    Notes: Summary The efficacy and toxicity of 120 mg/m2 etoposide and 100 mg/m2 carboplatin given i.v. daily x 3 together with 750 mg/m2 cyclophosphamide and 14 mg/m2 vincristine given i.v. on day 1 (ECCO) in a regimen given every 28 days for 6 courses was assessed in 90 (40 limited stage, 50 extensive stage) previously untreated patients with small-cell lung cancer. Mediastinal irradiation using 50 Gy in 25 fractions was given to limitedstage patients without progression after 3 courses of chemotherapy. Cranial irradiation with 30 Gy in 10 fractions was given to all patients attaining a complete response (CR). Objective responses were seen in 83% [CR, 60%; partial response (PR), 23%] of patients with limited and 76% (CR, 22%; PR, 54%) of those with extensive disease. The median relapse-free survival for objective responders with limited disease was 13.4 months, with a median of 8.0 months for extensive-stage patients. The median relapse-free survival for patients achieving a CR was 13.4 months, with a median of 7.8 months for those undergoing a PR. The median survival was 13.3 months for patients with limited disease, with a median of 9.6 months for those with extensive disease. The median survival following a CR was 18.2 months, with a median survival of 9.9 months for those showing a PR. The combination was well tolerated, with either no nausea or nausea only (WHO grade 0 or 1) in 56% of patients and minimal mucositis, renal toxicity, neurotoxicity or ototoxicity. Neutropenia measuring 〈1.0×109 WBC/l (WHO grade 3 or 4) was seen in 74% of patients, with two deaths due to infection occurring during neutropenia. Thrombocytopenia of 〈50×109 platelets/l (WHO grade 3 or 4) occurred in 24% of patients. ECCO is a new, active, welltolerated program for previously untreated patients with small-cell lung cancer.

    Notes: Summary Trimetrexate (TMTX) is a potent inhibitor of dihydrofolate reductase that circumvents the transport resistance seen with methotrexate and has a wide spectrum of preclinical activity. A total of 18 patients with advanced cancer were treated in a clinical and pharmacological phase I trial with TMTX given as a continuous 5-day intravenous infusion. Neutropenia, thrombocytopenia and stomatitis were the dose-limiting toxicities at the maximum tolerated dose of 50 mg/m2 per 120 h (10 mg/m2 per day for 5 days). There was one septic death associated with neutropenia. Other toxicities were mild rash, mild nausea and transiently raised serum transminase levels. Significant relationships between the dose given and the AUC of plasma TMTX and the steady-state plasma level were established. Significant, although weak, relationships between the percentage of change in neutrophils and platelets and both the AUC and steady-state plasma level of TMTX were also observed. No objective tumour responses were seen, although six patients had stable disease. The recommended phase II dose for a continuous infusion of trimetrexate is 40 mg/m2 per 120 h.

Goldfish acclimated either to 5 °C or to 25 °C were transferred to the opposite temperature and the changes in behavioral resistance to high temperature, the fluidity and fatty acid composition of isolated synaptosomal membranes were followed during acclimation to the new temperature with the purpose of establishing some correlation. 2. In 25 °C-acclimated goldfish, hyperexcitability was induced at 34.5 °C, loss of equilibrium at 37.6 °C and coma at 39.0 °C. In 5 °C-acclimated goldfish the corresponding temperatures were 29.2 °C, 32.0 °C and 33.0 °C. Time to attain 75% of the final acclimated state after transfer was approximately 4 days at 25 °C and 28 days at 5 °C. 3. Fluidity of synaptosomal membranes isolated from goldfish brains was estimated by use of the fluorescence polarization technique. Membrane viscosity decreased during acclimation to 5 °C, but increased during acclimation to 25 °C. The early stages of the transitions differed in time course from behavioral resistance acclimation but times to reach the new acclimated state were similar. 4. Fatty acid composition of synaptosomal phospholipids showed increased unsaturation during cold-acclimation and decreased unsaturation during warm-acclimation. 5. It is concluded that during acclimation, behavior shows changes in resistance to heat which are related to synaptic block. These are correlated in direction and overall time course with viscosity of synaptosomes as dictated by changes in the saturation of membrane phospholipids.

    Notes: Abstract This multicenter study assessed breast cancer screening uptake in 461 unaffected women at increased risk of developing breast cancer on the basis of family history who approached familial cancer clinics for advice about surveillance options. At the time of attending the clinic, 89% and 90% of participants were vigilant with respect to age- and risk-specific recommendations for mammography and clinical breast examination, respectively, and 51% reported practicing breast self-examination monthly or more frequently. The degree to which health outcomes are perceived to be under one's personal control (χ2=−2.09, p=0.0037) and breast cancer anxiety (χ2=8.11,p=0.044) were both associated with monthly or more frequent breast self-examination, while there were no associations with sociodemographic characteristics. A significantly lower percentage (56%) of women aged 〈30 were vigilant with respect to mammography recommendations, compared to 77%, 96% and 98% of women aged 30–39, 40–49 and 〉50, respectively (χ2=37.2,p〈0.0001). These relatively low rates of mammographic screening in young women may reflect concerns about increased cancer risk associated with early and repeated radiation exposure or lack of sensitivity in young women with radiographically dense breasts. If mammographic screening is ultimately shown to lower mortality in women at high risk, there will be a strong case to promote screening in young women. The need for regular mammographic screening would then need to be highlighted and reinforced amongst young women and their referring physicians. Awareness amongst general practitioners, who are largely responsible for referral to screening services, would also need to be increased.

