    Notes: To establish a three-dimensional in vitro test system mimicking the physiological situation of the oral cavity, organotypic co-cultures consisting of primary gingival cells on a collagen matrix with fibroblasts were generated. The histomorphological development after 7 and 14 d revealed close similarity with the non-keratinized gingiva epithelium. Furthermore, as epithelial specific markers synthesis and localization of keratins as well as the deposition of basement membrane components were assessed on frozen sections by immunofluorescence and keratin expression by in situ hybridization. Primary keratinocytes in conventional culture stained positive for keratin K14 and the mucosal differentiation-specific keratins K4 and K13, while primary fibroblasts, isolated from the same tissue source, and also some keratinocytes, were positive for vimentin. In organotypic co-cultures the keratinocytes formed a multilayered epithelium within 14 d containing basal cells and flattened cells in the uppermost layers. Comparable to native non-keratinized gingiva keratin 14 gene expression was clearly detectable in the basal cell compartment but showed extending immunolocalization. In addition, particularly at the early stage (7d), basally located keratinocytes were also vimentin positive. According to morphological differentiation K4 and K13 were detectable in suprabasal position at the RNA and protein level. The major basement membrane constituents collagen type IV and laminin increased with time revealing first an interrupted and later a fully extended staining underneath the basal cells. Maintenance of basal cell function was further demonstrated by cell proliferation (BrdU incorporation) which was initially high (7d) but declined towards the later stages (14–21 d). The results demonstrate i) that this co-culture system leads to a stratified surface epithelium with morphological and biochemical characteristics of the non-keratinized gingiva epithelium and ii) that a state of physiological tissue balance was reached, thus rendering a suitable model for tissue compatibility studies.

    Notes: Abstract. A decrease in gap junction-mediated cell-to-cell communication has previously been observed in monolayer cultures of human keratinocytes (HaCaT cells) expressing the human papillomavirus type 16 E5 (HPV16 E5) gene and attributed to the reduced phosphorylation of connexin 43, the most abundant connexin in HaCaT cells. In line with this observation, we have now analyzed the effect of HPV16 E5 on connexin 43 expression in raft cultures produced by transfected HaCaT cells. These keratinocytes transcribe HPV16 E5 under the control of a dexamethasone-inducible promoter. Our results show that treatment with dexamethasone leads to an almost complete disappearance of connexin 43 in rafts expressing the E5 gene but not in control rafts. In our study we discuss the possible effects of this downregulation on cell-cell communication and cellular malignant transformation.

    Notes: Abstract  Cell-matrix interactions and the ordered deposition of basement membrane (BM) components are of major importance for the maintenance of tissue homeostasis in complex epithelia. This aspect was studied in vitro in a coculture system designed as an oral mucosa model. As crucial epithelial features the kinetics of proliferation, expression of site-specific keratins as well as integrin patterns in correlation to synthesis of BM components were assessed by immunohistochemistry and in situ hybridization. Comparison with non-cornified gingiva as tissue of origin revealed different stages of epithelial development, eventually leading to complete reconstruction within a time frame of 1–3 weeks. First, the initial activated stage up to 1 week was characterized by (a) high keratinocyte proliferation, (b) extended expression of the basal cell-specific keratin K5 and (c) a patchy pattern of the differentiation-specific keratins K4 and K13. Second, after 2 weeks the improvement of histoarchitecture correlated to (a) predominant K5 expression in the basal and (b) extension of K4 and K13 within the suprabasal cell compartment, (c) high expression of integrins α3β1 and α6β4 including their ligand laminin-5 and (d) accumulating deposition of basement membrane components. Third, virtually complete tissue normalization at 3 weeks was indicated by (a) restriction of K5 to the basal cell area, (b) regular suprabasal localization of K4 and K13, (c) polarization of integrins to basal and parabasal cells and (d) linear codistribution of collagen IV, “classical” laminin (-1 or -10) and laminin-5 underneath the basal cells. Thus, these organotypic cocultures represent relevant equivalents for non-keratinized oral mucosa with typical gingival differentiation features and in addition suitable models for preclinical trials such as prospective dental material testing.

