    Notes: Abstract The culture fluorescence of Alcaligenes eutrophus JMP 134 was determined on-line by an Ingold Fluorosensor™ and correlated to the intracellular concentrations of reduced nicotinamide adenine dinucleotide (phosphate). The data were obtained from aerobic cultures of the strain growing chemostatically on phenol, phenol+sodium formate and fructose, as well as from aerobic/anaerobic transitions and substrate pulse experiments. The total culture fluorescence was corrected to take into account the inner filter effect of cells. Upon analysing the intracellular concentration of the dinucleotides using HPLC, it became evident that both NADH and NADPH contribute significantly to the fluorescence signal. A linear relationship between the sum of NAD(P)H and the net culture fluorescence was obtained from these data with a correlation factor of r=0.82. These investigations indicate that the measurement of culture fluorescence is a practicable tool for monitoring the redox state of a cellular culture, provided the total fluorescence signal is adjusted and the investigations are supported by direct measurements of intracellular levels of reduced dinucleotides.

    Notes: Synopsis Solid lipid nanoparticles (SLN) have been introduced as a novel carrier system for drugs and cosmetics. It has been found that SLN possess characteristics of physical UV-blockers on their own, thus offering the possibility of developing a more effective sunscreen system with reduced side-effects. Incorporation of the chemical sunscreen tocopherol acetate into SLN prevents chemical degradation and increases the UV-blocking capacity. Aqueous SLN dispersions were produced and incorporated into gels, followed by particle size examination, stability testing upon storage and thermoanalytical examination. Investigation of the UV-blocking capacity using different in vitro techniques revealed that the SLN dispersions produced in this study are at least twice as effective as their reference emulsions (conventional emulsions with identical lipid content). Placebo SLN even show greater UV-blocking efficacy than emulsions containing tocopherol acetate as the molecular sunscreen. Incorporation of tocopherol acetate into SLN leads to an overadditive UV-blocking effect. Furthermore, film formation of SLN on the skin and occlusivity were examined. The obtained data show that incorporation of tocopherol acetate into SLN leads to an improved sunscreen and skin care formulation.

    Notes: Abstract The Flow Visualization Gun (FVG), a novel time line visualization technique, has been used to investigate the flow field of a helicopter rotor with swept back tip shape in hover flight condition. After introducing the FVG-technique, the paper presents some visualization photographs of the rotor blade tip vortices and the rotor downwash. Using orthogonal sets of flow photographs and digital image analysis, the 3-dimensional time line displacement within the flow and the tip vortex structure are determined. The data of 16 time line experiments are interpolated in space and time to obtain velocity data on an evenly spaced 3-dimensional grid. Vorticity contour plots of the flow field show the complex arrangement of the tip vortices of the blade itself and of the preceding blade and some additional vorticity in the direct wake which may form a secondary vortex. Understanding and control of this vorticity distribution is important for the design of new efficient tip shapes. While the FVG technique has been used so far for qualitative investigation of complex flow patterns at local velocities of up to 20 m/s, a good comparison to laser velocimetry data validates the technique as an interesting tool for both qualitative and quantitative investigation.

    Notes: Abstract  Alcaligenes eutrophus JMP 134 was continuously (carbon-source-limited) grown on phenol to determine the maximum growth rates (μmax) as a function of the phenol assimilation pathways expressed. During growth on phenol as the sole source of carbon and energy, an almost exclusive expression of the ortho cleavage pathway (catechol 1,2-dioxygenase) was observed at initially low growth rates. This allowed a μmax of 0.28 h-1. The induction of the meta cleavage pathway (catechol 2,3-dioxygenase), which appeared at around 0.25 h-1, resulted in a further increase in the growth rate to 0.40 h-1 after the enzyme activities of this pathway had been correspondingly expressed. Hence, two maximum growth rates, one for the ortho and one for the meta cleavage pathway, exist for the growth of A. eutrophus JMP 134 on phenol. Growth on phenol was stimulated by formate, which served as an auxiliary energy source in this strain. The simultaneous utilization of phenol and formate at a molar ratio of 1:5.2 resulted in an increase of the yield coefficient from about 0.75 g dry mass/g phenol to 1.25 g/g. Furthermore, formate exerted a pronounced effect on the growth rate. At a molar ratio of phenol to formate of 1:4.2, the growth rate was increased to 0.42 h-1, despite the exclusive induction of the ortho cleavage pathway. The meta cleavage pathway was expressed during growth on this substrate mixture at about 0.4 h-1. However, this did not enable a significant increase of the growth rate beyond 0.4 h-1. This is attributed to an exhaustion of the capacity for formate oxidation at this rate. The results are discussed with respect to energy production capabilities when phenol is assimilated as an energy-deficient heterotrophic substrate.

