Notes: Abstract Bacterial products fmet-leu-phe (FMLP), muramyl dipeptide (MDP) and lipopolysaccharide (LPS) were assayed for their ability to alter the inflammatory response to λ carrageenan-induced pleurisy in Hooded Surgery rats. Continuously infused FMLP, or one initial i.v. dose of FMLP, MDP or LPS either ablated or partially suppressed the pleurisy. Total circulating leucocytes and neutrophils were suppressed by 55–65% when compared to the normal circulating leucocyte response to carrageenan pleurisy, excepting the protocol incorporating a single i.v. dose of FMLP where suppression was intermediate at 30%. There were also significant changes in the expression of FMLP receptors on circulating neutrophils. MDP and LPS induced a receptor number increase of 2 and 1.7 times initial value respectively, whilst a continuous FMLP infusion caused a receptor decrease to 0.3 times the initial value. The introduction of bacterial products at an alternative site to that of the pleurisy had an anti-inflammatory effect and the pleurisy was reduced.

Notes: Abstract The role of synchronism in systems of threshold elements (such as neural networks) is examined. Some important differences between synchronous and asynchronous systems are outlined. In particular, important restrictions on limit cycles are found in asynchronous systems along with multi-frequency oscillations which do not appear in synchronous systems. The possible role of deterministic chaos in these systems is discussed.

Notes: Summary The pharmacokinetics of isoxicam and piroxicam were compared in 12 young adults (〈40 years) and 12 elderly subjects (〉65 years). After a single oral dose of 200 mg isoxicam or 20 mg piroxicam blood samples were taken for 168 h and the plasma drug concentrations determined by HPLC. The elimination half life of piroxicam for the adults was 57.1±16.4 h (mean ± SD; harmonic mean 52.9 h) and for the elderly subjects was 57.8±22.1 h (harmonic mean 52.1 h). The corresponding values after isoxicam were 34.3±13.6 h (harmonic mean 31.6) for the adults and 39.1±22.7 h (harmonic mean 33.5) for the elderly subjects. Similarly no differences were noted in either the AUC0-∞ after piroxicam (adults 154.1±52.2 µg·h/ml, elderly 163.6±99.1 µg·h/ml) and isoxicam (adults 642.7±241.9 µg·h/ml, elderly 787.9±613.1 µg·h/ml) or the apparent oral clearance of piroxicam (adults 2.39±0.80 ml/min, elderly 2.51±0.90 ml/min) and isoxicam (adults 5.84±2.04 ml/min, elderly 5.59±2.12 ml/min). One adult and two elderly subjects exhibited slower elimination of both medicines than the remainder of each group. However determination of the oxidation phenotype using sparteine metabolism showed that this was not a likely determinant of the reduced clearance.

Notes: Summary The bioavailability of metoprolol was studied in eight healthy young and seven healthy elderly volunteers. Large interindividual differences in the bioavailability of metoprolol were observed in both groups. However, there was no significant difference in AUC, peak plasma concentration or elimination half-life between young and elderly, but time to peak concentration was significantly longer in the elderly. Pretreatment with metoclopramide had no effect on AUC but caused significant increases in peak concentration and decreases in time to peak concentration in both groups. Probanthine pretreatment (only to the young) resulted in a significant decrease in peak concentration of metoprolol and a significant increase in time to peak concentration but had no effect on the AUC. These results suggest that alterations in gastric emptying and gut motility due to ageing or other drugs have no effect on the overall availability of metoprolol to the systemic circulation but may have significant effects on the time to peak plasma concentration and peak concentration achieved after a single oral dose.

Notes: Summary The steady state pharmacokinetics of pirmenol was compared in twelve healthy young (aged 18 to 45 y) and 11 elderly subjects (over 65 y) subjects given pirmenol HCl 100 mg every 12 h for a total of 14 doses. In addition, the single-dose pharmacokinetics of pirmenol was determined following a 100 mg oral dose in the young subject group for comparison with the results of repeated administration. In the young subjects, the mean single-dose and steady-state CLR of pirmenol were similar; however, Ae was 29 % higher and CL/f was 22 % lower at steady state than after the single dose. Steady-state (fourteenth dose) Cmin, Cmax, tmax, λz, Ae, CL/f, CLR and V values were similar in the young and elderly subjects. Based on pharmacokinetic considerations, the dosage of pirmenol is unlikely to differ in young and elderly subjects.

Notes: Summary This study has compared the effect of repeated administration of charcoal and cholestyramine on the elimination of piroxicam. Eight young adults were given piroxicam as a single dose of 20 mg, on 3 separate occasions. On one of the occasions charcoal was also given. On another occasion cholestyramine was also administered. The mean elimination half-life after piroxicam alone was 53.1 h. This was reduced to 40.0 h by charcoal administration and to 29.6 h after administration of cholestyramine. In the second phase of the study 7 elderly subjects received piroxicam 20 mg for 14 days on two occasions. Cholestyramine administration at the end of one of the periods reduced the mean elimination half-life of piroxicam from 52.3 h to 27.3 h.

Description / Table of Contents: Zusammenfassung Anatomie, radiographische Darstellung und pathologische Bedeutung der tentoriellen Arterie, die ein Ast der kavernösen Verlaufsstrecke der A. carotis interna ist, werden besprochen.
Notes: Summary The anatomy, radiographic visualization and pathological significance of the tentorial artery, which is a branch of the cavernous portion of the internal carotid artery, is discussed. Eighteen patients with an enlarged tentorial artery are described. The diagnoses in these patients were almost equally divided among tentorial meningiomas (4), malignant gliomas (6) and arteriovenous malformations (4). In addition there was a single case in which this vessel fed an acoustic neuroma; this relationship has not been previously reported. In three cases the enlarged artery was not recognizably related to the lesion present.

Notes: Neutron diffraction and quasielastic neutron experiments are performed to investigate the effect of HCl adsorption on the structure and dynamics of an ultra-thin ice Ih film (5 H2O bilayers thick) deposited on a crystalline MgO(001) substrate. Three HCl coverages have been studied 0.3, 0.6, and 1 monolayer (ML) in the temperature range 190–270 K. At 0.3 and 0.6 HCl monolayer, no mobility is measured at T≤220 K. A translational mobility, which is a signature of a liquid phase, is observed at T=250 K. This phase occurs 15 K below the surface melting temperature of the bare ice film. The fraction of mobile molecules represents 30% (0.3 ML HCl) and 45% (0.6 ML HCl) of the film. At 1 HCl monolayer and T=220 K, HCl–dihydrate coexists with ice Ih, whereas at T=250 K the ice film becomes amorphous and only 9% of the film is mobile. The results are discussed within the context of atmospheric chemistry. © 2002 American Institute of Physics.
