    Notes: Zusammenfassung Elastisches Material aus dem Peritenonium der Rattenschwanzsehnen wurde elektronenmikroskopisch untersucht. Glatte, unverwundene Protofibrillen mit einem Durchmesser von 80 Å, die zur 10–15 zur Elastinfibrille (350 Å) vereinigt sind, wurden beobachtet. Die Elastinfibrillen sind zu Strängen von 800–1200 Å zusammengefaßt, verflochten und mit Kittsubstanz verbacken1. Es werden physikalische und chemische Daten erörtert sowie eine mögliche Theorie der molekularen Zusammenhänge.

    Notes: Abstract Two human ring chromosomes 18 were isolated intact in a Langmuir trough, critical-point dried, and studied by quantitative electron microscopy. The portion of deleted genetic material is calculated from dry-mass determinations and is found to be comparable to the total genetic material present in the bacterium Salmonella typhimurium.

    Description / Table of Contents: Zusammenfassung Es wurde die Wirkung von Pufferlösungen, die Dextran, Sucrose und verschiedene organische Ionen enthalten, auf das Volumen von Blut- und Asciteszellen während der Fixation mit Osmiumtetroxyd untersucht. Die Ergebnisse stimmen mit ähnlichen Befunden anGewebsstücken, über die früher berichtet wurde, überein. Die Gegenwart von Osmiumtetroxyd verursacht eine bemerkenswerte Zellschwellung, die mittels Hämatokrit-Technik bestimmt wurde. Die Schwellung kann durch eine angepaßte Zusammensetzung verschiedener Fixierungszusätze ausgeglichen werden. Über die Bedeutung dieser Zusätze wird diskutiert.
    Notes: Summary The effect of buffered media containing dextran, sucrose and different organic ions, on the volume of blood and ascitescells during fixation with osmium tetroxide has been studied. The findings are in agreement with similar studies ontissue pieces as reported earlier. The presence of osmium tetroxide causes a marked swelling of the cells as estimated by hematocrit techniques. The swelling can be balanced by an appropriate composition of the fixation vehicle, the significance of which is discussed.

    Notes: Abstract Quantitative electron microscopy was used to analyze surface-spread, critical-point-dried human interphase nuclei and chromatin. The following information is presented: (1) Unstimulated interphase nuclei of lymphocytes from peripheral blood have a mean dry mass of 50.30×10−12 g. The mean dry mass of stimulated nuclei of lymphocytes was determined to be 59.34×10−12 g, a significant statistical difference from the unstimulated ones. (2) Mean diameter of chromatin fibers and mean fiber mass per micron were 199ű15% coefficient of variation (C.V.) and 5.95×10−16g×29% C.V., respectively. (3) A line of regression of fiber mass on fiber diameter for 83 fibers indicated that a 200-Å fiber has a mass of 5.86×10−16g/μ, or almost the same as the mean fiber mass of 5.95× 10−16g/μ. (4) With the value 7×10−12g for the DNA content of an unstimulated lymphocyte nucleus, a total length of 215 cm is calculated for the DNA double helix. When this length is compared to the mean length of chromatin fiber per nucleus (7.59 cm), a ratio of 28.3 to 1 results, which is called the DNA-packing ratio. (5) This DNA-packing ratio of 28.3 is reasonably close to the packing ratio of 26.9 suggested from model calculations for the second DNA supercoil in a 200-Å chromatin fiber.

    Notes: Abstract Pairs of centrioles are a frequent finding in whole-mounted, critical-point-dried chromosome preparations from normal and irradiated human lymphocyte cultures. Most frequently they are found in association with a group C chromosome, although apparently free diplosomes are not uncommon. Fibers that in every respect are the morphologic equivalent of those seen in the body of the chromatid connect to the thick-walled open-ended part of the parent centriole. These features argue for the possibility that fibers connecting centrioles are an integral part of nuclear or chromosomal chromatin. It was observed that the smaller (daughter) centriole was connected and held in the well-known angular configuration to the thickwalled open end of the parent centriole by a few, probably only two, colchicine-resistant fibers.

    Notes: Abstract The fluorescent stains, dansyl chloride and fluorescamine, were used to indicate the amount of protein in G-banded and unbanded chromosomes 1 of the Chinese hamster relative to the average amount of protein in human erythrocyte. Fluorescence was found to be proportional to protein mass up to the equivalent of three erythrocytes. In G-banding, trypsin digestion resulted in an average protein loss of 35.4% compared with unbanded chromosomes.
