Time course analyses of nitrous oxide (N2O) production by bacteroides of Bradyrhizobium j japonicum strain 122 DES (uptake hydrogenase positive, Hup+) and of its isogenic uptake hydrogenase negative (Hup−) mutant strain PJ17 showed that rates of N2O production under microaerobic (1% O2) conditions were higher in the Hup− strain. When succinate was present in the incubation mixtures, dissimilatory nitrate metabolism proceeded immediately upon the addition of nitrate, whereas in the absence of succinate a 6-h lag period was observed before N2O could be detected. Inclusion of 10% H2 in the incubation mixtures decreased N2O production by 122 DES bacteroides, both in the presence and in the absence of succinate, and did not affect rates of denitrification by bacteroides of strain PJ17.
