    Notes: Abstract Membranes from roots of Lepidium sativum L. were investigated in situ and after fractionation by applying morphological and biochemical methods. After freeze-fracture combined with filipin labelling the tonoplast and the plasma membrane could be easily characterized by the frequency of intramembranous particles and the arrangement of filipin-induced lesions. On tonoplast vesicles, the filipin-induced lesions were arranged in clusters of different size whereas they were evenly distributed on plasma membrane vesicles. Enrichment of tonoplast and plasma membrane in different fractions was documented by filipin labelling, phosphotungstic acid staining and by the profiles of marker enzyme activities and ATP-dependent H+-transport. Additionally, the presence of rightside-out and inside-out vesicles of both tonoplast and plasma membrane could be demonstrated. It was found that filipin labelling used in combination with freeze-fracturing is suitable for quantitative determinations of the percentages of tonoplast and plasma membrane in membrane fractions, which have been found to be more than 40% for the tonoplast and about 40% for plasma membrane in the respective enriched fractions.

    Notes: Summary The properties of minicell producing mutants of Escherichia coli deficient in gentic recombination were examined. Experiments were designed to test recombinant formation in conjugal crosses, survival following UV-irradiation in cells, and the state of DNA metabolism in minicells. The REC- phenotypes are unaffected by min +/- genotypes in whole cells. In contrast to minicells produced by rec + parental cells, minicells from a recB21 strain have limited capacity to degrade linear, Hfr transferred DNA. The lack of a functional recA gene product, presumably involved in inhibiting the recBC nuclease action(s), permits unrestricted Hfr DNA breakdown in minicells produced by a recA1 strain. This results in an increase in TCA soluble products and in the formation of small DNA molecules that sediment near the top of an alkaline sucrose gradient. Unlike the linear DNA, circular duplex DNA from plasmids R64-11 or λdv, segregated into the minicells, is resistant to breakdown. By using in vitro criteria, and [32P]-labelled linear DNA from bacteriophage T7 for substrate, we found that the ATP-dependent exonuclease of the recBC complex (exo V) is present in rec + and recA- minicells, and is lacking in the recB21 mutant. In fact, the absence of a functional exo V in recBC- minicells results in isolation of larger than average Hfr DNA from minicells. We suggest that recombination (REC) enzymes segregate into the polar minicells at the time of minicell biogenesis. This system should be useful for studies on DNA metabolism and functions of the recBC and recA gene products.

    Notes: Summary The enzymatic properties of purified DNA polymerase I from a strain of Escherichia coli K12 with a mutation in the polA gene have been studied. The polymerizing activity of the mutant enzyme is similar to that of the enzyme from isogenic wild-type cells, when the activity is measured on exonuclease III treated calf-thymus DNA. Also the 3′–5′ exonucleolytic activity is not significantly different for both enzyme preparations. The 5′–3′ exonucleolytic activity of DNA polymerase I isolated from the mutant strain, however, is much lower than that of wild-type DNA polymerase I. The products formed by the action of the wild-type and the mutant enzyme on nicked circular double-stranded DNA of phage ϕX174′ (RFII DNA) were analysed by sucrose-gradient sedimentation and electron-microscopy. When RFII DNA was incubated with wild-type enzyme 80% of the molecules were converted into linear molecules. All linear molecules were shorter than one phage genome. Only 25% of the molecules were branched. After incubation of RFII DNA with the mutant enzyme 62% of the molecules have become linear. More than 90% of these linear molecules were branched and the majority of them was longer than one phage genome.

    Notes: Summary Preparative free-flow electrophoresis has been employed in combination with density gradient centrifugation to prepare fractions enriched in either tonoplast or plasma membrane from dark grown seedlings of cress (roots), zucchini (hypocotyls), soybean (hypocotyls) and maize (coleoptiles). A polyclonal antibody to the 72,000 Mr subunit to the maize tonoplast ATPase was used to identify the tonoplast fractions from the free-flow electrophoresic separations and to show the absence of tonoplast contamination in plasma membranes derived from the same homogenates. These findings confirm the identity of the tonoplast fraction based on the presence of the proton translocating ATPase determined previously from sucrose gradient fractionation and inhibitor studies to be a tonoplast marker. Using staining with phosphotungstic acid at low pH, the plasma membrane fractions obtained after free-flow electrophoresis were shown to be 〉 90% plasma membrane-derived with little or no cross-contamination of plasma membrane vesicles in the tonoplast-containing fractions. Finally, the composition of the fractions was correlated with the characteristic morphologic appearance after filipin treatment and freeze-fracture. By means of morphometric analyses using this criterion, both the identity and the purity of the tonoplast and the plasma membrane fractions received further confirmation. Essentially homogeneous fractions were obtained by subjecting fractions already enriched by a centrifugation method to the final separation by free-flow electrophoresis.
