    Notes: Recent development of electron cyclotron resonance (ECR) ion sources in China is reviewed. Emphasis is put on high charge state ECR ion sources which have been mainly developed in China by Institute of Modern Physics (IMP). Presently two ECR ion sources built by IMP for highly charged ion beams are put into operation for cyclotrons and atomic physics research. The development of high charge state ECR ion sources at IMP has progressed with a new magnetic field configuration, better condition for extraction of highly charged ions, high mirror magnetic field, large plasma volume, and special techniques to provide extra cold electrons. These techniques greatly enhance the production of highly charged ions from IMP ECR ion sources. So far more than 185 eμA of Ar11+ and 50 eμA of Xe26+ were produced by the IMP ECR ion sources. The metallic ion beam production was tested and the first beam 40Ca11+ was provided to the cyclotrons at IMP. The beam intensity of 40Ca11+ could reach 130 eμA. The next part of this article will report the latest progress of 2.45 GHz ECR ion sources in China. A 2.45 GHz compact permanent magnet proton ion source was designed and constructed by IMP. A new microwave feeding system is applied on this ion source. The ion source is able to deliver 90 mA of mixed ion beam (H1++H2++H3+) after preliminary commissioning. The article also mentions a small 2.45 GHz ECR ion source which was built by Sichuan University and used for industry applications. The dependence of plasma density and electron temperature on radio frequency power, neutral gas pressure, and different microwave windows was measured by a Langmuir probe on this ion source. © 2000 American Institute of Physics.

    Notes: The goldfish optic nerve can regenerate after injury. To understand the molecular mechanism of optic nerve regrowth, we identified genes whose expression is specifically up-regulated during the early stage of optic nerve regeneration. A cDNA library constructed from goldfish retina 5 days after transection was screened by differential hybridization with cDNA probes derived from axotomized or normal retina. Of six cDNA clones isolated, one clone was identified as the␣Na,K-ATPase catalytic subunit α3 isoform by high- sequence homology. In northern hybridization, the expression level of the mRNA was significantly increased at 2 days and peaked at 5–10 days, and then gradually decreased and returned to control level by 45 days after optic nerve transection. Both in situ hybridization and immunohistochemical staining have revealed the location of this transient retinal change after optic nerve transection. The increased expression was observed only in the ganglion cell layer and optic nerve fiber layer at 5–20 days after optic nerve transection. In an explant culture system, neurite outgrowth from the retina 7 days after optic nerve transection was spontaneously promoted. A low concentration of ouabain (50–100 nm) completely blocked the spontaneous neurite outgrowth from the lesioned retina. Together, these data indicate that up-regulation of the Na,K-ATPase α3 subunit is involved in the regrowth of ganglion cell axons after axotomy.

    Notes: Generations of efficient blue light at 447 nm and red light at 671 nm were achieved by frequency doubling and tripling of a diode-pumped, Q-switched 1342 nm Nd:YVO4 laser with a periodically poled LiTaO3 (PPLT). The blue light at 447 nm was generated by sum-frequency mixing of the fundamental at 1342 nm with the generated second harmonic at 671 nm. The first-order and third-order reciprocals of the PPLT compensated the phase mismatches of second-harmonic and sum-frequency processes, respectively, making them quasiphase matched. The resulting averaged blue light power of 51 mW and red light power of 207 mW under the averaged fundamental power of 500 mW indicate that the PPLT may be used to construct an all-solid-state blue and red dual wavelength laser. © 2001 American Institute of Physics.

    Notes: Abstract Simple sequence repeats (SSRs), or microsatellites, are a new class of PCR-based DNA markers for genetic mapping. The objectives of the present study were to develop SSR markers for barley and to integrate them into an existing barley linkage map. DNA sequences containing SSRs were isolated from a barley genomic library and from public databases. It is estimated that the barley genome contains one (GA)n repeat every 330 kb and one (CA)n repeat every 620 kb. A total of 45 SSRs were identified and mapped to seven barley chromosomes using doubled-haploid lines and/or wheat-barley addition-line assays. Segregation analysis for 39 of these SSRs identified 40 loci. These 40 markers were placed on a barley linkage map with respect to 160 restriction fragment length polymorphism (RFLP) and other markers. The results of this study demonstrate the value of SSRs as markers in genetic studies and breeding research in barley.

