    Description / Table of Contents: Summary A report is given on neoplasmata in the CNS of animals, observed at the Institute of Veterinary Pathology of the University of Munich during the years 1960–1961. These neoplasmata were the following: perineural fibroblastoma in the trigeminal nerve of a bastard; hamartoma (epithelial ependymoma?) in the spinal cord of a boxer; ependymoma in the olfactory bulb of a cat; glioblastoma multiforme in the cerebellum of a boxer; nonclassified tumour in the cerebellum of an Alsatian; oligodendroglioma in the piriform lobe of a dwarf bully. Due to the difficulties in classifying these cases with certainty their diagnoses are critically viewed, and put up for discussion.
    Notes: Zusammenfassung Bericht über die am Institut für Tierpathologie der Universität München in den Jahren 1960 und 1961 beobachteten Neubildungen des ZNS bei Tieren, und zwar: Perineurales Fibroblastom am N. trigeminus eines Bastards; Hamartie (epitheliales Ependymom?) im RM eines Boxers; Ependymom im Bulbus olfactorius einer Katze; Glioblastoma multiforme im Großhirn eines Boxers; unklassifizierter Tumor im Kleinhirn eines Schäferhundes und Oligodendrogliom im Lobus piriformis bei Zwergbully. Wegen der Schwierigkeit, alle beobachteten Fälle sicher zu klassifizieren, werden die Diagnosen eingehend kritisch beleuchtet und zur Diskussion gestellt.

    Notes: Summary During the past five years a disease involving mainly the CNS was observed among adult and older animals in a flock of goats. Debilitating factors appear to favour the onset of the clinical manifestation. These are usually ushered in by a short febrile episode. The course, as a rule, is chronic with frequent recurrences. Clinical signs include ataxia, coarse tremor, nystagmus, torticollis, trismus, excess salivation, weakness of the hindquarters or complete paralysis. The serum globulins are abnormally elevated, particularly the gamma fraction. High levels of total protein, glucose and transaminases are common in the CSF. Neuropathologically, two types of changes are found, often blending together in a smooth fashion, namely, a chiefly granulomatous lesion in periventricular, paraventricular regions and meningeal spaces respectively and a predominantly demyelinating process reaching out from the paraventricular areas into the adjacent nervous parenchyma. A neurologic illness of the domestic goat—clinically and pathologically very similar to the spontaneous gMEM—was repeatedly obtained by injection of a cell-free filtrate into the cisterna magna prepared from a saline suspension of organ-material of a gMEM-animal. Final etiologic elucidation of this affection in goats, however, is yet to be accomplished.

    Notes: Summary Ultrastructural studies of the retinae in two NCL-affected Dalmatian dogs revealed ubiquitous accumulation of lipopigments in numerous cell types of the retina, the fine structure of which closely resembled that seen in NCL-affected English setters. Photoreceptors and other retinal cell types were largely intact. These findings show that the retinal involvement in NCL of our Dalmatian dogs is identical to that of NCL-affected English setters. It also shows that in canine NCL a severe retinopathy, regularly encountered in human childhood NCL, does not develop. Thus, the NCL of Dalmatian dogs —and English setters — represents a reliable model to study human NCL, but for human retinopathia pigmentosa perhaps only at its earliest stage.

    Notes: Summary The human glioblastoma-derived cell lines 86HG-39, 87HG-28 and 87HG-31, used for the production of monoclonal antibodies (mAbs) against glioma-associated antigens (GAA), were characterized in terms of morphology, growth behaviour, chromosomes and antigen expression. In the primary tumours, differential expression of glial fibrillary acidic protein, S100 protein, Leu-7 and GAA as defined by mAbs MUC 2–39, MUC 2–63 and MUC 8–22 was demonstrated. Receptors for epidermal growth factor (EGFr) and nerve growth factor (NGFr) were found in many cells in short-term cultures, but the transferrin receptor (Tr) was found in only a few cells of 87HG-28. In permanent cell lines, differentiation antigens and EGFr decreased and Tr increased markedly. NGFr and GAA remained stable. Transplantation tumours of 86HG-39 were partly positive for Tr and GAA. Chromosomal analysis revealed that the 86HG-39 and 87HG-28 cell lines had a hypodiploid or diploid stem line with lines in the hypotetraploid to tetraploid region for 50 in vitro passages. The 87HG-31 cell line had chromosomal patterns in the hypotriploid to triploid region. A gain of chromosomes was seen in the groups C7, C8, C10, D14, F19, F20, G21, G22. The variability of antigens in these tumours and especially during long-term cultivation probably reveals an ability to influence the growth of malignant glioma cells via the respeective effector molecules.

