    Notes: Abstract We have used nascent strand determination analysis to map start sites of DNA replication in the mouse ribosomal gene cluster in which individual copies of the ribosomal genes are separated by intergenic spacer regions. One origin of bidirectional replication (OBR) was localized within a 3 kb region centered about 1.6 kb apstream of the rDNA transcription start site. At least one additional initiation site is situated near the 3′ end of the transcription unit. Adjacent to the OBR at the transcription start site are located two amplification-promoting sequences, i.e., APS1 and APS2. Nuclease-hypersensitive sites were identified in both the two APSs as well as in the OBR region, thus indicating that these sequences have an altered chromatin structure. In the OBR an intrinsically bent region, a purine-rich element and other prospective initiation zone components are found.

    Description / Table of Contents: Zusammenfassung Über Fortschritte bei der Wickbold-Verbrennungstechnik wurde berichtet. Ein Quarzkölbchen als Probenbehälter für feste und flüssige Substanzen, eine Zusatzeinrichtung für Flüssiggase, eine „Mikro“-Apparatur und ein neuer Saugbrenner mit regulierbarem Probendurchsatz wurden entwickelt. Für Makro-Chlor-Serienanalysen hat sich ein halbautomatisches Titrierverfahren nach Verbrennung und Intensiv-Absorption des Chlorids bewährt. Neue Anwendungsmöglichkeiten zur Elementanalyse durch Bildung und Abtrennung flüchtiger Chloride nach Cl2-Dotierung wurden aufgezeigt und Ergebnisse bei Spurenanalysen von Cd, Pb, Zn, As und Se in Polymeren und biologischen Proben diskutiert.
    Notes: Summary Improvements in the Wickbold combustion technique are reported. A quartz-flask sample-container for solids and liquids, an equipment for liquified gases, a micro combustion-apparatus and a new suction burner with adjustable flow control have been developed. A semi-automatic routine procedure for chlorine is suggested. There are new aspects in metal and non-metal analysis by H2/O2/Cl2-combustion with production of volatile chlorides. Results of Cd, Pb, Zn, As, and Se trace analysis in polymers and biological samples are discussed.

    Description / Table of Contents: Summary A combination method for analysis of small amounts of tensides consisting of blowing out/DC separation/IR microspectrometry is presented. Recent data on the behavior of different tensides on blowing out indicate the optimal conditions of working, the efficiency and the limits of the method as well as the feasibility of fractionated separations of tensidic substances. An IR spectroscopic micromethod subsequent to thin-layer chromatographic pre-separations enables impeccable spectra of substance quantities of less than 1μg under routine conditions using an IR microscope or a modern spectrometer with computer-controlled evaluation. Demonstration and identification of a few micrograms of tenside substance according to this method of working are illustrated by practical examples of analysis.
    Notes: Zusammenfassung Eine Methodenkombination Ausblasen/DC-Auftrennung/IR-Mi-krospektrometrie wird zur Analyse kleiner Tensidmengen vorgestellt. Neuere Erkenntnisse über das Verhalten verschiedener Tenside beim Ausblasen zeigen die optimalen Arbeitsbedingungen, die Leistungsfähigkeit und die Grenzen des Verfahrens auf und weisen auf die Möglichkeit fraktionierter Trennungen tensidischer Substanzen hin. Eine IR-spektroskopische Mikrotechnik im Anschluß an dünnschicht-chromatographische Vortrennungen ermöglicht einwandfreie Spektren von Substanzmengen ≦1μg im Routinebetrieb mit Hilfe eines IR-Mikroskopes oder eines modernen Spektrometers mit computergesteuerter Auswertung. Nachweis und Identifizierung einiger Mikrogramm Tensidsubstanz nach dieser Arbeitstechnik werden an praktischen Analysenbeispielen aufgezeigt.

    Description / Table of Contents: Zusammenfassung Durch Änderungen im Verbrennungsteil und bei der Gasführung wurde beim Antek-Stickstoff-Detektor eine deutliche Steigerung der Impulsausbeute und ein erweiterter Linearitätsbereich der Eichkurven erreicht. Mit 0,5 bis 10 mg bzw. 1 bis 10μl Probemenge lassen sich Gehalte an chemisch gebundenem Stickstoff von 0,5 ppm bis ca. 3% bestimmen. Die Intensivverbrennung bewirkt weitgehende Unabhängigkeit von der N-Bindungsart mit Einschränkungen bei Azo- und Hydrazo-Verbindungen. Vergleichende Mikro- und Spurenanalysen verschiedener chemischer und technischer Produkte belegen die Leistungsfähigkeit und die Vorteile des modifizierten Pyro-Chemilumineszenz-Reaktors gegenüber der Kjeldahl-Methode.
    Notes: Summary In order to develop a versatile method for microanalysis of chemically bound nitrogen, several parts of the Antek Nitrogen Detector system have been modified. These modifications have resulted in a fourfold increase in impulse rate and a linear calibration curve over four decades of concentration. Chemically bound nitrogen can be measured directly in 0.5–10 mg or 1–10μl samples from 0.5ppm up to about 3% N if appropriate operating parameters are chosen. Analyses of different types of nitrogen compounds have yielded significantly better recoveries than the Kjeldahl method, except for azo compounds. The method has been tested with various substances and proved superior to the Kjeldahl method in handling and efficiency, with acceptable results.

    Description / Table of Contents: Zusammenfassung Die Knallgasverbrennung nach Wickbold ist als Probenvorbereitung zur Hg-Bestimmung durch flammenlose Atomabsorption gut geeignet. Die Direktmessung in den Verbrennungsgasen in einer Verbrennungsküvette ist nur begrenzt anwendbar, etwa bei halogenfreien wäßrigen oder organischen Lösungen, z.B. Essigsäure. Sicherer ist die Naßabsorption (H2SO4/KMnO4-Lösung) mit anschließender Reduktion und Ausblasen des atomaren Hg-Dampfes, wobei ein Chlorüberschuß bei der Verbrennung (CCl4-Zugabe) quantitativ Hg-Absorption durch Bildung gut löslicher Hg-Chloride gewährleistet. Das Verfahren ist universell anwendbar und erfaßt einen weiten Konzentrationsbereich (0,1 ppb bis zu Prozentgehalten Hg).
    Notes: Abstract The Wickbold oxyhydrogen combustion is a suitable method for sample preparation in trace analysis of mercury, combined with flameless atomic absorption spectrometry. Samples free from halogens, especially aqueous or organic solutions (e.g. acetic acid), may be analysed by direct atomic absorption measurement of mercury in a combustion-absorption cell. The most reliable method is combustion in presence of chlorine (forms slightly soluble mercury-chlorine compounds), absorption of mercury in H2SO4/KMnO4 solution, followed by reduction, evaporation of mercury vapour by an air/N2-current and flameless atomic absorption spectrometry at 253.7nm. The method is very versatile and allows mercury analysis in a wide concentration range (10−8 to ≥ 10% Hg).
