    Notes: Abstract A polymerase chain reaction and an oligonucleotide probe method were developed for the specific detection and identification of the Gram-negative bacterium Pseudomonas stutzeri. This is an important species in environmental studies and can also be present in clinical samples. The primers for the amplification and the probe were designed from the analysis of 16S rRNA sequences. The PCR/oligonucleotide probe method, in combination with a direct lysis DNA extraction and an effective purification method, is an alternative to viable methods for the detection of P. stutzeri in environmental samples.

    Notes: Abstract R-Bodies have been found in a recently isolated pseudomonas-like free-living hydrogen oxidizing bacterium. Their isolation, fine structure and chemical composition are described and compared with the R-bodies from the kappa particles (Caedobacter), obligate endosymbionts of Paramecium aurelia. The 2K 1 R-bodies exhibited essential characteristics of the kappa R-bodies; however, their size and some other structural aspects proved that they represent a new type of R-bodies. The presence of phage tail-like particles in cells induced with Mitomycin C is in favour of the hypothesis that the R-bodies might be coded by defective prophages, or by extrachromosomal elements.

    Notes: Abstract The coryneform hydrogen bacterium strain GZ 29, assigned to Corynebacterium autotrophicum fixed molecular nitrogen under autotrophic (H2, CO2) as well as under heterotrophic (sucrose) conditions. Physiological parameters of nitrogen fixation were measured under heterotrophic conditions. The optimal dissolved oxygen concentration for cells grown in a fermenter with N2 was rather low (0.14 mg O2/l) compared with cells grown in the presence of NH 4 + (4.45 mg O2/l). C. autotrophicum GZ 29 had a doubling time of 3.7 h at 30°C with N2 as N-source and sucrose as carbon source and at optimal pO2. Acetylene reduction reached values of 12 nmoles of ethylene produced/minxmg protein. Although the oxygen concentration in the growing culture was kept constant, the optimal dissolved oxygen tension for the acetylene reduction assay shifted to higher pO2-values. The overall efficiency of nitrogen fixation amounted to 22 mg N fixed/g sucrose consumed; it reached a maximal value of 65 mg N fixed/g sucrose consumed at the beginning of the exponential growth phase. Intact cells reduced acetylene even under anaerobic test conditions; further anaerobic metabolic activity could not be ascertained so far.

    Notes: Abstract A 6.1 kilobase DNA fragment has been cloned from Pseudomonas stutzeri AN11, a naphthalene- degrading marine isolate. The cloned DNA constitutively expressed salicylate hydroxylase and catechol-2,3-dioxygenase activities in E. coli, both activities being slightly induced by salicylate. The structural genes have been located in the cloned fragment, which hybridized both with the chromosomal DNA and with a 60 kilobase plasmid of P. stutzeri AN11, and showed homology with the corresponding regions of TOL and NAH plasmids.

    Notes: Chlorella sp. strain VJ79 was isolated from a total heterotrophic count of a wastewater collector. It grows autotrophically, heterotrophically, and mixotrophically on a variety of organic substrates. Glucose and serine promote a mixotrophic growth from which the yield is higher than the sum of autotrophic and heterotrophic yields, but serine assimilation requires light. The interaction of glucose and light was studied in proliferating and nonproliferating cells by respirometry (IRGA and Warburg) and growth experiments. Glucose inhibits the photosynthetic CO2 fixation ten-fold and modifies the pigmentary system as it does in heterotrophic cultures. Light inhibits glucose uptake and assimilation, but under mixotrophic conditions maximal utilization of glucose is obtained. Mutants defective in autotrophic growth were isolated by mutagenesis with nitrosoguanidine. They show a degenerated pigmentary system and a mixotrophic growth yield equal to that of the heterotrophic growth. The analysis of the mixotrophic system shows that light energy, dissipated during autotrophic growth, is used under mixotrophic conditions. From the increase in growth, the increase in photosynthetic efficiency can be calculated as ca. sixfold.
