    Notes: Summary Correlation of morphological and electrophysiological data strongly suggest that in rat, the giant cells of the lateral vestibular nucleus (L.V.N.) are electrotonically coupled. 1. in addition to “active zones” large terminals synapsing on the perikaryon and/or the main dendritic trunk of the cells bear “gap” junctions which are interpreted as low electrical resistance pathways between neurons. 2. electrical activity of the giant cells was recorded intracellularly as the vestibulo-spinal tract was stimulated. Graded antidromic stimulation produced graded antidromic depolarizations (G.A.Ds) in 69% of cells with high threshold axons. 3. the latency of the G.A.Ds was too short to allow for chemical transmission through afferents or recurrent collaterals. 4. collision experiments demonstrated that directly evoked spikes blocked the antidromic spikes but did not block the G.A.Ds which thus were accounted for by activation of cells others than the impaled ones. 5. lesion experiments indicated that afferent fibers from the spinal cord terminate exclusively in the dorsal part of the L.V.N. Since G.A.Ds were recorded all throughout the nucleus, they were not excitatory post synaptic potentials (EPSPs) from spinal afferents. 6. when the strength of the spinal cord stimulation was increased EPSPs were also generated but they were distinct from the G.A.Ds by their latencies, time course and maximum amplitude. 7. since no direct contact is observed between neurons it is inferred that, as in other documented cases, coupling between giant cells is mediated by way of presynaptic fibers.

    Notes: Nicotinic acetylcholine receptors (nAChRs) are expressed in the midbrain ascending dopaminergic system, a target of many addictive drugs. Here we assessed the intracellular Ca2+ level by imaging fura-2-loaded cells in substantia nigra pars compacta in mouse brain slices, and we examined the influence on this level of prolonged exposures to nicotine using mice lacking the nAChR β2-subunit. In control cells, superfusion with nicotine (10–100 μm) caused a long-lasting rise of intracellular Ca2+ level which depended on extracellular Ca2+. This nicotinic response was almost completely absent in β2–/– mutant mice, leaving a small residual response to a high concentration (100 μm) of nicotine which was inhibited by the α7-subunit-selective antagonist, methyllycaconitine. Conversely, the α7-subunit-selective agonist choline (10 m m) caused a methyllycaconitine-sensitive increase in intracellular Ca2+ level both in wild-type and β2–/– mutant mice. Nicotine-elicited Ca2+ mobilization was reduced by the Na+ channel blocker tetrodotoxin (TTX) and by T-type Ca2+ channel blocking agents, whereas the choline-elicited Ca2+ increase was insensitive to TTX. Neither nicotine nor choline produced Ca2+ increase following inhibition of the release of Ca2+ from intracellular stores by dantrolene. These results demonstrate that in nigral dopaminergic neurons, nicotine can elicit Ca2+ mobilization via activation of two distinct nAChR subtypes: that of β2-subunit-containing nAChR followed by activation of Na+ channel and T-type Ca2+ channels, and/or activation of α7-subunit-containing nAChR. The Ca2+ influx due to nAChR activation is subsequently amplified by the recruitment of intracellular Ca2+ stores. This Ca2+ mobilization may possibly contribute to the long-term effects of nicotine on the dopaminergic system.

    Notes: Summary In the CNS, the exocytosis which accompanies transmitter releases occurs at the level of a resynaptic grid. Possible alterations in the grid as a function of this phenomenon were searched for at the synapses established by unmyelinated club endings on the Mauthner cell of teleosts. The number of vesicle openings generated by aldehyde fixation was diminished by cooling the preparation and enhanced after perfusion with a high-KCl Ringer solution. Morphometric analysis of the grid showed that under these conditions the mean distance between its constituent elements, the presynaptic dense projections, increased with the number of exocytotic events. Parallel changes were observed for the mean diameter of the spaces left free between these dense projections, suggesting that vesicle exocytosis produces a transient enlargement of the space where it takes place. These observations indicate that the presynaptic grid is more dynamically involved in the secretory process than previously conceived. It is therefore hypothesized that (i) the movement of the dense projections is a consequence of their interaction with the plasma membrane, and (ii) the distortion of the grid could underlie regulatory mechanisms by which the number of released vesicles is limited after each impulse. It is also proposed that the dense projections contribute to the stabilization of the plasma membrane, thereby preventing its randomization following intense release.
