    Notes: Abstract The injection of pregnant mice with Coxsackievirus B3 results in a reduction of fetal weights and retarded development of the albumin/α1-fetoprotein ratio which can be measured by using the Laurell two-dimensional immunoelectrophoretic technique. The litter weights tended to be lower in litters with a low albumin/α1-fetoprotein ratio and from an analysis of covariance the results indicate that the effect of the virus on fetal weights can not all be accounted for by the effect on maturation. It is suggested that the mouse model system studied may be of value for demonstrating retarded fetal and neonatal maturation as a result of inapparent virus infection, and possibly from other causes.

    Notes: Summary Total thyroxine and triiodothyronine levels are often reduced in long-term phenytoin therapy, a finding at variance with the euthyroid clinical state of these subjects. We have measured free (biologically active) thyroid hormone levels in 31 patients on phenytoin therapy and have found a significant reduction in measurements of both free thyroxine and free triiodothyronine. In contrast, addition of phenytoin to normal serum in vitro resulted in an increase in free thyroxine and free triiodothyronine concentrations. The findings in vitro are consistent with displacement of thyroid hormones from thyroxine binding globulin while the reduced free hormone levels in vivo confirm that the effects of phenytoin are not confined to binding inhibition and suggest that phenytoin has induced a change in the cellular metabolism of thyroid hormones.

    Notes: Abstract: Seventeen analogues of l-methyl-4-phenyl-1,2,3,6-tetrahydropyridine were synthesized using three reaction pathways: condensation of phenols with 1-methyl-4-piperidone, reaction of Grignard reagents with 1-methyl-4-piperidone followed by dehydration of the product, and aminomethylation of olefins. The identity of the products of synthesis was established by nuclear magnetic resonance spectroscopy, mass spectroscopy. and elemental analysis. Thirteen analogues were shown to inhibit the oxidation of benzylamine by bovine plasma amine oxidase. Increasing the length of the aliphatic chain of N-substituted analogues resulted in increased inhibition. In 4-phenyl-substituted analogues, both the position and electronic character of the substituent group affected the degree of inhibition.

    Notes: The urinary excretion of 3,5-diiodotyrosine was determined in euthyroid, hypothyroid and hyperthyroid individuals using a sensitive gas chromatographic mass spectrometric assay. This involved a multi-step extraction of the amino acid from urine (mean efficiency 28±5.7%) then conversion to the N,O -diheptafluorobutyryl methyl ester. The fragmentation of the derivative is discussed. Although the mean excretions in the two pathological states were significantly different from that of euthyroid individuals (P〈0.01 in both instances) there was considerable overlap with the normal range.

    Notes: The mass spectral properties of four classes of derivatives of thyronine are discussed (oxazolidinone, O-methyl oxazolidinone, O-acetyl oxazolidinone and N,O-diheptafluorobutyryl methyl ester). An assay for thyronine in human urine is described based on the N,O-diheptafluorobutyryl methyl ester. Results of To excretion in euthyroid humans were compared with those obtained previously using an assay based on the O-acetyl oxazolidinone derivative. Patients with frank hyperthyroidism had significantly higher To excretion than euthyroid subjects (2α 〈 0.002) and hypothyroid patients lower To excretions than enthyroid subjects (2α 〈 0.002). Some overlap between the two pathological ranges and the normal euthyroid range was evident.

    Notes: The components of a gas chromatographic mass spectrometric-selected ion monitoring (SIM) assay for thyroxine (T4) in human serum are described. The internal standard for the assay was synthesized from deuterium-labelled 3,5-diiodotyrosine and 3,5-diiodo-4-hydroxyphenylpyruvic acid. A novel method was developed for isolating the products of the coupling reaction. The results obtained by gas chromatography mass spectrometry SIM were compared with those of radioimmunoassay. The gas chromatographic mass spectrometric SIM assay would form the basis of a reference assay for T4.