    Notes: Abstract Wheat (Triticum aestivum L., 2n = 6x = 42, AABBDD) florets were emasculated and pollinated using two apomictic wheatgrass [Elymus rectisetus (Nees in Lehm.) A. Love & Connor, 2n = 6x = 42, SSYYWW] accessions, one of which produces 2n pollen. A 2n = 42 (BII) hybrid and four 2n = 63 (B III) hybrids were obtained. The spike morphology of the B II hybrid was intermediate to that of its parents. The pollen mother cells (PMCs) of this hybrid contained on average 38.361 and 1.62 II, which was consistent with its disparate genome composition (ABDSYW). Its pollen failed to stain and no BC1 progeny was obtained. The B III hybrids (reduced egg fertilized with unreduced sperm) were grasslike and had a full complement of E. rectisetus chromosomes, the synapsis of which was slightly impaired by wheat haplome and/or cytoplasm. Their PMCs contained on average 16.30 II, 25.72 I, and 1.54 multivalents (III plus IV). Pollen stainability in these hybrids was low (〈1%), and when they were used as females, one 54- and 60-chromosome BC1 were obtained. A mean of 13.25 II was observed in PMCs of the 54-chromosome BC1 and pollen stainability was 10%. Pollen stainability in the 60-chromosome BC1 was only 5%. The use of 2n-pollen-producing E. rectisetus accession accelerated hybrid and BC1 formation and may accelerate the ultimate transfer of apomixis to wheat.

    Notes: Abstract A size-fractionated TaqI genomic library of seashore paspalum (Paspalum vaginatum Swartz) was screened for the presence of (GA) n and (CA) n simple sequence repeats (SSRs). A total of 54 clones with a positive signal were detected among 13,000 clones screened. Forty-seven clones having repeats of n⩾ 3 were identified, of which 85% were perfect, 13% were imperfect and 2% were compound repeat sequences. Five of ten primer pairs synthesized to amplify selected loci resulted in a product in the expected size range and were subsequently used to examine SSR polymorphisms among 46 ecotypes of P. vaginatum. The number of alleles resolved on agarose or polyacrylamide gels were similar and ranged from 6 to 16 with an average of 14 per locus. Phenetic analysis of SSR polymorphisms revealed genetic relationships among the P. vaginatum ecotypes that were in general agreement with relationships determined previously by RAPD analysis of the same plant materials. Further screening of the genomic library did not identify (AT) n , trimeric or tetrameric repeats. Hybridization of an (ATT)8 oligonucleotide probe to genomic DNA isolated from I. batatas, E. coli, Citrullis lanatus and P. vaginatum suggested that the P. vaginatum genome contained significantly fewer ATT repeats than either the I. batatas or C. lanatus genome.

    Notes: Summary A time-dependent coupled-cluster approach may be employed to describe dynamic processes of many-electron systems. Atomic properties, such as the frequency-dependent polarizability, can be treated as a response of the system described by the coupled-cluster expansion to an external radiation field. The major difficulty in the realization of such a formalism is to deal with dynamic pair functions. The procedure reported here is to simplify the full set of single- and pair-excitation expansion equations to a subset of equations which includes polarization and relaxation effects to all orders and is solved by using a complete set of discrete basis functions. Calculations of excitation energies and frequency-dependent electric dipole polarizabilities for helium are presented. Application of the procedure to calculate photoionization cross sections is discussed.

    Notes: Summary Nineteen single-copy clones isolated from a PstI genomic library (cv. Maiden Plantain), and eight Vigna chloroplast DNA clones were used to probe total genomic DNA digests of 57 genotypes of Musa from India. The 19 genomic clones detected a total of 107 polymorphisms among the 57 genotypes. Principal coordinates and phenetic analyses of these data placed cultivars and species into distinct groups that were in general agreement with a previously published RAPD-based classification of these same plant materials. The 107 polymorphisms were sufficient to differentiate each clone from every other clone. Heterologous Vigna chloroplast DNA probes were used to characterize the cytoplasm of Musa cultivars and species. PCO analysis of these RFLPs were detected both within and between the generally recognized genome groups, indicating multiple hybridization pathways in the origin of hybrid clones. Data presented demonstrate that RFLPs are sufficiently abundant to classify Musa germplasm and that genetic relationships among Musa cultivars, based upon RFLP data, are in general agreement with relationships determined by analysis of morphology and RAPDs.
