    Notes: Preparative isoelectric focusing was employed to compare the association of protochlorophyllide and chlorophyllide with the enzyme NADPH-protochlorophyllide oxidoreductase (PCR; EC 1.3.1.33). Photoactive protochlorophyllide-PCR complexes were solubilized with 1-O-n-octyl-β-d-glucopyranoside from non-irradiated prolamellar bodies of wheat (Triticum aestivum). Also, chlorophyllide-PCR complexes were solubilized from prolamellar bodies irradiated under conditions either preventing or favouring a spectral shift of chlorophyllide to shorter wavelengths. Independently of the treatment prior to the solubilization, the pigments and the PCR focused together at pHs of 4 to 5. The results indicate that protochlorophyllide-PCR complexes are conformationally similar to chlorophyllide-PCR complexes. The results support the hypothesis that the spectral shift, referred to as the Shibata shift, reflects a breaking-up of large chlorophyllide-PCR aggregates to smaller chlorophyllide-PCR units, rather than a dissociation of the chlorophyllide from the enzyme protein.

    Notes: 3-Amino- 1,2,4-triazole (amitrole) provided to germinating barley at 20°C in the light led to bleached seedling leaves and photodynamic destruction of chloroplast structure, whereas normal greening and chloroplast ultrastructure was obtained when the seedlings developed in the presence of amitrole in the light at 30°C. Mass spectrometric analysis of the extractable herbicide demonstrated the same content of amitrole in leaves developed at 20 and 30°C. A very similar temperature-sensitive syndrome is characteristic for the nuclear gene mutant ligrina-o34 in barley. Amitrole and the mutation were shown to inhibit the cyclization of lycopene, leading to severe deficiencies in β-carotene and its xanthophyll derivative lutein. Besides accumulation of lycopene, also its precursors phytoene, phytofluene and ξ-carotene accumulated. Inhibition of carotenoid biosynthesis by amitrole and the mutation at 20°C in the light led to a strong reduction of both transcript and protein levels for chloroplastic glutamine synthetase (GS2) while transcript amount and protein of the cytosolic isoenzyme (GS1) were unaffected. At 30°C increased levels of mRNA for the chloroplastic isoform GS2 were observed in wild type, mutant and amitrole-treated seedlings, but protein levels remained unchanged. Turnover rates of the GS2 protein were the same at 20 and 30°C. This extensive translational control of chloroplastic GS2 synthesis was also observed in a heat shock experiment, which revealed transiently increased mRNA levels for chloroplastic GS2 but unchanged protein levels.Permissive synthesis of β-carotene and chloroplastic glutamine synthetase (GS2) at 30°C in the presence of amitrole or the tigrina-o34 mutation might be due to two alternative pathways of ionone ring formation using either lycopene or neurosporene as substrates for cyclization.

    Notes: The composition and amount of carotenoid pigments were determined in etiolated seedling leaves of 6 barley (Hordeum vulgare L.) mutants, comprising 1 xantha and 5 tigrina mutants. All mutants had on a mole basis approximately the same content of carotenoids as the wild type.The mutants xan-u21, tig-n32, and tig-33 contained significantly higher amounts of carotenes than the wild type, ranging from 32 to 68% of the total carotenoid content as compared to the 4–8% found in the wild type. In the mutants tig-b23 and tig-o34, only a slight increase in the amount of carotenes was notable. The carotene content and composition in tig-d12 was indistinguishable from that of the wild type.The carotenes extracted from xan-u21, tig-b23, tig-n32, tig-33, and tig-o34 were characterized by adsorption chromatography and spectrophotometry. Mutant xan-u21 is in the dark blocked in β-carotene synthesis, and accumulates the aliphatic polyenes: phytofluene, proneurosporene, poly-cis-lycopenes, neo-lycopene and lycopene. The other four mutants synthesize β-carotene, but accumulate in addition various higher saturated carotenes, the main components being ζ-carotene in tig-b23, a lycopenic pigment in tig-n32 and tig-33, and lycopene in tig-o34.Accumulation of higher saturated carotenes appears correlated with specific aberrations of the membrane structure in plastids. The regulation of carotene and protochlorophyllide syntheses in etioplasts are closely linked as shown by the single gene mutants which affect both pathways. However, several mutants have been identified which cause defects in protochlorophyllide synthesis only.

    Notes: Abstract Two chloroplast tRNAAsp species from barley were purified by chromatography on benzoylated DEAE-cellulose and sequenced. They differ in the modification at position 34, where queuosine (Q) is present in one of the species. The same chromatographic procedure yielded only one tRNAGlu species, corroborating the assumption that the same tRNAGlu species participates in both protein and chlorophyll biosynthesis. The level of tRNAGlu remains unchanged after light treatment of etiolated seedlings, whereas the amount of tRNAAsp decreases to about 50% relative to the level of dark-grown plants.
