MaTu, MT-1 and MT-3was tested in nude mice. Especially in theMaTu models the sterically stabilized hexadecylphosphocholine liposomes resultedin significantly reduced tumor growth in comparison toconventional hexadecylphosphocholine liposomes or free hexadecylphosphocholine. The enhancedtherapeutic efficacy of sterically stabilized hexadecylphosphocholine liposomes isprobably related to the extended circulation time ofthe formulation and its accumulation in tumors.

    Notes: Summary Percutaneous lumbar discectomy has gained growing interest during recent years as an alternative to open surgery for protrusions and non-sequestrated subligamentous intervertebral disc herniations. As a less invasive method it competes with chemonucleolysis. At least two modifications are known to date: automated percutaneous lumbar discectomy (APLD) with a 2-mm suction probe and non-automated, discoscopy-monitored percutaneous lumbar discectomy with a suction rongeur and a motor-driven shaver (NAPLD). In this study these two methods are compared for the amount of material extracted, dependence upon the degree of degeneration of the disc and intrinsic technical problems, using 20 human cadaver lumbar specimens for experimental testing. Total nucleotomy was not possible with either method. APLD yielded significantly less material and proved to be less effective in severely degenerated intervertebral discs than the NAPLD procedure, as preexisting gaps within the degenerated nucleus pulposus allowed the tissue to shift away from the tip of the 2-mm probe and facilitated displacement of the probe within the anulus fibrosus. By contrast, the rongeur, which first cuts the material to be removed and then carries it away by suction, was much more effective. Further data to support the advantages of non-automated percutaneous nucleotomy are discussed. Pretreatment of the disc with chymopapain did not result in a higher yield of nucleus material when combined with APLD.

    Description / Table of Contents: Summary A new technique is described which enables a selective tinctorial visualization of human blood basophils in autoradiographic preparations. Consecutive administration of 5-aminoacridine hydrochloride and aldehyde fuchsin rendered a sufficient preservation of basophils preventing leaching of their water soluble granules. Extensive control tests provided sufficient evidence to exclude chemographic artifacts produced by other histochemical procedures. The technique eliminates the obstacles to study the kinetics of human blood basophils and their relationship to atopic hypersensitivity reactions applying tritiated thymidine and radioiodine labeled lgE or corresponding antisera.
    Notes: Zusammenfassung Es wird eine neue Technik beschrieben, die eine selektive Darstellung der menschlichen Blutbasophilen in autoradiographischen Präparaten ermöglicht. Durch sukzessive Anwendung von 5-Aminoacridin-Hydrochlorid und Aldehyd-Fuchsin werden die wasserlöslichen Strukturen der menschlichen Blutbasophilen für den autoradiographischen Prozeß stabilisiert, ohne daß dabei chemographische Artefakte auftreten. Die Methode erlaubt es, die Kinetik der Blutbasophilen mit markierten Kernbasen sowie ihre Rolle im Rahmen atopischer Reaktionen durch Anwendung von radiojodmarkiertem Immunglobulin E oder von entsprechendem Antiserum zu studieren.

    Notes: Summary This study examines thein vitro andin vivo activity of alkylphosphocholines (APC) in experimental human breast carcinomas. Three analogs, hexadecylphosphocholine (HPC), octadecylphosphocholine (OPC) and eicosanylphosphocholine (EPC) were investigated. Three hormone receptor negative cell lines were sensitive to all three APCsin vitro whereas the receptor positive MCF-7 line was more resistant. Sensitivity was seen in 4/6 hormone receptor negative tumorsin vivo, with HPC being the most active analog. There were no antitumor effects in the four receptor positive models. The reasons for these differences in response between hormone receptor negative and -positive lines are not yet understood and require further study. Gastrointestinal toxicity and hemolysis, the major side effects of the APCs, were reduced by the use of liposomal preparations.

    Notes: Abstract The melting of monolayers of nine halomethanes and haloethanes physisorbed on exfoliated graphite is discussed on the basis of heat capacity and X-ray diffraction results. It is shown that the melting transition of incommensurate, triangular, orientationally disordered phase is continuous, whereas other two–dimensional solid phases melt discontinuously.

[3H]Thymidine-labelled basophils in buffy coat smears were fixed in methanol-formaldehyde followed by 5-aminoacridine hydrochloride and stained with basic Aldehyde Fuchsin prior to autoradiographic processing. The technique described represents a simple method for quantitative autoradiographic studies on basophil kinetics and the interaction of these cells with IgE-mediated atopias.
