    Notes: Abstract We present details of a method which allows for the determination of chondroitin sulphate turnoverin vivo using the guinea pig. Such methods have been utilised to examine the effects of diacetyl rhein, a compound with purported anti-osteoarthritic activity, and several related anthraquinone analogues on the turnover of chondroitin. Since the guinea pig develops spontaneous osteoarthritis, this may give useful information on the potential for such compounds to inhibit the progression of osteoarthritis. The results show that several of the anthraquinones are capable of reducing the turnover of chondroitin 4- but not 6-sulphate. This may indicate potential mechanisms for the breakdown of guinea pig cartilage aggrecans. We propose that these techniques could be useful for the screening of chemical agents with useful activity against osteoarthritis.

    Notes: 1. The effect of dietary salt restriction on blood pressure was measured in patients with moderate hypertension receiving antihypertensive medication. Patients were studied during a randomly allocated 6 week period of moderate dietary salt restriction and during a 6 week period of normal diet.2. Twenty-four of twenty-eight patients achieved a reduction of salt excretion exceeding 20%. In these patients mean urinary sodium excretion fell from 169 (s.e.m. = 13) mmol/24 h to 92 (s.e.m. = 7) mmol/24 h. Potassium excretion and body weight did not change, and with the exception of the erect systolic pressure, there was no overall change in blood pressure.3. When the data were grouped according to the patient medication, mean supine systolic pressure fell from 148.9 (s.e.m. = 4.4) to 140.3 (s.e.m. = 5.0) mmHg (n= 14, P〉0.05) and mean supine diastolic pressure fell from 100.8 (s.e.m. = 2.9) to 97.0 (s.e.m. = 2.8) mmHg (n= 14, P〉0.05) in patients taking diuretics. Blood pressure did not change in patients whose medication did not include diuretics.4. Moderate dietary salt restriction may be therapeutically useful in patients with moderate hypertension treated with diuretics but is of little value in those not receiving diuretics.

Because in vitro adenylate cyclase activity studies suggest that parathyroid hormone (PTH) and calcitonin (CT) increase renal tubular calcium and magnesium reabsorption by stimulating the same transport mechanism, the separate and combined effects of these hormones on calcium and magnesium transport was assessed in the rat.2. Fractional excretion of calcium was 9.5 ±0.5% in calcium-infused thyroparathyroidectomized (TPTX) rats and was reduced to 5.1 ±0.6 and 4.1 ±0.6% by maximal hypocalciuric concentrations of PTH and CT, respectively.3. Combined administration of maximal PTH and CT produced an effect on fractional and absolute calcium excretion similar to that recorded with maximal CT alone. The combined administration of half-maximal concentrations of PTH and CT also produced a comparable fall in the fractional excretion of calcium to 3.0 ± 0.7%, which was similar to that observed in the ‘maximal CT’ and ‘maximal PTH plus CT’ groups. The magnesium reabsorption data were comparable.4. These results support biochemical data suggesting that PTH and CT act upon the same transport site, presumably within the thick ascending limb of Henle's loop, to facilitate calcium and magnesium reabsorption.

    Notes: The molecular identity of a gene which encodes the pore-forming subunit (α1G) of a member of the family of low-voltage-activated, T-type, voltage-dependent calcium channels has been described recently. Although northern mRNA analyses have shown α1G to be expressed predominantly in the brain, the detailed cellular distribution of this protein in the central nervous system (CNS) has not yet been reported. The current study describes the preparation of a subunit specific α1G riboprobe and antiserum which have been used in parallel in situ mRNA hybridization and immunohistochemical studies to localize α1G in the mature rat brain. Both α1G mRNA and protein were widely distributed throughout the brain, but variations were observed in the relative level of expression in discrete nuclei. Immunoreactivity for α1G was typically localized in both the soma and dendrites of many neurons. Whilst α1G protein and mRNA expression were often observed in cells known to exhibit T-type current activity, some was also noted in regions, e.g. cerebellar granule cells, in which T-type activity has not been described. These observations may reflect differences between the subcellular distribution of channels that can be identified by immunohistochemical methods compared with electrophysiological techniques.

    Notes: Abstract Production of a granulomatous tissue adjacent to cartilage implants in subcutaneous tissues of mice by prior wrapping of cartilage with cotton, induces matrix depletion and a rise in serum levels of the acute phase protein haptoglobin. The granulomatous reaction to cotton could be inhibited partially, following treatment of mice with indomethacin, dexamethasone and cyclophosphamide but only the latter two reduced cartilage matrix loss.d-penicillamine was inactive. Regarding the acute phase response, dexamethasone andd-penicillamine appeared to lower and indomethacin and cyclophosphamide to elevate, serum levels of haptoglobin, but these effects were not statistically significant.
