    Notes: Abstract Acid phosphatase was localized by light and electron microscopy in chains of vacuoles in hyphal tip cells of Whetzelinia sclerotiorum. The Enzyme was present in these vacuoles whether or not conditions favored extracellular acid phosphatase secretion. Apical vesicles, microbodies, woronin bodies, and lipid bodies did not contain acid phosphatase. The implications regarding terminology of organelles in filamentous fungi are discussed with special reference to the fungal spherosome concept.

    Notes: A basic requirement of all photosynthetic organisms is a balance between overall energy supply through temperature-independent photochemical reactions and energy consumption through the temperature-dependent biochemical reactions of photosynthetic electron transport and contiguous metabolic pathways. Since the turnover of photosystem II (PSII) reaction centers is a limiting step in the conversion of light energy into ATP and NADPH, any energy imbalance may be sensed through modulation of the redox state of PSII. This can be estimated in vivo by chlorophyll a fluorescence as changes in the redox state of PSII, or photosystem II excitation pressure, which reflects changes in the redox poise of intersystem electron transport carriers. Through comparisons of photosynthetic adjustment, we show that growth at low temperature mimics growth at high light. We conclude that terrestrial plants, green algae and cyanobacteria do not respond to changes in growth temperature or growth irradiance per se, but rather, respond to changes in the redox state of intersystem electron transport as reflected by changes in PSII excitation pressure, We suggest that this chloroplastic redox sensing mechanism may be an important component for sensing abiotic stresses in general. Thus, in addition to its role in energy transduction, the chloroplast may also be considered a primary sensor of environmental change through a redox sensing/signalling mechanism that acts synergistically with other signal transduction pathways to elicit the appropriate molecular and physiological responses.

    Notes: Summary The organization of the mitochondrial genomes from two morphologically similar Phytophthora isolates, P. megasperma f. sp. glycinea (Pmg) and P. megasperma f. sp. medicaginis (Pmm), and the morphologically different species, P. parasitica var. nicotianae (Ppn), has been studied. The mtDNAs are circular, and their estimated sizes are 45.3 kb, 41 kb, and 39.5 kb for Pmg, Pmm, and Ppn, respectively. Physical maps were constructed for restriction endonuclease sites. Four genes (l-rRNA, s-rRNA, oxi-2, and cob) were found to have the same order in the three mtDNAs.

    Notes: Summary Microbodies were observed in the hyphal tips of all 14 fungi investigated. Their morphology varied among the fungi and their numbers were influenced by the growth medium. Microbodies were closely associated with mitochondria in one fungus and with the endoplasmic reticulum in several fungi. Catalase was not detected in microbodies with the diaminobenzidine cytochemical procedure even though catalase activity was present in extracts of these fungi. The activities of the glyoxylate-cycle enzymes isocitrate lyase and malate synthase were affected by the growth medium and were particulate in the two fungi studied by differential centrifugation. Microbodies are abundant, and they are ubiquitous among the fungi and in some cases they may contain glyoxylate-cycle enzymes.

    Notes: Summary The 3′-terminal 1221 nucleotides of a Zimbabwe isolate of cowpea aphid-borne mosaic potyvirus (CABMV) genome have been sequenced. The sequence comprises an open reading frame (ORF) of 990 nucleotides and a 3′ non-coding-region of 231 nucleotides followed by a poly-A. The ORF has high similarity to NIb and coat proteins (CP) of potyviruses. A potential CP Q/S cleavage site was identified, yielding a CP of 30.5 kDa containing 275 amino acids. The CABMV sequence is closely related to that of South African passiflora virus (SAPV) which should therefore be regarded as a strain of CABMV.

    Notes: Summary The distribution of glyoxylate-cycle enzymes between microbodies and mitochondria was examined in ethanol-grown Aspergillus tamarii Kita. Particulate activities of catalase and the two glyoxylate by-pass enzymes, malate synthase and isocitrate lyase, were localized in the microbodies. The microbodies had a buoyant density of about 1.23 g cm-3 after isopycnic centrifugation in linear sucrose gradients. Particulate activities of the other two glyoxycitrate synthase, together with that of succinate dehydrogenase were restricted to the mitochondria, which had a buoyant density of about 1.20 g cm-3. Catalase also appeared to be localized in a second particle, perhaps the microbody inclusions or the Woronin bodies, having a buoyant density of about 1.26 g cm-3.

    Notes: Summary Fifty-four land races of dry beans (Phaseolus vulgaris), indigenous to areas of Brazil where anthracnose (caused by Colletotrichum lindemuthianum) is a common problem, were evaluated in field nurseries for partial resistance to race Brazilian 1 (B1) of C. lindemuthianum using symptom severity classes (SSC) from 0 to 6. Plants were selected if symptoms were present and the SSC was less than the 95% confidence interval of the mean SSC of the susceptible cultivar Carioca. S1 progeny from selected plants were evaluated in air-conditioned chambers for partial resistance to races B1, delta, and kappa of C. lindemuthianum. Of 246 S1 families evaluated, 145 families were partially resistant to one or two of the races [symptoms present, but S1 family mean significantly (p〈0.05) less than the mean of Carioca] and susceptible to the third. Six families were partially resistant to all three races. The remaining families were either susceptible or segregated for reaction to race B1. Partial resistance to C. lindemuthianum showed race specificity in the air-conditioned chambers and field nurseries.

    Notes: Summary Four cytologically distinct zones defined by the type of vacuoles and vesicles present can be distinguished in young hyphae ofPythium paroecandrum. Acid phosphatase is not associated with the vesicles and the fibrillar vacuoles present in the first and second zones from the hyphal apex but is localized in the vacuoles with electron translucent contents in the third and fourth zones. Microbodies and mitochondria do not contain acid phosphatase. The association of acid phosphatase with vacuoles in fungi of various taxonomic groups is discussed.
