    Notes: Summary We review here the present knowledge about the pathway of import and assembly of porin into mitochondria and compare it to those of other mitochondrial proteins. Porin, like all outer mitochondrial membrane proteins studied so far is made as a precursor without a cleavble ‘signal’ sequence; thus targeting information must reside in the mature sequence. At least part of this information appears to be located at the amino-terminal end of the molecule. Transport into mitochondria can occur post-translationally. In a first step, the porin precursor is specifically recognized on the mitochondrial surface by a protease sensitive receptor. In a second step, porin precursor inserts partially into the outer membrane. This step is mediated by a component of the import machinery common to the import pathways of precursor proteins destined for other mitochondrial subcompartments. Finally, porin is assembled to produce the functional oligomeric form of an integral membrane protein wich is characterized by its extreme protease resistance.

    Notes: Abstract Various adenosine triphosphate (ATP)-dependent proteases were identified within mitochondria which mediate selective mitochondrial protein degradation and fulfill crucial functions in mitochondrial biogenesis. The matrix-localized PIM1 protease, a homologue of theEscherichia coli Lon protease, is required for respiration and maintenance of mitochondrial genome integrity. Degradation of non-native polypeptides by PIM1 protease depends on the chaperone activity of the mitochondrial Hsp70 system, posing intriguing questions about the relation between the proteolytic system and the folding machinery in mitochondria. The mitochondrial inner membrane harbors two ATP-dependent metallopeptidases, them- and thei-AAA protease, which expose their catalytic sites to opposite membrane surfaces and cooperate in the degradation of inner membrane proteins. In addition to its proteolytic activity, them-AAA protease has chaperone-like activity during the assembly of respiratory and ATP-synthase complexes. It constitutes a quality control system in the inner membrane for membrane-embedded protein complexes.

    Notes: Abstract To obtain data on the biochemical effects of edeine in the fungus Neurospora crassa, in vivo protein synthesis, in vitro protein synthesis, as well as in vivo RNA and DNA synthesis of the wildtype and an edeine resistant mutant were measured.—Incorporation of 3H leucine into conidia of both strains, which served as a measure for in vivo protein synthesis, was inhibited by 200 μg edeine/ml as follows: Wildtype approx. 40%, mutant approx. 6%.—Incorporation of 14C phenylalanine into polyphenylalanine in a cell free system with ribosomes from either the wildtype or the mutant, was inhibited between 74 and 95% by edeine at a ratio of 2 molecules edeine per ribosome.—Incorporation of 3H adenosine into conidia, serving as a measure for in vivo RNA synthesis, was inhibited in the wild-type (approx. 30% inhibition by 200 μg edeine/ml). It was, however, not influenced in the ed r mutant. Similarly, in vivo DNA synthesis was decreased in the wildtype, but not in the mutant.—These results suggest that edeine acts at more than one site. The resistance of the mutant ed r -29 (ed r -2 locus) is tentatively interpreted as due to a block in edeine uptake.

    Notes: Abstract Mitochondrial biogenesis requires the efficient import of hundreds of different cytosolically translated preproteins into existing organelles. Recognition and translocation of preproteins at the mitochondrial outer membrane is achieved by the TOM complex (translocase of the outer mitochondrial membrane). The largest component of this complex is TOM70, an integral outer membrane protein with a large cytosolic domain thought to serve as a receptor for a specific group of preproteins. To investigate the functional role of TOM70 in Neurospora crassa the tom70 gene was inactivated using the natural phenomenon of repeat-induced point mutation (RIP). Mutant strains were identified that harbored RIPed tom70 alleles and contained no immunologically detectable TOM70. Strains that lack TOM70 grow more slowly than wild-type strains, conidiate poorly, and contain enlarged mitochondria. In vitro preprotein import studies using TOM70-deficient mitochondria revealed a defect in the uptake of the ADP/ATP carrier. Other preproteins tested were imported at wild-type rates with the exception of the precursor of the mitochondrial-processing peptidase (MPP) which was imported more efficiently by TOM70-deficient mitochondria. These data support the view that TOM70 plays a role as a specific receptor for carrier proteins in mitochondrial-preprotein import. The presence of tetratricopeptide repeats (TPRs) in the TOM70 sequence and the enlarged shape of mitochondria lacking TOM70 raise the possibility that the protein also plays a role in the maintenance of mitochondrial morphology.

    Notes: Purpose - This paper identifies skills necessary in order to succeed in Vietnam and proposes a training program to develop such skills. Design/methodology/approach - To determine necessary skills, 74 managers were interviewed using critical incident methodology to identify training needs. Critical incident approach asks respondents to describe the incident, its nature and consequences to provide context for understanding the managers' problems. Findings - Local Vietnamese managers cited professional development (42 percent), basic business skills (29 percent), communication and cultural skills (18 percent) and legal understanding skills (11 percent) as most critical to succeed in international business. Foreign managers cited cultural understanding and awareness (34 percent), professional skills (23 percent), personal skills (20 percent), and interpersonal skills, language and communication skills. For Vietnamese managers, training includes leadership, problem solving, interpersonal skills, business basics, communication and culture. For expatriate managers, training presents the particular aspects of the local culture in a way that allows the foreign manager to become more effective through understanding and awareness. Research limitations/implications - The findings of this study and its training suggestions are based on findings from Vietnam. To be relevant outside Vietnam, the program may be adapted. Originality/value - This paper identifies the skills needed by Vietnamese and expatriate managers to be successful in international business. The findings and suggestions are valuable to managers and trainers involved in international business in Vietnam and Asia.
