    Notes: Abstract A thermotolerant methylotrophicBacillus sp. (KISRI TM1A, NCIMB 40040), isolated from the Kuwaiti environment and belonging to the group II spore-forming, bacilli, could not be correlated with any knownBacillus sp. It may, therefore, be a new species. It grew at temperatures from 37° to 58°C from pH 6.5 to 9.0 and on methanol up to 40 g l−1. It grew well in a chemostat. Its biomass yield coefficient was improved by about 30% by optimization of medium and growth conditions, reaching a maximum of 0.44g g−1 at 45°C pH 6.8 to 7.0, dilution rate 0.25 h−1 with methanol at 10 g l−1. Average crude protein and amino acid content were 84% and 60%, respectively, and maximum productivity attained under laboratory conditions was 5.06 g l−1h−1. It was concluded that this strain has good potential for use in single-cell protein production.

    Notes: Abstract A proprietary bacterial strain (Pet 1006) produced biosurfactants when grown on both glucose and an immiscible hydrocarbon as carbon sources. Pilot-plant-scale (1500 I) production gave, on repeated batch runs, 2 tonnes of culture broth containing active biosurfactant. The product was used as a substitute for chemical surfactants in a clean-up demonstration test carried out by Cargo Fleet Chemical Company Ltd. (UK) on an oil storage tank belonging to Kuwait Oil Company, Kuwait. The clean-up was successful in removing the sludge from the tank bottom, and it also allowed the recovery of more than 90% of the hydrocarbon trapped in the sludge. The recovered hydrocarbon had excellent properties and could be sold after being blended with fresh crude.

    Notes: Abstract The amphiphilic nature of 3, 6-bisdimethylaminoacridine (trivial name, acridine orange) can be used to characterize (polar/nonpolar) interfaces. This paper studies the properties of acridine orange and its dodecyl derivative at a solvent interface by internal reflection and tension determinations. In the case of absorbance measurements of crystallized acridine orange, the potenz value of the conventional acid dissociation constant pK a was found to be 9.6. A clear isosbestic point could be obtained after purification at a unique pH for different concentrations. In contrast, in the absorbance measurements of acridine orange dodecyl, no isosbestic point was observed; this was attributed to the free long dodecyl chain. The overlap in the absorbance spectrum and the normalized fluorescence spectrum was found in many solvents around 500 nm. Neutral species were more likely to transfer into organic phase upon shaking. For fluorescence measurements both steady-state and time-resolved spectra were investigated. Results showed that at the interface a bathochromic shift happened to the maxima. This means that chromophore species aggregated at the interface as the dimer or trimer or the macro oligomer. Fluorescence polarization in bulk was calculated and found to be ca. 0.4. For interfacial measurements a dynamic volume method was applied to obtain the interfacial tension. An extrapolated Gibbs plot was obtained. Increasing bulk concentration increased the concentration at the interface to a finite value representing saturation.

    Notes: The photosynthetic growth action spectrum of a green alga at three bands of visible light (blue, orange, and red) at fixed quanta input and under light-limiting conditions was measured in a batch cultivation system. Quantum efficiencies (biomass dry weight increment per quanta absorbed) were better in the yellow-red region than in the blue region. Results served as a basis for the design and optimization of a dye system that would shift the energy of solar radiation to the required wavelength range by absorbing ultraviolet to blue radiation and emitting in the yellow-red, thus enhancing algae growth. Direct incorporation of dyes into the growth medium, although theoretically expected to enhance growth, in fact resulted in dye decomposition, toxicity to algae and consequently in growth inhibition. Indirect application of dyes in a double tubular reactor (algae inside and dye solution outside) demonstrated growth enhancement for certain dyes with high quantum yields and stability, which had suitable absorption/emission spectra for artificial light sources used. The maximum indirect growth enhancement was obtained using rhodamine 6G at a concentration of 3×10-5M with tungsten filament lamp sources.

    Notes: The rationale behind KISR's involvement in SCP is explained and emphasis towards the use of aerobic thermophilic/thermotolerant methanol-utilizing bacterial cultures is stressed. An attempt is made to correlate different pilot plant harvesting procedures for biomass recovery with product quality. The latter is expressed as crude and true protein content, moisture and ash content, mineral composition of the ash, and amino acids profiles. The most suitable harvesting procedures were found to be those using a mild heat-acid or acid-heat treatment. Both of these procedures gave high recovery efficiencies and suitable product nutrition.
