    Notes: Abstract We measured anti M protein antibody (AMPA) titres in children with idiopathic mitral regurgitation (MR), streptococcal infection, rheumatic fever (RF), post-streptococcal acute glomerulonephritis (AGN) and normal healthy children. We investigated the association of MR with streptococcal infection and whether high AMPA titres can be used as persisting evidence of previous streptococcal infection. AMPA titres were measured with an enzyme-linked immunosorbent assay. We found significantly higher antibody titres in patients with MR and in streptococcal infection, RF, and AGN than in healthy controls. In the MR group (n=15), 54% patients had AMPA titres above the 90th percentile value that was found in normal controls. An elevated AMPA titre persisted for a long period even when the anti-streptolysin O titres had declined to normal in RF patients. Our data suggest that the high AMPA titres in MR should be further investigated to clarify the probable association with previous streptococcal infection. Conclusion High AMPA titre is a risk factor for developing complications after streptococcal infection. Our serological evidence suggests that in some patients. MR may be related to previous streptococcal infection.

    Notes: Abstract Recently, a possible relationship between Kawasaki syndrome (KS) and superantigen has been discussed since the report of selective expansion of specific Vβ family in the acute phase of KS. To further investigate the relationship between KS and superantigens, we examined 25 types of T-cell receptor Vβ family repertoire in patients with KS using the reverse transcription polymerase chain reaction. This is the first attempt to examine all of 25 Vβ families in KS. A non radioisotope method was used to quantify mRNA so that the experiment was safer, simpler, and faster. An expression index (EI) for each Vβ was defined as: (the amount of each Vβ mRNA)/(the sum of all Vβ mRNA) × 100. Ten patients with KS and ten normal children were studied. The Vβ9 and Vβ15 of acute phase of KS showed both significantly lower mean EI and significantly higher frequency of a decreased EI value as compared with control children. Selective expansion of the Vβ family in the patients with KS was not observed. Although highly increased EIs were observed in various Vβs, their frequency was not statistically significant. The pattern of increased Vβs did not show the specific pattern that indicates a particular superantigen. Conclusion In Kawasaki syndrome non-radioisotope method for analysing Vβ mRNA is useful in cases where many samples have to be handled. The depletion of Vβ9 and Vβ15 or highly increased expression index in the acute phase of Kawasaki syndrome might suggest a relationship to superantigens.

    Notes: Abstract The inhibitory action of azelastine hydrochloride (Azeptin) on the respiratory burst in peripheral polymorphonuclear leukocytes (PMN) and pulmonary alveolar macrophages (PAM) has been studied. Azeptin in vitro suppressed chemiluminescence and superoxide (O 2 - ) generation by human PMN in a dose-and time-dependent manner. Phorbol myristyl acetate (PMA) and formyl-methionyl-leucyl-phenylalanine (FMLP)-induced O 2 - generation were strongly suppressed by 10−6M and 10−5M Azeptin, respectively. PMN and PAM from rabbits injected with Azeptin 0.2 mg·kg−1 for 5 days showed lower chemiluminescence and O 2 - generation than cells from untreated rabbits. Nitroblue tetrazolium reduction activity in human PMN was suppressed by treatment of PMN with 10−6M Azeptin for 6 h. Inositol trisphosphate, intracellular free calcium, and protein kinase C activity were decreased by 10−6M to 10−5M Azeptin. The tyrosine phosphorylation of many proteins, especially a 115 kDa protein, was suppressed by 10−5M Azeptin. However, superoxide dismutase activity in PMN, PAM, and lung tissue samples was only slightly decreased, even when the rabbits were treated with 1.0 mg·kg−1 Azeptin for 5 days. The results suggest that Azeptin suppresses multiple signal transduction steps in the respiratory burst of PMN. This suppressive action should be very useful in the prevention and treatment of reactive oxygen-associated disorders.

