    Notes: SUMMARY— Alter extraction and separation by thin-layer chromatography (TLC) on cellulose, nine flavonol glycosides were detected and purified in the fruit of strawberry from various sources. The structures of these were investigated by TLC, spectral properties and several types of hydrolysis. Four of them mere completely identified as the -3-ß-monoglucosides and -3-ß-glucuronides of quercetin and kaempferol. Three others were characterized less fully as kaempferol-7-monogluco-side, a kaempferol-7-glucoside and an unreported quercetin-3-glucoside which was not the -3-ß-monoglucoside or a diglucoside. A comparison between the flavonol glycosides and anthocyanins of this fruit was made with respect to the amounts and types present.

    Notes: Murine splenocytes which contained B cells activated by in vivo exposure to affinity-purified goat anti-mouse IgD (GaMD) antibody were utilized to present major histocompatibility complex (MHC) and non-MHC minor lymphocyte-stimulating (Mlsa) determinants in a primary mixed lymphocyte reaction (MLR). As the time in hours after in vivo exposure to GaMD increased, splenocytes from adult mice showed a co-ordinate increase in cell size, expression of public and private MHC class II antigenic determinants and MHC and Mlsa antigen-presenting capacity. This augmented alloantigen-presenting capacity was demonstrable with either irradiated or mitomycin C-treated adult splenocytes. In contrast, GaMD-treated neonatal splenocytes from 10-day-old mice demonstrated no significantly increased class II expression or enhanced MHC stimulatory capacity, but nevertheless triggered augmented responder cell proliferation across an Mlsa barrier. Thus, increased class II expression or presenting capacity may not be required for an augmentation in splenocyte Mls-stimulating ability to occur. In vitro exposure of T cell-depleted splenocytes or highly purified small resting B cells to GaMD or lipopolysaccharide (LPS) induced a substantially increased ability in those populations to present MHC and Mlsa antigens in a primary MLR. Hence in vivo or in vitro activation of B lymphocytes in a stimulator cell population may yield more effective presentation of MHC and non-MHC determinants.

    Notes: Summary: Homeostatic mechanisms regulating mast cell numbers and function in peripheral tissues have largely focused on cytokines, such as stem cell factor, interleukin (IL)-3, IL-4, and IL-10, which regulate mast cell maintenance and proliferation. Despite these advances, little attention has been paid to the mechanisms that mediate mature mast cell turnover, and control of mast cell hyperplasia generated during Th2-mediated responses. These are important issues, as mast cells are now known to be multi-functional effector cells, that have the capacity to mediate both innate and Th2-induced immune responses. Numerous secretagogues may elicit mast cells to release a large number of important mediators that can cause chronic inflammation. Therefore, how mast cell homeostasis is regulated may have significant effects on normal physiology, and contribute to the genesis of inflammatory disease. Our laboratory has characterized an in vitro model of mast cell homeostasis, by which long-term exposure of murine bone-marrow-derived mast cells to the Th2-derived cytokines IL-3, IL-4, and IL-10, will induce downregulation of critical mast cell effector proteins such as Kit and FcεRI, followed by mast cell apoptosis. These data offer a novel role for Th2 cytokines, acting to both initiate and resolve mast cell activation and proliferation. Loss of these signals may contribute to a multitude of diseases, such as mastocytosis and allergy.

    Notes: Abstract A consequence of reducing early graft failure due to acute rejection has been that more patients are at risk of chronic rejection, something which has become an increasingly important cause of graft loss. We examine the graft survival rates and reasons for failure in our unit from 1981 to 1986. Patients were divided into two series according to treatment of acute rejection episodes. From 1983 onwards, by treating acute vascular (poor prognosis) episodes with antilymphocyte globulin (ALG), we have significantly improved the 6-month actuarial graft survival rate. However, the percentage of total graft failure due to chronic rejection in this second series has significantly increased. The need for greater understanding of the aetiology of chronic rejection, together with its present unsatisfactory treatment, is discussed.

    Notes: Abstract. In 1979, a mass poisoning of more than 2000 people occurred in central Taiwan due to consumption of rice-bran oil contaminated with PCBs and their heat-degraded byproducts. The incident was later referred to as Yucheng (oil disease). Serum samples from 56 women with the 1979 exposure were collected in February 1992 and analyzed for their contaminant content using sample enrichment and isotope dilution mass spectrometry. In most of the samples, levels of PCDFs and PCBs were detectable, and the median values of 2,3,4,7,8-PCDFs and 1,2,3,4,7,8-PCDFs were 1,030 and 2,220 ng/kg serum lipid, respectively. The median level of the total PCBs on a whole weight basis was 8,730 ng/kg. The PCB/PCDF concentrations in Yucheng women 14 years after the toxic exposure were still one to two orders of magnitude higher than controls. Concentrations of PCB levels in 1992 were positively correlated with the 1980–1981 measured PCB levels in these women and both PCBs and PCDFs were negatively correlated with the total duration when these women breast fed their children between 1979 and 1992. It is concluded that serum levels of congener-specific PCBs/PCDFs in exposed women are good indicators of previous exposure and may provide important information for more reliable estimation of dose-response relationship.
