    Notes: Summary A highly cellulolytic Cellulomonas mutant, CS1-17, has been shown to be improved over the original parent strain, CS1-1, with respect to xylanase and β-xylosidase activities. From induction studies during growth on xylan, crystalline cellulose and carboxymethylcellulose it can be deduced that, although both activities have been similarly affected by the mutation, xylanolytic activity is distinct from cellulolytic activity; however, the possibility of some cross-specificity has not been eliminated.

    Notes: Summary A Cellulomonas mutant previously shown to be improved over its parent with respect to cellulolytic and xylanolytic activities, has now been shown to produce enhanced activity towards galactomannan. The mutant produced a range of glycosidase activities during growth on various carbohydrate substrates. Changes in the proportions of β-glucosidase, β-mannosidase, β-xylosidase and α-arabinosidase levels with variation in growth substrate indicate specific produciton patterns for these four enzymic activities.

    Notes: Candida utilis was grown with glucose as growth-limiting carbon source in batch culture, steady-state continuous culture, and non-steady-state continuous culture. High cytochrome concentrations were routinely recorded in cells harvested in the latter stages of batch culture. They were occasionally recorded in cells from imprecisely controlled steady-state cultures, but precise control of the steady-state dissolved oxygen tension stabilized the cytochromes at relatively low levels. Controlled non-steady-state continuous cultures, imposed by pulse additions of ethanol, routinely produced cells with high cytochrome concentrations. A mechanism is proposed whereby maintenance of cytochrome derepression in continuous culture is dependent upon indefinitely prolonging an “overshoot” response in gene expression.

    Notes: (1) Samples of yeast which varied over an arbitrary 0-100% scale with respect, to cytochrome concentration were prepared. (2) The reflectance spectrum of each sample was scanned. (3) These reflectance spectra were analyzed numerically on a digital computer to determine the bandwidths and intensities as a function of cytochrome concentration. Band shapes using the profiles of Cauchy, Gauss, and Giovanlelli were used. The correlations may be used to estimate cytochrome concentrations with a standard deviation of about 3%.