    Notes: The Fourier-expansion density matrix method is developed to describe the multiphoton processes of molecules interacting with the heat bath under the strong laser light. This method is applied to deriving expressions for the absorption spectra in the case in which a single-mode laser interacts with the molecular system of a three-level taking into account the dynamic Stark effect. Model calculations are performed to demonstrate the dynamic Stark effects on the absorption ability in the three-level system, and compared with the results of the weak laser light case. The calculated band shapes of the two-photon absorption are analyzed in terms of the simultaneous and sequential mechanisms. The sequential mechanism is characterized by bath-induced intramolecular transitions. As the laser intensity is increased the simultaneous mechanism makes a dominant contribution with increased dynamic Stark effects.

    Notes: An expression for the probability of time-resolved three-photon ionization via coherently excited resonant states of a molecular system is derived in the perturbative density matrix formalism with the aid of the Liouville space Feynman diagram. It is shown how the time evolution of the molecular coherence in the first resonant state can be detected as a function of the delay time between the pumping and probing lasers through the probing two-photon ionization process. The three-photon ionization process consists of the simultaneous and sequential processes which are classified by the Liouville space Feynman diagrams. The possibility of appearance of quantum beats in the pump–probe three-photon ionization is discussed by performing model calculations of the probability of ionization. The role of the direct and redistributed processes is discussed. The redistributed process is induced by intramolecular processes in the resonant state. The theoretical treatment is applied to the (1+2) three-photon ionization of pyrazine observed by Knee et al. It is demonstrated that the fast decay component (110 ps) of the pyrazine spectra originates from the intramolecular dephasing of the vibronic coherence created by the pumping pulse laser, and this component appears only when the redistributed process (the ionization process through triplet levels) is absent.

    Notes: Effects of the coherence transfer induced by the molecule–heat bath interactions on the ultrashort time-resolved coherent anti-Stokes Raman scattering (CARS) from molecules in liquids are theoretically studied. Based on the perturbative density matrix formalism an expression for the CARS intensity is derived taking into account the coherence transfer between the Raman active vibrational transitions of two molecules in liquids. The coherence transfer constants and dephasing constants are properly incorporated with the aid of Liouville space Feynman diagrams. The structure of the coherence transfer matrix element which expresses the time evolution of the coherence between the relevant transitions is clarified by solving the Master equation with the coherence transfer and dephasing constants in the Markoff approximation. Frequency shifts of the quantum beats appear in the time-resolved CARS as a result of the coherence transfer. A multispherical layer model is adopted in evaluating the coherence transfer effects in liquids in femtosecond time domains. Model calculations of time-resolved CARS spectra have been carried out to demonstrate the coherence transfer effects in both short and long range coherence transfer cases. It is predicted that the quantum beats are amplified in the time-resolved CARS spectra of molecules in liquids in a long range coherence transfer case when there exist differences in the coherence transfer constants between each spherical layer.

    Notes: Effects of vibration–rotation (Coriolis) couplings on the coherent polarization anisotropy are theoretically studied in a time-resolved two-photon ionization of a symmetric top molecule. This polarization anisotropy originates from a coherent excitation of the resonant rovibronic molecular eigenstates (rovibronic coherence) whose zeroth order states are mixed through the Coriolis coupling. Expressions for the time-dependent degree of polarization after the coherent excitation of the rovibronic states produced by the Coriolis coupling are derived as a function of the delay time in the pump–probe two-photon ionization. Model calculations of the time-dependent degree of polarization as well as the probabilities of the two-photon ionization are performed to demonstrate the Coriolis coupling effects in the low excess energy regions of the resonant intermediate state. It is shown that oscillatory behaviors in the time-dependent degree of polarization should be observed as a result of the creation of the rovibronic coherence. It is demonstrated that oscillations of the degree of polarization involve both contribution of the purely rotational J-coherence and that of the rovibronic coherence in the resonant manifold when the rotational branches are coherently excited and the characteristic rotation–vibration interaction energy is larger than a typical free rotational energy under jet-cooled condition.
