    Notes: Summary Motor evoked potentials (MEPs), monitoring the motor function directly, are superior to somatosensory evoked potentials (SSEPs) in monitoring the motor system during spinal surgery. Reliable MEPs are difficult to elicit under normal anaesthesia. Using intravenous anaesthesia with either ketamine or etomidate infusion, we performed intraoperative MEP monitoring in 12 spinal operations for 11 cases from February 1992 to May 1992. For anaesthesia, ketamine was used in 5, etomidate in 7, fentanyl was supplemented in all, muscle relaxation at 30% to 50% of pre-anaesthetic muscle power was maintained with atracurium or vencuronium infusion. Transcranial bipolar electrical stimulation was used to induce MEPs. Concomitant SSEP monitoring was performed in 3. No significant anaesthesia related side effects were noted except one episode of unpleasant dream occurred in the ketamine anaesthesia group. Successful monitoring was achieved in 10 sessions. In 5 of which warning to the surgeons was made due to sudden MEP deterioration, which recovered followed by definite management in four and persisted in one. In the other 5 sessions, no warning was made due to stationary or gradual change in MEPs. Bilateral two-channel recordings were used in 3 sessions. In 2 of which unilateral transient change was noted. Loss of SSEPs was noted in one despite unchanged MEPs, in whom only new sensory deficits occurred postoperatively. Compared to the baseline MEPs in terms of latency and amplitude, the final MEPs improved in 5 sessions, did not change significantly in 4 sessions, deteriorated in one session, and were correlated well with the immediate postoperative motor status. In our small series, the intraoperative MEP monitoring showed neither false negative nor false positive result. It is concluded that the intraoperative MEP monitoring is feasible under intravenous ketamine or etomidate anaesthesia and valuable in spinal surgery.

    Notes: Measurements of TlBaCaCuO-based dc superconducting quantum interference devices (SQUIDs) performed using a practical feedback circuit gave a flat frequency response and an energy sensitivity of 6×10−30 J/Hz at 5 kHz and 77 K with an 80 pH SQUID. Flux-locked loop operation was demonstrated to be strongly effective in eliminating the magnetic field hysteresis of the SQUID. The SQUIDs were operated in the flux-locked loop at temperatures over 90 K and the 1/f noise was found to decrease with increasing temperature near 77 K.

    Notes: Low-temperature magnetic properties and crystal structures of melt-spun Sm2Co7B3 compound were characterized. The magnetic measurements in the temperature range 77–450 K indicated that a spin reorientation took place at about 150–160 K. A huge anisotropy was observed (Ha=135 kOe at 300 K, 725 kOe at 77 K) for Sm2Co7B3, although the magnetic moment is rather low. The crystal structure of the Sm2Co7B3 compound was analyzed in detail by Rietveld analysis of powder diffraction for the first time, and revealed that B(4h) atoms are not placed in the Sm(2e) layer but in between the Sm(2e) and Co(6i1) layers.

    Notes: We have made a series of single-level dc superconducting quantum interference devices (SQUIDs) from 4-μm-thick TlBaCaCuO films with large grain sizes and operated them in liquid nitrogen. Although device characteristics could not be precisely controlled, some devices had white-noise levels that approached thermally limited noise above ∼1000 Hz. In addition, devices with 5 and 80 pH loop inductances had 1/ f noise levels at 10 Hz of 2×10−29 and 5×10−29 J/Hz, respectively. The noise levels at these frequencies are comparable to commercial rf SQUIDs operating in liquid helium, but the hysteresis of the voltage-flux characteristic of the high Tc SQUIDs remains large.

    Notes: Aromatic l-amino acid decarboxylase (AADC) is necessary for conversion of l-DOPA to dopamine. Therefore, AADC gene therapy has been proposed to enhance pharmacological or gene therapies delivering l-DOPA. However, addition of AADC to the grafts of genetically modified cells expressing tyrosine hydroxylase (TH) and GTP cyclohydrolase 1 (GCH1), which produce l-DOPA in parkinsonian rats, resulted in decreased production of l-DOPA and dopamine owing to feedback inhibition of TH by dopamine. End-product feedback inhibition has been shown to be mediated by the regulatory domain of TH, and site-specific mutation of serine 40 makes TH less susceptible to dopamine inhibition. Therefore, we investigated the efficacy of using TH with serine 40 mutated to leucine (mTH) in an ex vivo gene-therapy paradigm. Primary fibroblasts (PF) from Fischer 344 rats were transduced with retrovirus to express mTH or wild-type rat TH cDNA (wtTH). Both cell types were also transduced with GCH1 to provide the obligate TH cofactor, tetrahydrobiopterin. PF transfected with AADC were used as coculture and cografting partners. TH activities and l-DOPA production in culture were comparable between PFwtTHGC and PFmTHGC cells. In cocultures with PFAADC cells, PFmTHGC cells showed significant reduction in the inhibitory effect of dopamine compared with PFwtTHGC cells. In vivo microdialysis measurement showed that cografting PFAADC cells with PFmTHGC cells resulted in smaller decreases in l-DOPA and no reduction in dopamine levels compared with cografts of PFAADC cells with PFwtTHGC cells, which decreased both l-DOPA and dopamine levels. Maintenance of dopamine levels with lower levels of l-DOPA would result in more focused local delivery of dopamine and less potential side-effects arising from l-DOPA diffusion into other structures. These data support the hypothesis that mutation of serine 40 attenuates TH end-product inhibition in vivo and illustrates the importance of careful consideration of biochemical pathways and interactions between multiple genes in gene therapy.