    Description / Table of Contents: Summary The aim of this study was to examine to what extent excess space must be taken into consideration as an etiological factor in upper canine impaction with special attention paid to the role of the adjacent laterals. To clarify this question the pre- and post-treatment models of 63 patients with a total of 84 impacted cuspids were measured and the cephalograms of 116 patients havign 144 impacted cuspids were analyzed. The results of this study revealed: 85.5% of the cuspids were palataly impacted while 15.5% were labially impacted. An arch deficiency was found in only 18% of the palatally impacted cuspids, whereas there was an arch-length deficiency in 46% of the buccally impacted. In 35% of the cases there was a correlation between peg shaped laterals and palatal impaction, however, such was not present in te patients with vesbibular retention. Finally the cephalometric data revealed horizontal growth characteristics in 80% of the palatally impacted canines as opposed to 23% in those labially impacted.
    Notes: Zusammenfassung Ziel dieser Untersuchung ist es festzustellen, inwieweit ein Platzüberschuß als ätiologischer Faktor für die Impaktierung der Oberkieferenckzähne, unter besonderer Berücksichtigung der Rolle der seitlichen Schneidezähne, in Betracht gezogen werden muß. Zur Klärung dieser Frage wurden sowohl die Anfangs- und Abschlußmodelle von 63 Patienten mit 84 impaktierten Eckzähnen als auch die Fernröntgenseitenbeilder von 116 Patienten mit 144 verlagerten Eckzähnen vermessen. Die Ergebnisse dieser Studie zeigen: Die palatinale Verlagerung Überwog mit 84,5% deutlich gegenüber der verstibulären Retention mit 15,5%. Ein Platzmangel konnte lediglich bei 18% deer Patienten mit palatinaler Verlagerung im Gegensatz zu 46% bei vestibulärer Impaktierung festgestellt werden. Hypoplastische seitliche Schneidezähne traten in 35% der Fälle mit palatinaler Verlagerung, edoch bei keinem Patienten mit vestibulärer Retentiion auf. Die kephalometrische Analyse zeigte ebenfalls einen unterschiedlichen Gesichtsschädelaufbau bei palatinaler und vestibulärer Verlagerung. Während 80% der Patienten mit palatinaler impaktierung einen horizontalen achstumtyp aufwiesen, wurde ein solcher nur in 23% der Fälle mit vestibulärer Retention gefunden.