    Notes: Abstract The agarophyte red seaweed Gracilaria conferta was used as a model plant to study the relationship between growth and environmental factors. This species was cultured in small outdoor tanks with continuous seawater supply during three years. Seaweeds were kept under constant density by weekly thinning and were also pulse-fed weekly with nitrogen and phosphate. Water temperature and underwater irradiance increase had opposite effects on the weekly growth rate in two seasons: negative in summer and positive in non-summer. Therefore, a dichotomic separation between summer (June–August) and non-summer (September–May) seasons was utilized in the proposed linear model. The linear model, of the analysis of covariance type, accounted for an explained percentage of total variation (R2) of 0.567, with significant coefficients of all variables included. A standardized model showed that season was the dominant variable, with its coefficient being twice that of temperature in summer, and zero in the non-summer season. Water temperature affected the growth rate twice as much as irradiance, and epiphytes showed a significant negative effect on growth only in the summer. This model aids in the prediction of growth on a seasonal basis under local conditions.

    Notes: Abstract Assimilate translocation has been identified and characterized in Gracilaria cornea under different conditions. Carbon fixation and translocation were carried out by inserting the base part of the thallus into a bicarbonate labeled solution in seawater and exposing its upper part to the air (open system) or to a non-labeled solution above a rubber septum (closed system). After a pulse-chase treatment in the light, three separate sections of each thallus were extracted by DMF (high moleuclar weight photosynthates) or by ethanol (low molecular weight). The results indicate a high rate of active photosynthate translocation which is directly related to inorganic carbon gradients in the thallus, and probably also to sugar gradients in the thallus. Translocation parameters of Gracilaria cornea are lower than of brown algae, as Gracilaria does not contain specific translocation tissues.

    Notes: Abstract A strain selection procedure using Gracilaria verrucosa gametophytic sporelings was found to be an efficient tool for the improvement of Gracilaria strains. Two strains, C-2 and A-18, which were isolated and grown clonally, showed higher growth rates under high and low temperature conditions, respectively, than the local Gracilaria conferta. Growth rate, photosynthesis and chlorophyll, which were measured under different temperature and photon flux densities, demonstrated an overall advantage of the selected strains over the wild type strains of both G. verrucosa and G. conferta. Growth rates were also generally in positive correlation with the carboxylase activity of Rubisco. The G. verrucosa wild type also had a 40% higher agar content than G. conferta. The selected strains thus showed higher potential for outdoor cultivation than local wild type populations.

    Notes: Abstract Photosynthetic (oxygen evolution) and growth (biomass increase) responses to ambient pH and inorganic carbon (Ci) supply were determined for Porphyralinearis grown in 0.5 L glass cylinders in the laboratory, or in 40 L fibreglass outdoor tanks with running seawater. While net photosynthetic rates were uniform at pH 6.0–8.0, dropping only at pH 8.7, growth rates were significantly affected by pH levels other than that of seawater (c. pH 8.3). In glass cylinders, weekly growth rates averaged 76% at external pH 8.0, 13% at pH 8.7 and 26% at pH 7.0. Photosynthetic O2 evolution on a daily basis(i.e. total O2 evolved during day time less total O2 consumed during night time) was similar to the growth responses at all experimental pH levels, apparently due to high dark respiration rates measured at acidic pH. Weekly growth rates averaged 53% in algae grown in fibreglass tanks aerated with regular air (360 mg L-1 CO2) and 28% in algae grown in tanks aerated with CO2-enriched air (750 mg L-1 CO2). The pH of the seawater medium in which P. linear is was grown increased slightly during the day and only rarely reached 9.0. The pH at the boundary layer of algae submerged in seawater increased in response to light reaching, about pH 8.9 within minutes, or remained unchanged for algae submerged in a CO2-free artificial sea water medium. Photosynthesis of P. linearissaturated at Ci concentrations of seawater (K0.5560 μM at pH 8.2) and showed low photosynthetic affinity for CO2(K0.5 61 μM) at pH 6.0. It is therefore concluded that P. linearisuses primarily CO2 with HCO3 - being an alternative source of Ci for photosynthesis. Its fast growth could be related to the enzyme carbonic anhydrase whose activity was detected intra- and extracellularly.
