    Notes: A size-selected library constructed from DNA of the whiting Merlangius merlangus was screened. From about 3200 recombinant clones, 43 microsatellite loci were detected. Thirteen were sequenced in full. Primers were designed from the sequence of the flanking regions for six loci and used to test the allelic variability at these loci using the polymerase chain reaction (PCR). In addition, five primer pairs developed for the stickleback and another seven for cod were tested. Only six primer pairs revealed at least three alleles per locus. The three useful loci Gmo2, Mmer-UEAW01 and Mmer-UEAW02, had 14–23 alleles per locus in 370 samples. Estimates of genetic structure (φ) were not statistically significant. However, estimates of genetic differentiation (Fst) were significantly different from zero. Heterogeneity χ2-analysis of allele frequencies among populations suggested relatively low levels of differentiation among samples. Significantly different allele frequency distributions were found for Borgensfjord and northern and southern North Sea samples for at least one locus, and between the latter samples for Mmer-UEAW02 and Gmo2. There were significant excesses of homozygotes in all samples, over expectation for randomly mating populations in Hardy-Weinberg equilibrium. The estimated frequencies of null alleles were 14.3%, for Mmer-UEAW01, 10.2% for Mmer-UEAW02 and 11.6% for Gmo2. This result calls for a careful interpretation of the significance of these microsatellite data.

    Notes: We analysed a random sample of 10 three-spined stickleback nests by DNA fingerprinting. DNA from the guardian male and a random subsample of LO fry per nest were probed with pYNZ 132, a human single-locus probe for VNTR-loci (variable number of tandem repeats). On average this probe produced DNA fingerprints of 12 scoreable bands. By comparing the bands present in each individual, we calculated band sharing indices (BSIs) between the guardian male and its fry. The BSls varied between 0.40 and 0.77 with an average (± S.D.) of 0.59 ± 0.09. We therefore conclude that the guardian male was the true father of the fry in all these nests. Once the paternal bands in each fry were determined, we compared the maternal bands among the fry of each nest. Based on the BSIs obtained with these comparisons, we found that one guardian male enticed three females to spawn in its nest, six enticed two females and three enticed one female.

    Notes: Abstract A method for the determination of mercury in solid samples using laser ablation coupled with atomic fluorescence spectroscopy has been developed. An Nd-YAG laser was used for ablation and the vaporised and atomised material was rapidly led to an atomic fluorescence detector, where excitation and emission took place. The experimental approach was applied to the assessment of different procedures as sensitive as possible for implementing standard addition methods. Calibration curves were recorded using the prepared standards, which exhibited linear ranges between 0.5–100 μg/g, with excellent regression coefficients in all instances (0.9907). The precision, expressed as RSD %, was 3 and 4% for contents of 1 and 30 μg/g, respectively, in the same pellet; and 7 and 12% for the same contents and different type of pellets. The method has been applied to the determination of mercury in CRM of sewage sediment and a sludge sample with a known amount of mercury determined by an interlaboratory study. The results obtained show good agreement with those expected.

    Notes: Summary After 2 h intraocular injections of 19 and 190 nmoles ibotenic acid in the rat retina produced an intensive vacuolization of the inner plexiform layer and cellular alterations, in the inner nuclear layer and ganglion cell layer. These alterations consisted of either cytoplasmic swelling accompanied by clumping of the nuclear chromatin or darkening of the cytoplasm along with nuclear condensation. A week later the retinas were thinner than the controls due to the disappearance of the affected cells. Pre-treatment with diazepam prevents the morphological alterations induced by 19 nmoles ibotenic acid; mainly the swelling, which was completely prevented, while the darkening was reduced drastically, although some vacuolization of the inner plexiform layer is still present.
