    Notes: Summary The effect of varying oral doses of 11R, 16, 16-trimethyl prostaglandin E2 (TmPGE2) on meal-stimulated gastric acid secretion and serum gastrin concentrations was studied in 10 male subjects with asymptomatic duodenal ulcer disease. A liquid protein meal was infused intragastrically 0.5 h and 3.5 h after drug administration. TmPGE2 inhibited gastric acid secretion in a dose dependent manner during the first meal and no significant effect was observed during the second meal. Except for the highest dose, no TmPGE2 was detected in plasma 3 h after drug administration. The degree of inhibition of meal-stimulated gastric acid was positively correlated with the plasma level of TmPGE2, but it was not due to inhibition of postprandial gastrin release. The results indicate that oral TmPGE2 inhibits meal-stimulated gastric acid secretion but not gastrin release in humans with asymptomatic duodenal ulcer disease.

    Description / Table of Contents: Zusammenfassung. Obwohl eine Vielzahl der an malignen Tumoren erkrankten Patienten kein oder nur ein minimales Ansprechen auf eine Chemotherapie zeigen, werden diese Patienten dennoch mittels standardisierter Therapieregime behandelt. Ein Grund hierfür ist die Unmöglichkeit, vor Therapiebeginn das Ansprechen auf eine Chemotherapie vorherzusagen. Die Bestimmung biochemischer Faktoren im Gewebe von Tumorpatienten, welche ein Tumoransprechen auf eine bestimmte Chemotherapie voraussagen können, könnte jedem Patienten eine auf einer rationalen Basis stehende, individuell angepasste und optimale Behandlung ermöglichen. In diesem Artikel werden wir die wesentlichen Studien zusammenfassen, welche sich mit der Fragestellung beschäftigt haben, ob eine quantitative Analyse der Gen- oder Proteinexpressionen von am Medikamentenstoffwechsel beteiligter Enzyme im Tumorgewebe, das Ansprechen eines Tumors auf ein Medikament im voraus identifizieren kann. Diese Hypothese wurde bisher ausführlich in bei 5-FU basierenden Therapien von Patienten mit colorectalen Carcinomen untersucht. Es zeigte sich, daß eine Assoziation zwischen der quantitativen Analyse der Genexpressionen der Thymidylat Synthase (TS), der Thymidin Phosphorylase (TP) und der Dihydropyrimidin Dehydrogenase (DPD) und dem Tumorresponse bei auf 5-FU basierender Chemotherapien besteht. Die Kombination dieser Marker resultierte in einer Identifizierung aller Therapieresponder. Als Resultat dieser Studien läßt sich zusammenfassen, daß mittels neuester PCR-Analysemethoden die Identifizierung von Genexpressionen möglich ist, welche eine individualisierte Chemotherapie ermöglicht.
    Notes: Abstract. Although a majority of cancer patients show no response or minimal response to any given chemotherapy, all patients are nevertheless placed on standard therapy regimens because there has been no way to identify beforehand those patients who are destined not to respond. Determining the biochemical factors relevant to drug response in each patient's tumor cells prior to treatment should allow optimal therapy to be selected for each patient on a rational basis. In this paper, we summarize studies aimed at determining whether analysis of the quantitative expression levels of genes or proteins involved in cancer drug activity in clinical specimens of tumor tissue could predict the effect of the drug on a tumor. This hypothesis has been tested mostly in 5-fluorouracil (5-FU)-based regimens in colorectal cancer. In the case of 5-FU, the quantitative expression levels of thymidylate synthase, thymidine phosphorylase, and dihydropyrimidine dehydrogenase were found to be associated with tumor response. Using all three of these markers together resulted in very effective identification of responding patients. The salient results of these studies were that, using the PCR enhanced by newly developed methodology, gene expressions can be identified and measured in pretreatment biopsies that are quantitatively associated with either response or non-response to drugs that are used in treating colorectal cancer.

    Description / Table of Contents: Abstract. Systems of economic ratios are very important in the field of operative controlling. Functional equations of economic quantities may be analyzed by comparing actual current values of these variables with planned values or values of a previous period using methods of deviation analysis. In this paper different methods of deviation analysis are compared. They are based on conditions, which should be satisfied by methods used to determine the effects on a depending quantity caused by the change of its components. In some aspects these conditions are different from conditions for methods of deviation analysis used in cost accounting. Based on an approximation of the temporal change of economic quantities a new method of deviation analysis is developed. This concept satisfies all conditions formulated at the beginning of the paper.
    Notes: Zusammenfassung. Große Bedeutung im operativen Controlling haben Kennzahlensysteme. Dabei auftretende funktionale Zusammenhänge zwischen den Kennzahlen lassen sich unter Nutzung der Methoden der Abweichungsanalyse analysieren, indem Soll- und Ist-Werte oder Angaben verschiedener zeitlicher Perioden miteinander verglichen werden. Gegenstand dieser Arbeit ist ein Vergleich verschiedener Konzepte der Abweichungsanalyse. Dazu werden Kriterien formuliert, deren Erfüllung für die Nutzung der betreffenden Methoden im Controlling und für eine möglichst einfache und eindeutige Interpretation der Ergebnisse wünschenswert erscheint. Aus der Sicht der Kennzahlensysteme ergeben sich dabei teilweise andere Anforderungen als die in der Kostenrechnung diskutierten Bedingungen. Für die Analyse der zeitlichen Entwicklung wird ein neues Konzept der Abweichungsanalyse entwickelt, das allen geforderten Kriterien genügt.
