    Notes: Abstract  Two monoclonal antibodies (Vx-BB8 and Vx-EA11) to the chemical warfare agent Vx were produced and characterized. A competitive inhibition enzyme immunoassay was developed to detect Vx concentrations as low as 3.7×10-7–3.7×10-6 mol/l in biological samples. Vx-BB8 400 μg given intravenously immediately before 1×LD95 Vx or 400 μg Vx-BB8 intraperitoneally 1.5 h–3 days before 1×LD95 Vx could protect all the tested mice from death.

    Notes: Abstract  Two artificial antigens, N α N ɛ-di(O, O-diisopropyl) phosphoryl L-lysine (DIP)- bovine serum albumin (BSA) conjugate (DIP-BSA) and DIP-KLH (keyhole limpet hemocyanin), were synthesized. Antibodies against sarin (O-isopropyl methylphosphonofluoridate) were obtained after immunization of rabbits with DIP-KLH conjugate. A competitive inhibition enzyme immunoassay (CIEIA) was developed to detect the organophosphorus nerve agent sarin. The antibody solutions could be inhibited by sarin as low as 10-6 mol/l, and the standard curve was linear over 3 orders of magnitude. The coefficients of intraassay and interassay variation of this method were 5.4–6.2% (n=11) and 8.0–9.5% (n=6) at a sarin concentration range of 10-3–10-6 mol/l, respectively. The recovery of sarin in water samples at the concentration of 5×10-5 mol/l was in the range of 96.8–102.5%. The specificity of the antiserum was assessed by comparing the inhibition induced by sarin with soman, Vx, isopropyl alcohol and isopropyl methyl phosphonic acid. The results showed that less than 5 mmol/l soman, 2 mmol/l Vx, 16 mmol/l isopropyl alcohol and 8 mmol/l isopropyl methyl phosphonic acid did not influence the determination of sarin in water samples.

    Notes: Summary The fluorescence enhancement of terbium(III) by nucleotides (AMP, ADP, ATP, GMP, GDP, GTP) and polyhomonucleotides [poly(A), poly(G), poly(C), poly(U)] in the presence of phenanthroline (phen) was studied. Investigation of the composition of the terbium(III)/ANP(AMP, ADP, ATP)/phen complexes and conditions of optimization suggest a 1:2 molar ratio of terbium(III) and phen for the ternary complexes. The results showed that the presence of phen enhanced the net fluorescence of terbium(III)/ANP, poly(A), poly(C) or poly(U) from several fold to more than one-hundred fold, while it has little effect on the fluorescence of terbium(III)/GNP(GMP, GDP and GTP) or the poly(G) system. The possibility of spectrofluorimetric measurements of these compounds were studied under optimal conditions (pH 7.0 in tris-HCl buffer; Ex=298 nm, Em=543.5 nm). The detection limits were 2.0×10−7, 6.0×10−7 and 1.0×10−6 mol/l for AMP, ADP and ATP, respectively. The relative standard deviations (6 replicates) were within 2.0% in the middle of the linear range.

    Notes: Summary A rapid fluorimetric method employing the complexes of Al3+ with tetracycline (TC) and anhydrotetracycline (ATC) has been used for the measurement of the concentrations of TC and ATC in about 10−7–10−6 mol/l. The assay offers simplicity and rapidly compared with other methods. The recoveries in the determination of synthetic samples are in the range of 95–103% for TC and 101–120% for ATC. Several determinations of TC and ATC in serum and urine have been carried out with satisfactory results.

    Notes: Abstract A new fluorometric immunoassay for methotrexate (MTX) was developed using the mimetic enzyme Mn(III)-tetrakis(sulfophenyl) porphine (Mn-TPPS4) as a labelling reagent to catalyze the fluorescence reaction of 4-hydroxyphenylacetic acid (HPA) and hydrogen peroxide. In the competitive assay MTX (antigen) from human serum and Mn-TPPS4 labelled MTX competitively react with antibody (anti-MTX) coated on a polystyrene 40-well plate. The Mn-TPPS4 in the bound fraction, after the separation from the free fractions, was determined by measuring the fluorescence intensity between HPA and H2O2 catalyzed by bound Mn-TPPS4 and MTX conjugate, which was anti-proportional to the concentration of MTX in the serum. MTX can be determined between 0.5 and 10 μg/well with a detection limit of 100 ng/well.
