    Notes: Abstract The yeast SIN3 gene functions as a transcriptional repressor, despite the fact that Sin3p does not bind DNA directly. We have conducted a two-hybrid screen to look for proteins that interact with Sin3p, using the PAH2 domain of Sin3p as bait. Five new genes, STB1–STB5 were identified, as well as the STB6 gene, which is similar to STB2. STB1, STB2, STB3, and STB6 are novel genes, and STB4 and STB5 encode C6 zinc cluster DNA-binding proteins. None of these genes is essential for viability, and several of these genes may encode transcriptional activators. Several special problems were encountered in using a transcriptional repressor in a two-hybrid screen. For example, the STB genes will interact with a LexA-Sin3(PAH2) fusion protein containing a region of Sin3p, but a LexA-Sin3p fusion protein containing full-length Sin3p, along with a STB clone, does not produce two-hybrid activation of a transcriptional reporter. In addition, a sin3 mutation reduces the transcriptional activation by two-hybrid partners, suggesting that a sin3 mutation reduces the transcriptional efficiency of the Gal4p and VP16 activation domains. We have shown previously that Sin3p is part of a large multiprotein complex, and we show here that Stb1p and Stb2p are present in this complex.

    Notes: Summary During a six-year period five patients withCampylobacter fetus subspecies fetus infections were seen at the Mayo Clinic in Rochester, Minnesota. Bacteremia was observed in two patients, one presenting with aortic valve endocarditis and the other with abdominal atherosclerotic aortic aneurysm.C. fetus subsp. fetus was isolated from tibial tissue of a patient with osteomyelitis. Diarrhea was the main complaint of two further patients, and was also mentioned by the patient with the aortic aneurysm. Despite the use of incubation conditions and selective media geared to detect onlyCampylobacter jejuni, C. fetus subsp. fetus was isolated from stool specimens of the two patients with gastrointestinal symptoms. The fact that three of fiveC. fetus subsp. fetus infections observed in this study were associated with intestinal symptoms further supports the importance of the gastrointestinal tract in the pathogenesis ofC. fetus subsp. fetus infections.

    Description / Table of Contents: Résumé Listeria monocytogenes est de plus en plus souvent impliqué dans des infections sur prothèses articulaires. Ces infections ont tendance à être plus insidieuses et moins douloureuses que celles à Staphylococcus aureus. Elles peuvent survenir chez des patients immunodéprimés en raison de cancers ou d'autres maladies, ou chez des sujets âgés non immunodéprimés. Un traitement anti-microbien prolongé, autre que par céphalosporines, est généralement indiqué dans les infections sur prothèses. Sur les vingt-sept cas de listériose recensés pendant la période d'investigation, trois (11%) étaient associés à la présence de corps étrangers (prothèse de hanche, prothèse du genou et greffe artérielle).
    Notes: Summary Listeria monocytogenes is increasingly recognised as a cause of prosthetic joint infection. These infections tend to be more insidious and indolent in contrast to similar infections with Staphylococcus aureus. They can occur in patients who are immunocompromised due to malignancy or other illness or in nonimmunocompromised elderly patients. Listeria infections should not be treated with cephalosporins and prolonged antimicrobial treatment is generally advised for prosthetic joint infections. We noted a high affinity of L. monocytogenes for foreign bodies. Three of twenty-seven cases (11%) of listeriosis under the period of investigation were associated with foreign bodies (hip prosthesis, knee prosthesis and arterial graft).

    Notes: Summary Synovial fluids of patients suffering from rheumatoid arthritis contain elevated levels of granulocyte (PMN) elastase in complex with alpha 1-proteinase inhibitor (alpha 1-PI), whereas free-elastase activity is usually not detectable. This absence of free enzymatic activity in joint effusions has cast some doubt on the pathophysiological relevance of PMN elastase in inflammatory joint destruction. Our in vitro experiments using bovine nasal cartilage demonstrate that incubation with elastase and alpha 1-PI in equimolar concentrations to or even in excess of the serum proteinase inhibitor resulted in significant tissue destruction as assessed by histological staining for proteoglycans, release of uronic acid from the matrix and loss of mechanical stability. Though in the supernatants containing alpha 1-PI, free-elastase activity was not detectable, immunofluorescent staining for elastase evidenced penetration of the enzyme into the matrix. Simultaneous measurements of the incubation media employing a sandwich enzyme-linked immunoadsorption assay (ELISA) revealed PMN elastase in complex with alpha 1-PI but without correlation to the parameters of tissue degradation. In comparison with the results obtained using the chromogenic substrate Suc-Ala-Ala-Ala-pNA (SAPA) for titration of alpha 1-PI against elastase, the employment of cartilage matrix showed that a fourfold increase in inhibitor concentration was necessary to achieve 100% enzyme inhibition. Hence, cartilage surface obviously interferes with the interaction between alpha 1-PI and elastase. Measurements of elastase-inhibitor concentrations or free enzymatic activity in synovial fluid seem to have limited value in predicting cartilage destruction.

Das aerobe System von Hoffman-La Roche (Roche Septi-Chek) erwies sich im Hinblick auf den Nachweis vonCampylobacter-Bakteriämien sowohl der anaeroben Tryptic Soy Broth von Difco als auch dem Isolatorsystem von Du Pont mit Bebrütung der Platten in 5% CO2 als überlegen.
    Notes: Summary The efficiency of three commercially available blood culture systems for isolatingCampylobacter sp. was investigated. Thirteen of 189,688 blood cultures from patients seen at the Mayo Clinic from 1984 through 1990 were positive forCampylobacter sp. Eleven out of 13 blood cultures positive forCampylobacter sp. were part of a complete three bottle set of cultures (Isolator from Du Pont, Roche Septi-Chek from Hoffman-La Roche, and non-vented Tryptic Soy Broth from Difco), none of them providing a specific microaerophilic mileu. The aerobic Roche Septi-Chek seems to be more efficient for the detection ofCampylobacter sp. in blood cultures than either the anaerobic Tryptic Soy Broth (Difco) or the Isolator (Du Pont) using the incubator condition of 5% CO2.
