    Notes: Summary Milbemycin production byStreptomyces hygroscopicus RB4569D was examined in media containing different carbohydrates. Total milbemycin titer could be increased by substitution of fructose for glucose and by selection of the appropriate starch type. Total titer could be further enhanced by increasing the concentration of fructose and/or starch in the medium. Rates of carbohydrate utilization were shown to be independent of their initial concentration and increased titers in high carbohydrate media were shown to be due to a prolonged production phase rather than an increased accretion rate. The pattern of individual milbemycin components was governed by the carbon:nitrogen ratio of the medium rather than carbohydrate concentration and there was a critical C:N ratio below which no milbemycin was produced.

    Notes: Abstract Nasopharyngeal aspirates and throat swab specimens were compared in a polymerase chain reaction (PCR)-based test forMycoplasma pneumoniae. The pathogen was detected in 50 % and 45 % of throat swab specimens and aspirates, respectively. However, in specimens negative forMycoplasma pneumoniae by PCR, amplification inhibitors were detected in 0 % and 36 % of throat specimens and aspirates, respectively. Further investigations confirmed that no throat specimens, but one-quarter of aspirates, are likely to be rejected for containing inadequate respiratory material or excess amplification inhibitors. Because rejection of most of the unsuitable specimens is possible only after PCR, the use of aspirates is less cost-effective. This, and the reluctance to subject patients to aspiration, make the aspirate an inferior specimen for detection ofMycoplasma pneumoniae by PCR.

    Notes: Abstract The stability and mechanical properties of polypyrrolep-toluene sulphonate films prepared under various preparation conditions were studied and are reported here. Relatively high retention of conductivity and flexibility properties in air at room temperature were found for the films. More stable films were produced from propylene carbonate diluent than from acetonitrile and solvent mixtures. Acid treatment caused conductivity to increase and the mechanical properties to decrease slightly. However, both properties decreased dramatically after exposure of the films to sodium hydroxide solution. The effect of solutions containing different vanadium ions was also observed, the films being stable in V3+ and V4+ acid solutions, but unstable in V5+ solutions. The thermal stability of polypyrrole films grown at different temperatures and from different solvents was considerably high and fairly constant. Anisotropy in the mechanical properties was also observed for the two directions (along and across) within the same plane of the film. Incorporation of the plasticizer, di-iso-butyl phthalate (DIBP) into the films improved the mechanical properties. A high extendable acrylic-polypyrrole composite film was also prepared, capable of straining over 100%.

    Notes: Abstract Purpose: To evaluate the efficacy of a novel multiday schedule of vinorelbine and cisplatin in patients with advanced NSCLC. Patients and methods: Thirty patients were enrolled, including 27 patients with stage IV disease, and 11 patients with performance status of 2. They received a maximum of four chemotherapy cycles with cisplatin 20 mg/m2/day and vinorelbine 15 mg/m2/day intravenously (i.v.) for four consecutive days, every three weeks, with prophylactic filgrastim. Results: Sixteen patients responded (53%, 95% confidence interval (95% CI): 34%–72%), including two complete and fourteen partial confirmed responses. Median survival for all patients was 8.1 months, with actuarial one-year and two-year survival rates of 40% and 15%. Despite prophylactic filgrastim, the delivered vinorelbine dose intensity of 16.8 mg/m2/week caused febrile neutropenia in 48% of patients (16% of cycles), resulting in one treatment-related death. Common nonhematologic toxicities included delayed emesis, asthenia, and constipation. Conclusions: This multiday vinorelbine–cisplatin schedule is highly active against advanced NSCLC but results in frequent neutropenic complications. The myelotoxicity and antitumor efficacy of vinorelbine in NSCLC patients may be schedule-dependent.

    Notes: A technique is described for the dissociation and fractionation by isopycnic centrifugation (4,000 × g, 60 minutes, 4°C) in linear bovine albumin density gradients (12 ml, pH 5.2 real osmolality 333 mmol/Kg water, 1.030-1.075 g/cm3) of cells (≤ 3 × 107/gradient) released by a strictly standardised combination of mechanical and enzymatic means from a transplantable methylcholanthrene induced BALB/c fibrosarcoma. Optimal conditions for reproducible localisation of cell bands with maintenance of both satisfactory resolution and satisfactory viable cell recovery (〉 80%) were established by means of a series of simultaneous double fractionation experiments. When rebanding was performed under these conditions the density of the median of the cell count of the refractioned cells shifted less than 0.0005 g/cm3. Experiments also showed that close adherence to certain aspects of the tissue dissociation and fractionation protocol was necessary to avoid reduced cell yields and viabilities, density-dependent selective cell losses, reductions in resolution and shifts in the location of cell bands. Other aspects of the protocol were tolerant to variation without introducing artefacts.

    Notes: A transplantable methylcholanthrene-induced fibrosarcoma of female BALB/c mice (the MC-2 fibrosarcoma) was dissociated by combined mechanical and enzymatic means, then fractionated by isopycnic centrifugation in linear albumin gradients.In some experiments recovered cells were both cultured in soft nutrient agar and inoculated subcutaneously into syngeneic recipients. In these experiments a highly significant correlation was observed between subsequent colony number and rapid growth phase tumor size suggesting identity of clonigenic and tumorigenic cells.It was consistently found that clonigenic cells were markedly depleted from the low density extremes of the cell density distribution profiles suggesting that the low density neoplastic cells had irreversibly left the growth fraction.With increasing tumor age, sequential studies showed that both total and clonigenic cell density distribution profiles were variable, showing no obvious trend, suggesting that in the age (13-35 days) and size (2-8 g) range studied growth fraction changes had little selective effect on cells of any specific density. These results imply that a marked selective depletion of low density clonigenic cells (or selective accumulation of low density non-proliferative cells) must mainly occur during an earlier phase of tumor growth. Studies on several other murine solid tumors also showed maximal depletion of clonigenic cells from the least dense fractions, suggesting that this situation may be common.
