Summary Sixteen β-lactam producing actinomycete strains were screened for the presence of large linear plasmids. Among these, four strains contained linear plasmids of 12–450 kb in size. Southern blot analysis using a synthetic oligonucleotide for the isopenicillin N synthase gene and plasmid pBROC137 containing thecefD andcefE genes in the cephalosporin biosynthesis, respectively, showed no hybridization. This result suggests that these linear plasmids are not involved in the biosynthesis of β-lactam antibiotics.
