    Notes: Abstract A population of cells in the anterior substantia nigra pars compacta (SNPc) of the guinea-pig have been reported previously that differ from classical dopaminergic neurons in terms of their active and passive membrane properties. To investigate this population further, anterior nigral neurons (n=17) were compared with neurons in the adjacent subthalamic nucleus (STN; n=26). The anterior nigral neurons were found to be indistinguishable from STN neurons in their action potential characteristics, firing rate, resting membrane potential and input resistance. A low-threshold calcium conductance and anomalous rectification could be demonstated in cells from both groups. Furthermore, the gross morphological characteristics of anterior nigral neurons and STN neurons were very similar, as assessed following the intracellular injection of biocytin. A further similarity was seen in the response of the two cell groups to cyanide (200 μM) and apomorphine (500 μM). Cyanide hyperpolarised the membrane potential of all STN neurons and the majority (77.8%) of anterior nigral neurons, in both cases producing a concomitant reduction in firing rate. These changes were accompanied by an increase in membrane conductance for potassium ions. Apomorphine depolarised the membrane potential of all STN neurons and anterior nigral neurons, in most cases increasing the input resistance (83.3% of STN neurons and 100% of anterior nigral neurons). In both groups of cells, when firing rate was affected, an increase was usually seen. Given the physiological, morphological and pharmacological similarities of STN and anterior nigral neurons, the most parsimonious interpretation is that the anterior nigral neurons belong to the STN. However, the anterior nigral neurons were found in slices that, when resectioned, contained tyrosine hydroxylase (TH)-immunoreactive cell bodies in every section, in a location corresponding to the SNPc. The implication is that in the guinea pig the SNPc and STN (usually considered to be anatomically distinct nuclei) intermix at this level for several hundred microns. This close association of the STN and the compacta was further demonstrated by the presence of TH-positive varicose and non-varicose neuronal processes within the STN.

    Notes: Summary Acetylcholinesterase is released from substantia nigra neurons, independently of cholinergic transmission. In an attempt to discover the functional significance of this phenomenon, the behavioural effects of injecting acetylcholinesterase into one substantia nigra of the rat were investigated. Following a single injection of the enzyme, intraperitoneal amphetamine evoked circling behaviour in a direction away from the side of injection. Purified acetylcholinesterase with a similar electrophoretic mobility to the endogenous secreted form, was far more potent in eliciting circling than much higher activities of commercial enzyme, consisting of several molecular species of acetylcholinesterase. Similar infusions of butyrylcholinesterase did not induce circling. Depending upon the amount of enzyme initially given, the behavioural effects of a single injection of acetylcholinesterase persisted for up to thirty days. During this period apomorphine, administered systemically, induced transient circling towards the acetylcholinesterase-treated side. It is concluded that secreted acetylcholinesterase has a functional significance within the substantia nigra, independent of cholinergic transmission. This released enzyme could exert long-term changes in the activity of the nigrostriatal system, involving modification of dopamine striatal receptors.

    Notes: Summary It is well established that neuroleptics increase the firing rate of nigrostriatal neurons. However, the action of these drugs on firing pattern has received scant attention. The effect of local administration of neuroleptic agents was investigated on the firing pattern, and also firing rate, of nigro-striatal neurons. Chlorpromazine or haloperidol was microinjected into the substantia nigra of the urethane-anaesthetized guinea pig during extracellular recordings from identified nigrostriatal cells. Both neuroleptics induced the familiar increase in firing rate of nigrostriatal neurons. More significantly, however, these drugs also caused a dose-dependent change in firing pattern, specific to neuroleptic action. This change consisted of a drug-induced transition from slow irregular single or double spikes with a short interspike interval (‘beat’ firing) to irregular bursts of spikes with a longer interspike interval (‘burst’ firing). Furthermore, the transition from beat to burst firing occurred sharply and sometimes continued to switch in a regular cyclical fashion between the two modes for up to three hours following infusion of the neuroleptic. Administration of neuroleptics may thus provide a useful tool for studying the physiology of the firing pattern of nigrostriatal neurons.

