    Notes: Summary Changes in intracellular Ca2+ release in the diaphragm muscle of alloxan-diabetic mice were compared with changes in normal muscles and non-diabetic denervated muscles. We measured Ca2+ transient aequorin luminescence by direct electrical stimulation of these muscles. External Ca2+-free solution readily decreased the Ca2+ transient in normal muscles but had less of an effect in diabetic muscles. Only when the muscles were pre-injected with EGTA (reducing intracellular levels of free Ca2+) did the Ca2+ transients decrease significantly in diabetic muscles, however, there was no effect in denervated muscles. The caffeine-induced increase in Ca2+ transients, however, was delayed in both diabetic muscles and non-diabetic denervated muscles. The caffeine response was observed in normal muscles under the external Ca2+-free conditions even after EGTA-pretreatment, whereas it was suppressed, after a brief increase, in both diabetic and non-diabetic denervated muscles. These results demonstrate (1) the insensitivity of intracellular Ca2+ mobilization to external Ca2+ levels and the ready accumulation of intracellular Ca2+ in the cytosol in the diabetic state, (2) increased permeability to Ca2+ in the denervated state and (3) impairment of the Ca2+ pool which responds to caffeine in both diabetes and the non-diabetic denervated state. Diabetic neuromyopathy thus appears to be a state of abnormal Ca2+-mobilization caused secondarily by high levels of blood glucose.

    Description / Table of Contents: Zusammenfassung Mit der Doppeldiffusion und der Immunoelektrophorese mit der Zellulose-Acetat-Membran wurde die Kompliziertheit der Hb-A, Hb-F-Antigene und des Anti-Hb-A-Antiserums nachgewiesen, obwohl im anti-Hb-F-Antiserum nur ein Antikörper gegen Hb-F enthalten war. Die Antigenität der Hämoglobin-Ketten wurde mit der Harnstoff-Immunoelektrophorese untersucht. Die elektrophoretische Trennung erfolgte auf einer Zellulose-Acetat-Membran unter Einfluß von 8 m Harnstoff und Hinzufügen der Antiseren nach der Entfernung des Harnstoffs. Trotz der Kompliziertheit des benutzten Systems wurden 2 Linien, eine für die schnell wandernde α-Kette-und eine für die langsamere β-Kette, im Anti-Hb-A-Antiserum und Hb-A-System nachgewiesen. Die Linie der α-Kette im Anti-Hb-A-Antiserum und Hb-F-System und die Linie der γ-Kette im Anti-Hb-F-Antiserum und Hb-F-System wurden auch demonstriert.
    Notes: Summary By double diffusion and cellulose acetate membrane immunoelectrophoresis the complexity of the Hb-A, Hb-F antigens and anti-Hb-A antiserum was demonstrated though the anti-Hb-F antiserum contained only 1 antibody against Hb-F. The urea added immunoelectrophoresis which consists of the electrophoretical separation of subunit chains on the cellulose acetate membrane under the existence of 8 m urea and the application of antiserum after the removal of urea was used to investigate the antigenicity of hemoglobin subunit chains. In spite of the complexity of system used, 2 lines, for the fast moving α-chain and for the slow moving β-chain, were demonstrated in anti-Hb-A antiserum and Hb-A system. Also the α-chain line in anti-Hb-A antiserum and Hb-F system and the γ-chain line in anti-Hb-F antiserum and Hb-F system were demonstrated.

    Description / Table of Contents: Summary Runt disease which occurred after transfusion of C57BI bone marrow into supralethally (900r) irradiated A/Jax mice was delayed, in some cases suppressed by the application of rabbit anti-mouselymphocytic serum. Without antiserum survival time in this system was 15,6 days. A 6 days' treatment with antiserum of the donor mice increased the survival time to 27,8 days. Survival time after an additional treatment with antiserum from the 7th day after irradiation on for 3 weeks was 34,8 days. The heterologous bone marrow transfusion from rat into mouse was also favourably influenced by antilymphocytic serum.
    Notes: Zusammenfassung Das Sckundärsyndrom, welches nach Transfusion von Knochenmark von C57BI Mäusen auf supraletal (900r) bestrahlte A/Jax Mäuse auftrat, wurde durch Kaninchen-anti-Mäuselymphozytenserum verzögert, in einigen Fällen unterdrückt. Während die Überlebenszeit nach homologer Knochenmarkstransfusion in diesem System bei 15,6 Tagen lag, überlebten A/Jax Mäuse mit Knochenmark von C57BI Mäusen, welche 6 Tage lang Anti-Lymphozytenserum erhalten hatten, durchschnittlich 27,8 Tage. A/Jax Mäuse, die zusätzlich noch 3 Wochen lang mit Anti-Lymphozytenserum vom 6. Tage post irradiationem an behandelt wurden, überlebten durchschnittlich 34,8 Tage. Auch die heterologe Knochenmarkstransfusion von Ratte auf Maus wurde durch Anti-Lymphozytenserum günstig beeinflußt.

