    Notes: Abstract The effects of temperature (60°–70°C) and medium composition (complex and defined) on ethanol tolerance ofBacillus stearothermophillus LLD-15, an L-lactate dehydrogenase mutant have been determined in shake flasks under aerobic conditions. In all cases, there was complete inhibition of growth in the presence of 6%v/v ethanol.B. stearothermophillus LLD-15 was found to be less tolerant to ethanol at 70°C than at 60°C and also less tolerant to ethanol in a defined medium than in a complex medium.

    Notes: Abstract Xanthobacter Py2 grows on propene as sole carbon source, converting propene to propene oxide (epoxypropane) using an alkene-specific monooxygenase, as the first step in catabolism. Four mutants, NZ1–4, with a propene− propene oxide+ phenotype were isolated by 1-methyl-3-nitro-1-nitrosoguanidine mutagenesis or by enrichment with the suicide substrate vinylidene chloride, and were shown to have lost the ability to convert alkenes to epoxides. All four mutants were complemented by a number of clones of Xanthobacter Py2 chromosomal DNA in the broad-host-range cosmid pLAFR5, some of which appeared to be non-overlapping. Representatives of the different clones obtained were transferred into Xanthobacter autotrophicus JW33 and one, pNY2, the most frequently isolated clone, was shown to express an inducible, fully functional propene monooxygenase. Subcloning revealed that all four mutants were complemented by a 2.4-kb EcoRI-PstI fragment situated at one end of the cosmid insert. However, activity in X. autotrophicus JW33 could only be expressed from pNY2, containing the complete insert (25 kb), suggesting a large operon or some form of long-range control. pNY2 failed to express in E. coli. In X. autotrophicus JW33 [pNY2] at least three new polypeptides were evident after induction with propene compared with a control carrying only the cosmid pLAFR5.

    Notes: Abstract  Pseudomonas fluorescens NCIMB 11671 metabolises α-pinene via α-pinene oxide and 2-methyl-5-isopropylhexa-2,5-dienal. Mutants unable to grow on α-pinene and/or α-pinene oxide have been isolated by N-methyl-N′-nitro-N-nitrosoguanidine mutagenesis, including an unexpected phenotype able to grow on α-pinene but not on α-pinene oxide. The mutants have been classified on the basis of their α-pinene monooxygenase, α-pinene oxide lyase and aldehyde dehydrogenase activities. Biotransformation of α-pinene by the wild-type and mutant strains has revealed evidence for alternative routes for pinene metabolism to that already proposed.

    Notes: Summary The intracellular location of methane mono-oxygenase (MMO) activity in the methanotroph Methylococcus capsulatus (Bath) has been shown to depend primarily on the availability of copper. MMO activity was observed in the particulate fraction of cell extracts under conditions of copper excess but switched to a soluble location in response to copper stress. The two activities could be differentiated by sensitivity to a range of inhibitors and by major changes in the polypeptide banding patterns on denaturing polyacrylamide gels. MMO activity concomitant with the oxidation of ethanol was only observed in cells with particulate MMO activity but could be lost independently in response to copper stress. Examination of other methanotrophs indicated that the copper effect could explain a similar switch in intracellular location observed in Methylosinus trichosporium OB3b but that some methanotrophs do not have the capacity to overcome copper stress in this way.

    Notes: Continuous fermentation with cell recycle proved very effective in increasing the ethanol volumetric productivity of the thermophilic facultative anaerobe, Bacillus stearothermophilus strain LLD-15, on sucrose at 70°C. When complete cell recycle was used, cell viability decreased after a few residence times and sucrose consumption was reduced. Operation using a constant bleed rate resulted in greater stability and higher ethanol volumetric productivities. A mathematical model based on maintenance energy requirements provided an adequate description of the system. © 1994 John Wiley & Sons, Inc.

    Notes: Abstract Methionine, biotin, nicotinic acid and thiamine were found to be absolute requirements for growth ofBacillus stearothermophilus strain LLD-15, an L-lactate dehydrogenase mutant, at 70°C. A defined medium for aerobic growth did not support anaerobic growth. Physiological and subsequent taxonomic studies showed that strain LLD-15 is not a derivative ofB. stearothermophilus NCA 1503 as originally thought, but bears some similarity toB. caldotenax.