    Notes: Summary Acetylcholinesterase is released within the substantia nigra from the soma/dendrites of nigrostriatal neurons. Previous work suggests that this phenomenon is independent of cholinergic systems, but rather serves to modulate the sensitivity of dopamine-containing nigrostriatal cells to synaptic events. This hypothesis was tested directly in the anaesthetized guinea-pig. Micro-infusion of acetylcholinesterase into the substantia nigra led to an increase in spontaneous firing of nigrostriatal neurons. Furthermore, the pattern of firing evoked by stimulation of the striatum was markedly enhanced. By contrast, administration of butyrylcholinesterase had no effect. It thus appears that acetylcholinesterase has a modulatory action on the firing of nigrostriatal cells, independent of hydrolysis of acetylcholine.

    Notes: Abstract The substantia nigra has one of the highest levels of ATP-sensitive K+ channel in the brain. Since this channel is controlled by cell metabolism, the aim of this study was to see how closely it is associated with nigral dopamine systems, which are decreased in Parkinson's disease. In a sub-population of neurons within the rostral substantia nigra pars compacta of the guinea-pig, a brief period of hypoxia resulted in a tolbutamide (100–500 μM) sensitive hyperpolarisation [input resistance (IR) decrease from 144.88±14.04 Mω pre-hypoxia to 105.91±13.25 Mω during hypoxia]. Maximal blockade of this decrease was seen in presence of 500 μm tolbutamide [IR decrease only from 161.35±32.82 Mω to 155.02±34.29 Mω]. Reserpine (which depletes dopamine stores) but not α-methyl-para-tyrosine (which decreases de novo synthesis of dopamine) caused a marked attenuation of this hyperpolarisation [IR decrease only from 163.32±44.42 Mω pre-hypoxia to 154.42±50.97 Mω during hypoxia]. This observation suggests that blockade of dopamine storage, but not of de novo synthesis, leads to a loss of responsiveness of certain mid-brain neurons to hypoxia, rendering them potentially more susceptible to subsequent degeneration. The possible link between nigral dopamine systems and ATP-sensitive K+ channels is discussed.

    Notes: The isotropic phase of nematogenic substances above the nematic–isotropic phase transition temperature Tni is characterized by short range orientational correlation of the molecules and the existence of pseudonematic domains. This microscopic structure has a strong influence on reorientational dynamics in the liquid. We present time resolved transient grating optical Kerr effect measurements of pentylcyanobiphenyl between 35 and 120 °C, which allow us to monitor dynamics on timescales from subpicoseconds to tens of nanoseconds. The well-known collective component characterizing the reorientational dynamics of the pseudonematic domains follows Landau–deGennes behavior up to 30–35 degrees above Tni where the theoretical correlation length ξ≈three molecular lengths. Additionally fast relaxation channels (with time constants between 1 and 300 ps) are observed which are associated with local reorientational and intermolecular dynamics within the pseudonematic domains. The local dynamics are temperature independent in the range where the pseudonematic domains dominate the liquid structure, demonstrating a decoupling of the molecular motions from the bulk viscosity. Only at high temperatures, T(approximately-greater-than)70 °C, where the pseudonematic domains cease to exist, is a viscosity dependence observed. At these elevated temperatures, the slowest contribution to the fast molecular dynamics approximately follows hydrodynamic theory.

    Notes: Subpicosecond transient grating optical Kerr effect measurements have been used to evaluate the reorientation of biphenyl molecules in neat biphenyl and n-heptane solutions. Besides an ultrafast (100 fs time scale) component associated with librational damping/dephasing, two reorientational relaxation components are observed. The slow reorientation is due to rotation around the short axes of the molecule (tumbling motion), the fast reorientation is associated with internal rotation around the central C–C bond and/or rotation of the whole molecule around its long axis (spinning motion). Whereas the tumbling motion has been observed in earlier depolarized light scattering data, the time resolved Kerr data presented here are the first ones to reveal the dynamics of the fast reorientation component and the ultrafast librational dynamics. It is shown that the diffusive reorientational relaxation must be coupled to the ultrafast librational dynamics, and implications of this coupling are pointed out.