    Description / Table of Contents: Zusammenfassung Die Zusammenhänge der Antigenität zwischen nativen Hämoglobinen und deren Unterketten wurden mit der Absorption der Antiseren und der Kombination der Harnstoff-Immunelektrophorese und Doppeldiffusion untersucht. Die α-Kette zeigte Identität mit Hb-F, aber nur partielle Identität mit der β-Kette und Hb-A. Die β-Kette war in ihrer Antigenität mit Hb-A identisch, die α-Kette und Hb-F waren teilweise identisch mit der β-Kette. Die ψ-Kette zeigte die Identität mit Hb-F; es wird angenommen, daß ihre Antigenität verschieden von der α-oder β-Ketten ist. Für das Auftreten der Linien der α-, β- und ψ-Ketten müssen Anti-Hb-A-oder Anti-Hb-F-Antikörper in den absorbierten Antiseren vorhanden sein, außerdem fusionieren die schwächeren Linien der Doppeldiffusion nicht mit irgendwelchen Linien der Unterketten. Auch gereinigte α- oder β-Ketten wurden zur Feststellung ihrer Linien benutzt.
    Notes: Summary The correlation of the antigenicities among native hemoglobins and their subunit chains were investigated by the absorption of antisera and the combination of urea added immunoelectrophoresis with double diffusion. Alphachain showed identity with Hb-F but partial identity with β-chain and Hb-A. Beta-chain showed identity with Hb-A but α-chain and Hb-F showed partial identity with this chain. Gamma-chain showed identity only with Hb-F and its antigenicity was considered as being different from those of α- or β-chains. The lines of α-, β-and ψ-chains were reconfirmed from the facts that the appearance of them depended always on the existence of anti-Hb-A or anti-Hb-F antibodies in the absorbed antisera and the minor component lines of

    Description / Table of Contents: Summary 5 different antibody specificities can be differentiated in heterologous antilymphocytic antiserum of the rabbit: 1 anti-erythrocyte/leukocyte/thrombocyte/liver/kidney antibody. 2 anti-leukocyte/thrombocyte/liver/kidney antibody. 3 anti-leukocyte antibody. 4 anti-erythrocyte antibody. 5 anti-serumprotein antibody. Anti-erythrocyte and anti-leukocyte antibodies were found in the 19 S and 7 S globulin fraction, whereas antibodies directed against an antigen common to erythrocytes and leukocytes could only be demonstrated in the 7 S fraction. The practical implications of this latter type of antibody were discussed. No blood group specificity was demonstrated. The crossreactivity of anti mousecell antibodies with blood cells of the rat did not include all the antibodies in the antiserum.
    Notes: Zusammenfassung Im heterologen Anti-Mäuse- bzw.-Rattenlymphocytenserum von Kaninchen ließen sich die folgenden Antikörpertypen differenzieren: 1 Anti-Erythrocyten/Leukocyten/Thrombocyten/Leber/Nierenantikörper. 2 Anti-Leukocyten/Thrombocyten/Leber/Nierenantikörper. 3 Anti-Leukocytenantikörper. 4 Anti-Erythrocytenantikörper. 5 Anti-Serumproteinantikörper. Während anti-erythrocytäre und anti-leukocytäre Antikörper sowohl in der 7 S als auch in der 19 S Fraktion auftraten, wurden Antikörper, die gegen ein den Erythrocyten und Leukocyten gemeinsames Antigen gerichtet sind, nur in der 7 S-Fraktion nachgewiesen. Die Bedeutung des letzteren Antikörpertyps, insbesondere für die Absorption der Erythrocytenagglutine wird diskutiert. Neben dem Fehlen der Gruppenspezifität wird gezeigt, daß die Kreuzreaktion mit Rattenblutzellen auf einen Teil der Antikörper beschränkt ist.

    Notes: Abstract The temperature and flow field of natural convection in a Hele-Shaw cell is visualized by using a liquid-crystal tracer. The tracer photographs obtained by this method are compared with the interferograms of previous experiments using the same experimental setup, and the applicability of the present methods is validated. Quantitative data of the temperature and velocity were obtained by applying a colour-image-processing technique to the visualized images.

    Notes: Abstract An acyl-coenzyme A carboxylase that carboxylates acetyl-CoA, butyryl-CoA, propionyl-CoA, and succinyl-CoA was purified from Myxococcus xanthus. Since the enzyme showed maximal rates of carboxylation with propionyl-CoA, the enzyme is thought to be propionyl-CoA carboxylase. The apparent K m values for acetyl-CoA, butyryl-CoA, propionyl-CoA, and succinyl-CoA were found to be 0.2, 0.2, 0.03, and 1.0 mM, respectively. The native enzyme has a molecular mass of 605–615 kDa and is composed of nonidentical subunits (α and β) with molecular masses of 53 and 56 kDa, respectively. The enzyme showed maximal activity at pH 7.0–7.5 and at 25–30°C, and was affected by variation in concentrations of ATP and Mg2+. During development of M. xanthus, the propionyl-CoA carboxylase activity increased gradually, with maximum activity observed during the sporulation stage. Previous work has shown that a propionyl-CoA-carboxylase-deficient mutant of M. xanthus reduces levels of long-chain fatty acids. These results suggest that the propionyl-CoA carboxylase is also responsible for the carboxylation of acetyl-CoA to malonyl-CoA used for the synthesis of long-chain fatty acids during development.
