    Notes: Abstract The mixed microbial flora of 3 lakes in Ohio with differing histories of hydrocarbon pollution was examined in relation to the ability to use hydrocarbons. Weathered kerosene was spiked with naphthalene, pristane, 1,13-tetradecadiene, andn-hexadecane and added to water-sediment mixtures from the 3 lakes, and utilization of the 4 marker hydrocarbons was measured. Each of the marker hydrocarbons was metabolized; naphthalene was the most readily used and pristane was the most resistant. Values for dissolved oxygen suggest that oxygen did not limit hydrocarbon degradation in the water column at any site examined. Nutrient addition studies indicated that nitrogen and phosphorus limited hydrocarbon degradation at all sites examined. Maximum numbers of heterotrophic bacteria were detected when the water temperature was 10°C or higher. The data indicate that temperature limits hydrocarbon degradation in the winter, except at a site which had been impacted by an oil spill and which received chronic inputs of hydrocarbons and nutrients. In samples from that site, all 4 marker hydrocarbons were degraded at 0°C. Results of temperature and nutrient-addition experiments suggest that different seasonal populations of hydrocarbon users are selected at that site, but not at other lake sites.

    Description / Table of Contents: Zusammenfassung Blut gerinnt, wenn das Gerinnungsferment Thrombin aktiviert wird und Fibrinogen in Fibrin umwandelt. Störungen der Thrombin- oder Fibrinbildung können Bluterkrankheiten verursachen. Die Ursache angeborener Bluterkrankheiten liegt entweder in der quantitativen Verminderung oder in qualitativen Strukturanomalien eines Gerinnungsfaktors. Immunologische Methoden im Verein mit einer beginnenden, direkten Strukturabklärung zeigten für das Fibrinogen überzeugend, dass beide Möglichkeiten vorkommen. Unterschiedliche Funktionsausfälle von Fibrinogen lassen vermuten, dass verschiedenartigen Strukturanomalien für Störungen der Substrateigenschaften gegenüber Thrombin, der Polymerisierung oder der Fibrinstabilisierung verantwortlich sind. Ähnliche Untersuchungen an den Faktoren II, VII, VIII, IX, X und XIII zeigen, dass wahrscheinlich die Mehrzahl der bekannten Bluterkrankheiten durch Strukturanomalien verursacht werden. Die eingehende Untersuchung solcher Strukturdefekte wird zukünftig um so interessanter und bedeutungsvoller werden, je besser die Struktur und Funktion der normalen Gerinnungsfaktoren bekannt sind.
    Notes: Summary Blood coagulation reflects, the complex biological system resulting in the formation of thrombin and the subsequent conversion of fibrinogen into fibrin. Disorders of either thrombin or fibrin formation may cause bleeding states. Congential hemorrhagic disorders are characterized by either reduction of a coagulation factor or impairment of its functional capacity by structural aberrations. Immunological studies and direct molecular evaluation of fibrinogen demonstrate that both types of abnormalities occur. Functional evaluation of abnormal fibrinogens suggests that many different structural variants exist and variably influence interaction with thrombin, fibrin polymerization and fibrin ‘stabilization’. Similar studies performed on factors II, VII, VIII, IX, X and XIII indicate that qualitative abnormalities are more frequent than ‘true’ deficiency states. Such molecular abnormalities will become increasingly interesting and important as the biochemistry of normal coagulation progresses.

    Notes: Summary Addition of human placental umbilical cord serum to bone marrow cultures reproducibly increased the number of granulocyte colonies in vitro. This stimulatory effect was significantly greater than that of fetal calf serum which was seen in cultures of human bone marrow under the conditions described.

    Notes: Abstract Bovine factor VIII/platelet aggregating factor was adsorbed onto gold granules and the protein-gold complex added to either formalin-fixed or fresh washed human platelets. Following aggregation, binding of gold granules to the platelets was measured by monitoring the optical density of colloidal gold remaining in the supernatant. Scatchard analysis of binding data indicated that multiple classes of binding sites were present. The number of high affinity binding sites per formalin-fixed platelet dependend on the concentration of ristocetin: 420 gold granules were calculated to bind at 1.4 mg/ml of ristocetin, 610 at 0.6 mg/ml of ristocetin and 875 when no ristocetin was added. Fresh washed platelets bound 1350 granules per cell in the absence of ristocetin. We conclude that during platelet aggregation, induced by bovine factor VIII, the binding sites on the platelet surface are only partially occupied.

    Notes: Summary Dextran, glycerol and dimethyl sulfoxide (DMSO), alone or in combination, were used for cryoprotection of human bone marrow cells. The viability of cryopreserved cells was assessed by culture of myelopoiesis-committed stem cells (CFU-c) in vitro. A significantly better protection against freezing injury was obtained by 9% dextran in combination with 3 or 5% DMSO, and also with 5 or 10% DMSO alone, than with either 15% glycerol or 9% dextran with 1% DMSO.