    Description / Table of Contents: Zusammenfassung Bei 8 graviden Kaninchen wurde Äthylnitrosoharnstoff (ÄNH), gelöst in Citronensäure/Dinatrium-Phosphatpuffer (pH 6,0), in einer Konzentration von 50 mg/kg intravenös appliziert. Die Injektion der jeweils frisch angesetzten ÄNH-Lösung (0,1M) erfolgte am 8. bzw. 10. Graviditätstag in die Ohrrandvene (Vena auricularis lateralis). Von den 21 Nachkommen konnten nur 12 Tiere aufgezogen werden. Acht davon, die sich normal entwikkelt hatten, zeigten nach 2 1/2- bis 3jähriger Latenzzeit neurologische Symptome. Nach Tötung und histologischer Untersuchung konnte das Vorliegen von Tumoren im Bereich des peripheren (9) und zentralen (2) Nervensystems festgestellt werden. Die Tumoren des peripheren Nervensystems lassen sich als Neurinome reticulären Charakters (Antoni B-Typ) klassifizieren, deren Zellen stellenweise von basalmembranartigen Strukturen umgeben werden. Bei den neoplastischen Prozessen des Hirns handelt es sich um mikroskopisch nachweisbare Transformationsareale sowie um Gliome (2) in der Umgebung des III. Ventrikels. Die vorliegenden Befunde zeigen, daß die ÄNH-Exposition während der Organogenese zur Entstehung neurogener Tumoren im Erwachsenenalter der pränatal geschädigten Kaninchen führt.
    Notes: Summary The oncogenic effect of ethylnitrosourea (ENU) on rabbits during the organogenesis was studied. ENU, dissolved in sodium phosphate (pH 6.0), was injected in a 50 mg/kg dosis intravenously in pregnant dams between the 8th and 10th day of gestation. From 12 offspring 8 grew up normally and appeared unremarkably until 30–36 months of age when neurologic signs first developed. Six rabbits showed one or more tumors along the peripheral nerves. Two rabbits were bearing brain gliomas in the vicinity of the 3rd ventricle. The tumors of the peripheral nervous system (PNS) were of variable size and soft consistency; they had a cut surface of spongy and even cystic appearance. Histologically, the PNS tumors resemble Schwannomas of the Antoni B type. An ultrastructural study of these neoplasms disclosed cells featuring a basal lamina type of lining characteristics of Schwann cells. These findings demonstrate that the exposure of rabbits to ENU during organogenesis results in the induction of neurogenic tumors which become clinically manifest late in life. It seems, therefore, that the selective oncogenic effect of ENU on the nervous tissues of the exposed animals depends exclusively on the stage of organogenesis at which the drug is brought to bear upon the target organ anlage.