    Description / Table of Contents: Abstract With AWPS (Amplitude Weighted Phase Structuration), a new signal processing scheme is demonstrated for the simultaneous determination of zeta potentials and particle sizes. It allows the measurements of a small electrophoretic mobility in the presence of large particle diffusion and constant velocity, e. g. due to thermal convection. Laser light scattering techniques instead of the former methods determine electrophoretic velocity more objectively and precisely. The applicability of laser measurement techniques by analysis of the frequency spectrum is limited for particles ≪50 nm or very low potentials, because of the broadening of the spectral peak by Brownian motion. In contrast to AWPS a separation of the various kinds of collective motion is not possible. The presented results demonstrate that this separation is of considerable significance in the acquisition of reliable values. Additionally the novel signal processing scheme allows a significant increase in sensitivity and therefore the application of an oscillating field (50–100 Hz) with a very small field strength. The system is feasible for particle sizes in the range of a few nm up to several μm. Its high resolution allows experiments with low fields or with small zeta potentials, even in the critical particles size range of a few nanometers.
    Notes: Zusammenfassung Mit AWPS (Amplitudengewichteter Phasenstrukturation) wird ein neues Signalverarbeitungssystem zur gleichzeitigen Bestimmung von Zetapotential und Partikelgröße vorgestellt. Es erlaubt die Messung geringer elektrophoretischer Beweglichkeiten in Gegenwart einer starken Diffusion der Partikel sowie einer überlagerten konstanten Geschwindigkeit, z. B. aufgrund eines Konvektionssthermes. Laserstreulichtverfahren anstatt der bisherigen Methoden bestimmen die elektrophoretische Geschwindigkeit objektiver und präziser. Die Anwendbarkeit der Lasertechniken, die über die Analyse des Frequenzspektrums arbeiten, ist begrenzt für Partikel ≪s50 nm oder sehr niedrige Potentiale, aufgrund der Verbreiterung des spektralen Peaks durch die Diffusionsbewegung. Im Gegensatz zu AWPS ist eine Auftrennung in die verschiedenen Komponenten der kollektiven Bewegung nicht möglich. Die vorgelegten Ergebnisse belegen, daß diese Auftrennung von beträchtlicher Bedeutung ist, um verläßliche Werte zu erhalten. Das neue Signalverarbeitungssystem ermöglicht zusätzlich einen bedeutenden Zuwachs an Empfindlichkeit und daher die Anwendung eines oszillierenden Feldes (50–100 Hz) mit sehr geringer Feldstärke. Das System ist anwendbar für Partikelgrößen im Bereich von wenigen nm bis zu einigen μm. Seine hohe Auflösung ermöglicht Experimente mit kleinen Feldern oder mit niedrigen Zetapotentialen, besonders im kritischen Teilchengrößenbereich von einigen Nanometern.

    Notes: Abstract Polyalkylcyanoacrylate nanoparticles were labelled with the fluorescent marker propidium iodide to follow their cellular distribution. Fixation of propidium iodide onto the nanoparticles can be achieved by its addition to the polymerization medium after initiating the polymerization process. However, the charged propidium iodide was found to distort the anionic polymerization process. Therefore, polymerization conditions were optimized with regard to the time of marker addition and marker concentration, loading capacity, and monodispersity of the nanoparticles size distribution.
