Notes: Summary An enzyme histochemical study of experimental herpes simplex encephalitis of the mouse has revealed a decrease in the number of capillaries displaying alkaline phosphatase activity. Glial cells showed increased Inosine 5 diphosphatase and ATPase activity. These enzyme histochemical changes were distributed throughout the nervous parenchyma while the lesions, seen by light microscopy, are localized to well defined areas. Mice inoculated with a pure culture of irradiated HSV failed to show the above mentioned modifications.

Notes: Summary The lectinRicinus communis agglutinin I (RCAI) was topically applied to transected mouse sciatic nerve or to neuromas formed 2 months after a nerve transection. Fluorochrome-labelled ricin was transferred to the corresponding dorsal root ganglia where it accumulated selectively in the nerve cells, apparently as a consequence of retrograde axonal transport. The ricin caused an almost total loss of the dorsal root ganglionicneurons and, consequently, could prevent formation of neuromas or eliminate an already existing neuroma. The hybrid toxin wheat germ agglutinin (WGA)-ricin-A chain caused no apparent increased sensitivity for neuronal destruction. The drugs doxorubicin and ethidium bromide, similarly applied, labelled satellite and other cells in addition to neurons in the ganglia, and caused only a moderate neuronal loss. The presented method to eliminate neuromas by selectively destroying sensory neurons may provide a means to study pain mechanisms in neuromas.

Notes: Summary Following intranasal instillation of Sendai virus in newborn mice an extensive virus infection of respiratory epithelium and olfactory mucosa was observed by immunoperoxidase technique. Viral antigen appeared in olfactory nerves and in neurons of the trigeminal ganglia. Selective labeling of neurons in trigeminal ganglia was also seen after virus injection into the snout. This shows that respiratory infections may not be restricted to the respiratory mucosa but may also spread to peripheral ganglia after uptake of virus at axonal terminals and somatopetal axonal transfer to the nerve cell bodies. Following intracerebral injection into newborn mice viral antigen persisted in scattered neurons in the thalamus and mesencephalon after 24 weeks. The majority of the mice developed hydrocephalus, for which obliteration of the aqueduct seems to have been a major cause.

Notes: Summary The involvement of visceral organs was studied in three typical clinical and pathological cases of juvenile amaurotic idiocy (Batten-Spielmeyer-Vogt disease). Abundant intracellular deposits of a substance showing the same staining reactions and primary yellow fluorescence as that previously described in the nerve cells was found particularly in the myocardium, liver, kidneys, spleen and lymphnodes. The resemblance between this substance and lipofuscin and its different chemical composition as compared with the lipid accumulated in infantile amaurotic idiocy (Tay-Sachs disease) is briefly discussed.

Notes: Summary Uptake of horseradish peroxidase (HRP) in sensory nerve terminals in sinus hair in the nose of the mice was studied after local (s.c.) or i.v. injection. HRP spread into the sinus hair follicles and surrounded the nerve terminals of the Merkel disc endings and lanceolate terminals. It was incorporated into vacuoles and vesicles of these terminals. Subsequently, HRP was demonstrated in nerve cell bodies in the ophthalmomaxillary part of the trigeminal ganglion, indicating a somatopetal axonal transport of the tracer. In this way the sensory neurons may be subjected to various influences from the periphery

Notes: Summary The two vestibular cortical projection areas in the anterior suprasylvian sulcus and post-cruciate dimple regions were defined by evoked potential technique in anaesthetized cats. The thalamic location of neurons with axon terminals in these fields was determined using the method of retrograde axonal transport of horseradish peroxidase. The ascending vestibular pathway appeared to be separated also at the thalamic level, where cells in the ventro-posterolateral nucleus were found to project to the post cruciate dimple and cells in the posterior nuclear group to the anterior suprasylvian vestibular cortical fields.

Notes: Summary The total activities of lactate dehydrogenase (LD) and the LD isoenzyme distribution were determined on homogenates from 61 benign and malignant intracranial tumours, and in serum and cerebrospinal fluid from 45 and 28 of the tumour patients respectively. The LD activity and the LD isoenzyme distribution in serum were not changed in patients with intracranial tumours. The LD activity in the cerebrospinal fluid was raised in about 50% of the tumour patients, but without any significant differences between patients with benign and malignant tumours. Except for very low activities in acoustic schwannomas and in pituitary adenomas, the tumour tissue did not differ significantly from normal brain in total LD activity. The LD isoenzyme distribution showed a pronounced cathodal shift in the grade 3–4 astrocytomas and the metastatic carcinomas, but an anodal shift in the grade 1–2 astrocytomas and the oligodendrogliomas. The meningiomas and the acoustic schwannomas showed a midzone isoenzyme pattern with a dominant LD3 fraction. Knowledge of the LD isoenzyme distribution can thus sometimes be a complement to the histological examination in the classification of brain tumours.