    Description / Table of Contents: Summary In orthodontics, removable acrylic appliances are preferably produced from methylmetacrylates. However, in the adjacent oral mucosal tissue, cell damage may be caused by the evaporation of residual monomer. The aim of this study was to modify the classic agar overlay assay in order to apply histochemical methods by comparing conventionally used transformed mouse fibroblasts (L-929) with primary human gingival fibroblasts to further elucidate the term toxicity. While proliferation was assessed via the incorporation of the base analogon bromdesoxyuridine, differentiation was investigated by detection of the fibroblast-specific intermediate filament vimentin. After the monomers had taken effect, reduced proliferation extending over the inhibition area was observed by indirect immunofluorescence, independent of cell type. In contrast to this observation, a few cells within the inhibition area which could not be clearly detected by neutral red staining still exhibited mitotic activity. Detection of the differentiation-specific intermediate filament vimentin was comparable with the degree of neutral red fading visible in the classic agar overlay assay. The study showed that the inhibition areas with primary gingival fibroblasts were smaller (approximately 1/3) after monomer action compared with conventionally applied transformed fibroblasts. The results indicate that the modified assay is comparable with the classic method. Evaluation of neutral red staining with respect to toxic material influence can moreover be supplemented by histochemical studies of typical cell properties.
    Notes: Zusammenfassung In der Kieferorthopädie werden zur Herstellung herausnehmbarer Geräte hauptsächlich Kunststoffe auf der Basis von Methylmethacrylaten verwendet. Durch die Restmonomerabgabe besteht dabei die Gefahr einer toxinbedingten Schädigung von Zellen des Umgebungsgewebes. Ziel der vorliegenden Arbeit war es, durch Erweiterung des klassischen Agar-Overlay-Tests histochemische Verfahren zum Nachweis von Zellschädigungen zu prüfen sowie durch den Vergleich konventionell eingesetzter transformierter Mausfibroblasten (L-929) mit primären humanen Gingivafibroblasten den Toxizitätsbegriff näher zu charakterisieren. Untersuchungskriterien waren die Zellproliferation, welche durch die Inkorporation des Basenanalogons Bromdesoxyuridin analysiert wurde, und die Zelldifferenzierung durch den Nachweis des fibroblastenspezifischen Intermediärfilaments Vimentin. So konnte nach Einwirken der getesteten Monomere unabhängig vom Zelltyp fluoreszenzoptisch eine hemmhofübergreifende Verminderung der Proliferation beobachtet werden. Demgegenüber waren noch einzelne mitotisch aktive Zellen im Zentrum des Hemmhofes sichtbar, die mit Neutralrot nicht immer zweifelsfrei anfärbbar waren. Der Nachweis des differenzierungsspezifischen Intermediärfilaments Vimentin war größenordnungsmäßig mit dem Entfärbungsgrad des klassischen Agar-Overlay-Tests nach Neutralrot-Gabe vergleichbar. Die Untersuchungen ergaben ferner, daß unter dem Einfluß der Monomere die Hemmhöfe bei primären Gingivafibroblasten deutlich kleiner waren (etwa ein Drittel) als bei der konventionell eingesetzten Mausfibroblasten-Zellinie. Die Befunde zeigen, daß der modifizierte Test mit der klassischen Vorgehensweise vergleichbar ist. Durch histochemische Untersuchungen charakteristischer Zelleigenschaften kann darüber hinaus die Bewertung der Neutralrot-Vitalfärbung bezüglich einer toxischen Materialeinwirkung ergänzt werden.

    Description / Table of Contents: Summary A total of 33 different substances used in orthodontic therapy were investigated for their potential to irritate mucous membranes. As an alternative to animal testing, the HET-CAM test (Hens Eggs Chorioallantois Membranes Test) was employed. The effects of the substances on the vessels of the chorioallantois membrane, accordance with manufacturers' instructions, plastic materials used for removable orthodontic appliances and bracket bonding caused no irritation. Liquid primers for bracket bonding and monomer substances were highly irritating. Etching gels for decalcification of the enamel surfaces were far less irritating than liquid etching substances. Thus, primers should be handled with care; the use of liquid monomers should be avoided in the oral cavity; and etching gels should be preferred to liquid etching substances.
    Notes: Zusammenfassung Insgesamt 33 verschiedene Materialien, die in der kieferorthopädischen Behandlung Anwendung finden, wurden auf ihre schleimhautreizende Wirkung untersucht. Als Alternative zum Tierversuch wurde der HET-CAM-Test (Hens Egg Chorioallantois Test) angewendet. Die Wirkung der Substanzen auf die Blutgefäße der Chorion-Allantois-Membran (CAM), wie Hämorrhagie, Lysis und intravasale Koagulation, wurde auf Video aufgezeichnet und ausgewertet. Kunststoffe zur Herstellung herausnehmbarer Apparaturen und zur Bracketbefestigung verursachten keine Reizung, sofern sie entsprechend den Anweisungen der Hersteller verarbeitet wurden. Primer zur Bracketbefestigung und Monomere waren hingegen stark reizend. Ätzgele zum Anätzen des Zahnschmelzes führten zu weit geringerer Reizung als flüssige Ätzmittel. Aus der Untersuchung folgt, daß Primer vorsichtig und sparsam angewendet werden sollten. Monomere sollten nicht in Schleimhautkontakt kommen, zum Beispiel beim direkten Unterfüttern; Ätzgele sollten flüssigen Ätzmitteln vorgezogen werden.
