    Notes: Abstract  Using a battery of monoclonal antibodies specific for rat proteins, immunohistochemistry was carried out on the developing myoepithelial cells (MECs) of the rat major salivary glands. The proteins examined were α-smooth muscle actin (αSMA), h1-calponin (calponin), keratin 14 (K14), β subunit of S-100 protein (S-100β), vimentin and glial fibrillary acidic protein (GFAP). The MECs exhibited immunoreactivity for αSMA, calponin and K14, but not that for S-100β, vimentin and GFAP. Immunoreactivity for αSMA appeared in the MECs from the time when the microfilaments were initially deposited in these cells, i.e., at 20 days in utero in the sublingual and submandibular glands and at birth in the parotid gland. Calponin immunoreactivity was seen 1 day earlier than αSMA. The appearance was almost at the same time as the onset of the MEC differentiation in each gland. A small number of the MECs expressed weak K14 immunoreactivity from the time when the acinus-intercalated duct structure was established, i.e., at 21 days in utero in the sublingual gland, at 5 days after birth in the perotid gland and after 5 weeks post-natally in the submandibular gland. In addition, K14 immunoreactivity was observed in the basal cells of the striated and excretory ducts. The first appearance of K14 in these cells again coincided with the emergence of the duct system in each gland, i.e., at 20 days in utero in the sublingual gland, at 21 days in utero in the submandibular gland and at 3 days after birth in the parotid gland. Finally, the MECs in all the glands were found to redistribute as the acini matured. As the acini grew rapidly during the weaning period in the parotid and the sublingual glands, the MECs ceased to surround the acini. Thereafter, they disappeared from the acini in the parotid gland, whereas they reappeared in the sublingual gland. In the submandibular gland, the MECs were confined to the terminal tubules until 4 weeks after birth. Thereafter, the acini were established and invested by the MECs. In conclusion, immunohistochemistry of calponin and αSMA is a useful tool for identification of the MEC during its earliest differentiation, which has hitherto been possible only electron microscopically. In addition, it is suggested that the MEC is heterogeneous and the functionally differentiated MEC appears after weaning around acini of the mucous and seromucous glands.

    Notes: Abstract One of the pyrimidine compounds, 2-piperadino-6-methyl-5-oxo-5,6-dihydro(7H)pyrrolo[3,4-d]pyrimidine (MS-818), has neurotropic effects in vitro. Therefore, we studied the effect of MS-818 on the regeneration of the peroneal nerve in C57BL/6J mice after a crush injury. Two test groups, which received a daily intraperitoneal injection of 5 mg/kg or 10 mg/kg MS-818, respectively, were compared with controls, which received daily intraperitoneal injections of physiological saline, over a 14-day period. The maximum foot-width ratio (crushed side/uncrushed side) was obtained on days 1, 8 and 14 after the crush injury, and the various morphometric parameters were evaluated at both 5 and 10 mm distal to the proximal portion of the crush site. The significant effects of MS-818 included a larger maximum foot width (P〈0.04) and a greater number of unmyelinated axons per nerve at both levels (P〈0.003) in both test groups than in controls. MS-818 had no significant effects on body weight, the increase of total transverse fascicular area after the crush injury, the total number of myelinated fibers with their size distributions, or the number of nuclei of Schwann cells and macrophages. Therefore, we conclude that MS-818 promotes axonal sprouting and elongation after a crush injury in mice.

    Notes: Summary To evaluate the three-dimensional pathology of lumbar primary sensory neurons in p-bromophenylacetylurea intoxication, the number and size distribution of neurons and of myelinated fibers were evaluated at the L-6 spinal ganglion level and at proximal and distal levels of sural nerve and thoracic (proximal) and cervical (distal) levels of Goll's tract, respectively, 2 and 6 weeks after the intoxication in rats. The number and size distribution of ganglion neuron cell bodies were not significantly different between intoxicated and control rats. The distal level of sural nerve had, significantly fewer large myelinated fibers than did control, and a significantly higher frequency of fibers undergoing degeneration. Proximal levels of sural nerve showed similar, but less severe changes. Similarly, the myelinated fibers of Goll's tract were significantly more affected at cervical than at thoracic level. Therefore, by morphometric criteria both centrally and peripherally directed myelinated fibers are most affected distally and less affected proximally while neuron cell bodies are not affected at all. These three-imensional morphological changes must be taken into consideration in formulating possible mechanisms for the development of this neuropathy.

    Notes: Summary Examination of photo and electron microscopical preparations of a biopsied sural nerve from a patient with von Recklinghausen's disease is reported. In the toluidine blue stained Epon-sections the number of myelinated fibers per square millimeter was moderately decreased and the number of large myelinated fibers was severely reduced. Large, circular, lamellated structures which resemble the onion bulb were observed. These structures are different from the previously reported onion bulb in their size and core structure which contains usually a degenerated nerve fiber. Some consideration was given to the origin of these onion bulb-like lamellated structures.

Relative configuration at C-2, C-3, and C-1′ was determined as 2S,3R,1′S by the conversion from (2S,3R,1′R)-stegobinone. This new sex pheromone elicits the pheromonal response from the species.

    Notes: Abstract Stegobinone, (2S, 3R, 1′R)-2,3,-dihy dro-2,3,5-trimethyl-6-(1′-methyl-2′-oxobutyl)-4H-pyran-4-one, the sex pheromone of drugstore beetles (Stegobium paniceum L.), elicited the pheromonal response from the males of the species in our bioassay system; however, the synthesized diastereomeric mixture of this compound was actually inactive to the males. Although the stegobinone isolated from the beetles of this species had significant activity, its enantiomeric [(±)-2S,3R,1′S-] and diatereomeric [(±)-2S,3S,1′RS-] isomers were inactive. Adding the (±)-2S,3R,1′S isomer to stegobinone significantly reduced the male response. Furthermore, the activity of Stegobinone vanished on keeping it at room temperature for two weeks. In such a stored stegobinone sample, the presence of 2S,3R,1′S isomer, the inhibitory component, was confirmed. This isomer might be produced by C-1′ epimerization during storage.