    Description / Table of Contents: Zusammenfassung Die onkogene Wirkung von N-Methyl-N-Nitrosoharnstoff wurde beim Hanford-Miniaturschwein untersucht. Die Substanz wurde jeweils unter Zusatz von 1% NaH2PO4 in physiologischer Kochsalzlösung gelöst (pH 6,1 – 6,2) und den Versuchstieren peroral in 14tägigen Abständen bis zum Versuchsende verabfolgt. Nach einer Versuchsdauer von 4 1/2 Jahren wurden die klinisch unauffälligen Tiere getötet und untersucht. Hierbei zeigte sich, daß alle Tiere Magentumoren entwickelt hatten. Bei den induzierten Tumoren handelt es sich - von einer Ausnahme abgesehen- um multiple, exophytisch wachsende, größtenteils gestielte, blumenkohlartige Schleimhauttumoren, die vorwiegend in der Kardia- bzw. Fundusregion lokalisiert sind. In einem Falle fand sich zusätzlich ein im Bereich der Submucosa der Kardia lokalisierter Tumor. Aufgrund ihrer histologischen Struktur lassen sich die Schleimhauttumoren als adenomatöse Polypen klassifizieren. Einige dieser in der Regel benignen Tumoren tragen cytologische Merkmale einer beginnenden malignen Entartung. Bei dem in der Submucosa lokalisierten Tumor handelt es sich vermutlich um ein malignes Neurinom. Die vorliegenden Ergebnisse zeigen, daß die Organotropie bestimmter onkogener Nitrosamide u.a. auch speciesabhängig sein kann. Diese Erkenntnis dürfte nicht nur hinsichtlich der Morphogenese von Magentumoren sondern darüberhinaus auch der Ätiopathogenese von Bedeutung sein.
    Notes: Summary The oncogenic effect of N-methyl-N-nitrosourea was studied in Hanford miniature-pig. The compound - dissolved in 0,9% NaC1 (pH 6,1 – 6,2) and supplemented with 1% NaH2PO4 - was administered per os to the animals in intervals of two weeks during 4,5 years. Finally the animals, which had behaved clinically inapparent during the whole run of the experiment, were sacrificed and autopsied. All animals of the experimental group had developed stomach tumours, whereas control animals showed normal conditions of gastric mucosa. Except one, all tumours were grouped macroscopically as multiple, exophytic, papillary, cauliflower-like neoplasms of the mucosa, located predominantly within cardia- and fundus-regions. Histologically they are to be classified generally as benign adenomatous polyps. However, a few of them also possess cytological characteristics of an early malign dedifferentiation. One - above excepted - tumour probably belongs to the class of malign neurinomas. The results of the experiment demonstrate in general that the obvious organotropic properties of certain oncogenic nitrosamides might be in a way also species-depending ones. This perception seems to be important in view of morphogenesis as well as of etiopathogenesis of stomach tumours.

    Description / Table of Contents: Zusammenfassung Unter allen überprüften Versuchstieren stellt das Schwein ein hervorragendes Versuchstier speziell für Untersuchungen auf den Gebieten des Fettstoffwechsels und der vergleichenden und experimentellen Arterioskleroseforschung dar. Die Tendenz zur frühzeitigen Entwicklung sogenannter spontaner Arterio- oder Präarteriosklerosen scheint allen bislang untersuchten Schweinerassen und selbst dem europäischen Wildschwein eigen zu sein. In ihrer formalen wie kausalen Pathogenese erweist sich die Arteriosklerose des Schweines als ein polyätiologisches Phänomen, das die grundsätzlich gleichen Wesenszüge wie die humane Arteriosklerose erkennen läßt. Dem Nachteil, das Hausschwein wegen seiner Größe und seiner aufwendigen Haltung und Fütterung als Versuchstier nur selten einsetzen zu können, soll in Zukunft durch die Verwendung sogenannter Miniaturschweine in der medizinischen Forschung begegnet werden. Aus diesem Grunde werden gegenwärtig umfangreiche Untersuchungen mit diesem Schweinetypus durchgeführt.
    Notes: Summary With regard to research-work concerning lipid-metabolism and arteriosclerosis the porcine species offers optimal conditions compared with all other experimental animals. Liability to develop very early so-called spontaneous arteriosclerosis or prearteriosclerotic lesions appears to be characteristic for all breeds of swine including European wild boars. Arteriosclerosis in swine may be defined as a polyetiological phenomenon, very similar to human arteriosclerotic lesions in regard to morphology as well as to distribution within the different areas of the arterial system. Contrary to these advantages in research-work, however, one should consider the expensiveness of managing and feeding of domestic swine and the very high body-weight obtained in long term experiments normally. Therefore, studies are carried out at the present time in order to determine, whether so-called miniature pigs do meet the requirements of research-work in the fields of lipid-metabolism and arteriosclerosis.
