    Notes: Summary In crosses involving three allelic pab markers of the same cistron, the recombination frequencies between any two of the markers were such, that a consistent order of the three markers could be established. In these crosses two closely linked markers (0,9% distance, on each side) were also present. The segregation of the pab independent types showed, with respect to the outside markers, a frequent appearance of all four types of outside marker combinations, which in the crossing-over theory would belong to one crossover between the pab markers and 0, 1, or 2 crossovers in the adjacent regions. This deviation from ordinary crossingover expectations, called correlation effect, is confined to very small dimensions of the genome. In the present case to regions of about 0,05% map units.-Inspite of this effect, which sometimes nearly equals out the four types of outside marker combinations, the two recombinant types were, in all present crosses, significantly different, such that the pab markers could be ordered with respect to the outside markers. This ordering is in agreement with that established independently from recombination frequencies. Thus the frequencies of the different types of outside marker combinations are in agreement with the assumption, that the linear genes are built lengthwise into the chromosomal thread, and in disagreement with the assumption that genes or even larger chromosomal parts are sidechains of the chromosome. For this work a subdividing technique has been used by which it was relatively easy to obtain the double mutants between close markers. The same technique can also be applied for the partial selection of double mutants of allelic markers, provided that relatively close markers are available. In determining the linkage relationships it has been found, that me-3 seems to involve a fairly large deletion.

    Notes: Abstract To detect possible changes in the regulation of glutamate/γ-aminobutyric acid (GABA) enzymes at the level of gene expression in a thioacetamide-induced rat model of acute hepatic encephalopathy, we have examined changes in the mRNAs of four glutamate/GABA enzymes by quantitative RNA blot hybridization analysis. Such changes could reflect cell adaptation to excess ammonia or some other associated metabolic stress. The mRNA levels of glutamate dehydrogenase (GDH) decreased similarly in three different brain regions, whereas those of glutamine synthetase (GS) and glutaminase (GA) increased. The mRNA levels of glutamate decarboxylase (GAD) were unchanged. The results indicate that some effect of liver damage, presumably hyperammonemia, affected the expression of some, but not all, genes associated with ammonia and glutamate metabolism in the brain. This adaptation of gene expression to secondary effects of ammonia on brain amino acid neurotransmitter metabolism or brain energy metabolism could play a role in the physiological changes observed in hepatic encephalopathy.

    Notes: Summary Sporulation of the standard strain of Bacillus subtilis can be induced by decoyinine (a specific inhibitor of guanosine monophosphate synthesis) in the presence of excess ammonium ions, rapidly utilizable carbohydrates and phosphate. Certain developmental mutants unable to sporulate in usual sporulation media can also be induced to sporulate under these conditions. Among the inducible strains are mutants blocked in the citrate cycle, in phosphoenolpyruvate carboxykinase or one of the glycerolphosphate dehydrogenases (NAD dependent or independent). A mutant lacking phosphoglycerate kinase activity and able to grow on glucose and L-malate but not on either carbon source alone, can also be induced to sporulate in a medium containing both glucose and malate. Among other sporulation mutants whose biochemical deficiency is not known, some are weakly inducible whereas many, blocked at sporulation stages O, II, III, or IV, are not inducible. The results show that the functions of the citrate cycle (used for ATP synthesis) and of gluconeogenesis (used for glycerol phosphate and D-glucosamine-6-phosphate synthesis), normally needed for sporulation under conditions of carbohydrate deprivation, are no longer required when sporulation is induced by decoyinine in the presence of carbohydrates. Mutants in which sporulation is highly inducible can be used as a sensitive tool for the detection of other inducing compounds because their spontaneous (uninduced) background of spores is very low.
