    Notes: Abstract Biochemical modulation is one of the most interesting fields in cancer chemotherapy. Interferon-α (IFNα) is a cytokine that is able to influence the pharmacodynamics of 5-fluorouracil (5FU) through a number of mechanisms. With the aim of confirming some data emerging from the literature, we initiated a multicentric randomized study comparing the combination of 5FU and IFNα-2a with 5FU alone in the treatment of advanced or metastatic colon cancer. A group of 205 colon cancer patients (104 in the 5FU arm and 101 in the 5FU+IFNα-2a arm) were included in the final intention-to-treat analysis. Rectal cancers were not considered eligible. All patients had measurable disease, were aged 75 years or less, had a Karnofsky index of at least 60 and had good bone marrow, renal, liver and cardiac functions. No previous chemo-immunotherapy was allowed. The treatment was 750 mg/m2 5FU (4 h i.v. infusion) on days 1–5 and then i.v. bolus weekly, starting from day 12, with or without IFNα-2a given s.c. three times weekly (starting dose 3 × 106 IU rising to 9 × 106 IU, if tolerated). Patients were treated until progression or, if responsive, for a maximum of 48 weeks and then observed for a period of 2 years. The primary end-point of the study was objective clinical response (OR); secondary parameters were time to progression, overall survival, and time to death after progression. WHO criteria were used for both clinical response and toxicity measurements. Dose reduction was planned a priori in the event of significant toxicity due to 5FU, IFNα-2a or both. Association between primary and secondary end-points and treatment was studied by univariate and multivariate analysis. Altogether, 47 patients achieved a documented response. A 25% OR was observed in the combination arm while a 21% OR was seen in the 5FU arm; this difference is not statistically significant (P=0.6). Patients with a small tumour burden (below 5 cm2) showed a higher probability of response in both arms. Patients in the experimental arm had a higher but not statistically significant cumulative progression-free probability. Median survival was 47.1 weeks overall, while it was 43.7 and 48.5 weeks in the control and experimental arms, respectively. The combination was clearly more toxic than 5FU alone, leukopenia being the most frequent side-effect in the experimental arm and nausea and vomiting in the control arm. In conclusion these results are quite disappointing and 5FU + IFNα-2a can not be considered a standard treatment for advanced colon cancer.

    Notes: Summary The behavior of Protozoa can be studied by using the ethogram as a conceptual tool capable of giving an almost complete picture of the motor biology of these microorganisms. A new semi-automatic, computerized method for drawing ethograms is described here: it allows a time-saving of about 70%, in comparison with the similar manual procedure. Microorganism movements are photographed by a Pentax LX camera from the screen of a TV monitor, connected to the stereomicroscope by a TV camera, and resolved into single images using a stroboscopic apparatus. The pictorial data are introduced into the computer by means of a digitizing tablet, and the track analysis is performed semi-automatically. The measurements recorded are then processed using a commercial statistics package in order to obtain a general view of the quantitative parameters of each ethogram.

Duodenal and jejunal pressures and duodeno-caecal transit were measured in eight healthy male volunteers during euglycaemia (blood glucose 4–6 mmol/l) and hyperglycaemia (blood glucose 12–15 mmol/l). Duodenal and jejunal pressures were recorded with a manometric assembly during intraduodenal infusion of 100 ml nutrient liquid comprising 14 % protein, 31.5 % fat and 54.5 % carbohydrate together with 15 g lactulose. Duodeno-caecal transit was determined by a breath hydrogen technique. The number of duodenal (p 〈 0.05) and jejunal (p 〈 0.01) pressure waves, excluding phase III episodes was reduced during hyperglycaemia compared to euglycaemia. Hyperglycaemia was associated with earlier onset of phase III activity (30 ± 12 vs 132 ± 20 min; p 〈 0.05). Duodeno-caecal transit was slower during hyperglycaemia when compared to euglycaemia (114 ± 17 vs 49 ± 6 min, p 〈 0.01). We conclude that induced hyperglycaemia has major effects on postprandial small intestinal motility. The reduction in duodenal and jejunal motor activity is likely to explain the retardation of small intestinal transit during hyperglycaemia. [Diabetologia (1996) 39: 984–989]

    Notes: : Oxidative modification of low-density lipoprotein (LDL) appears to play a pivotal role in atherosclerosis. In the present study the effect of several concentrations of rutin, its aglycon quercetin, cyanidin 3-O-β-d-glucoside, its aglycon cyanidin, hydroxycinnamic acids, and a standardized red orange (Citrus sinensis varieties) extract (ROE) on LDL oxidation was tested. Lipid peroxidation was monitored by conjugated diene formation and by different electrophoretic mobility of native LDL and oxidized LDL. Results obtained in the present study suggest specific effects of flavonoids tested and, in particular, of ROE on the prevention of LDL oxidation involved in the development of cardiovascular diseases.

    Notes: : The major natural polyphenols of wine are flavonoids. Red wine also contains a natural phytoalexin called resveratrol which recently has been the subject of conflicting reports regarding its protective role against cardiovascular diseases. We have investigated the free radical scavenging capacity of resveratrol, its effects on xanthine oxidase (XO) activity, spontaneous membrane lipid oxidation, and DNA cleavage. Resveratrol showed a dose-dependent free radical scavenging activity, significant inhibition of XO activity, an anti-lipoperoxidative capacity, and a protective effect on DNA cleavage. The antioxidant capacity of resveratrol is ascribed to the concomitant activities of scavenging free radicals, metal chelating, and inhibition of some enzymes involved in free radical generation.

    Notes: : Red oranges are characterized by high levels of anthocyanins, with a high content of other antioxidant compounds such as flavones, hydroxycinnamic acids, and ascorbic acid. In this work, we investigated effects of standardized extract from red oranges on DNA cleavage and its free radical scavenging capacity. In addition, its effect on xanthine oxidase activity and lipid oxidation in rat-liver microsomes was evaluated. The extract showed a protective effect on DNA cleavage and a dose-dependent free radical scavenging capacity. These results were confirmed by significant inhibition of xanthine oxidase activity and an antilipoperoxidative capacity. The evidence suggests that, because of its antioxidant activity, the extract may offer excellent photoprotection for skin and may be useful in treatment of diseases where oxidative stress plays a role.
