    Notes: In the presence of retinoic acid, cultured human neuroblastoma SH-SY5Y cells grow processes indicative of neuronal differentiation. A voltage-gated Ca current is already present in undifferentiated cells. A gradual increase of the Ca current density occurs during cell differentiation. According to kinetic and pharmacological properties, Ca currents in differentiated cells are indistinguishable from those elicitable in undifferentiated cells and resemble features of the high-voltage activated currents present in mammalian neuronal cells. ω-conotoxin strongly depresses high-voltage activated currents, both in undifferentiated and in differentiated SH-SY5Y cells. Interestingly, the Ca agonist Bay K 8644 is effective, albeit with great variability from cell to cell, only in differentiated cells and only when barium is the current carrier through the Ca channels. A diversity of high-voltage activated Ca channels of distinct pharmacology has been recently observed in other kinds of neurons. This requires a redefinition of the role that voltage-dependent Ca channel subtypes can play in mammalian neurons.

    Notes: Abstract  The ability of action-potential-like waveforms (APWs) to attenuate opioid-induced inhibition of N-type Ca2+ channels was investigated in the neuroblastoma × glioma cell line NG108–15 using whole-cell voltage clamp methods. In in vitro differentiated NG108–15 cells, the opioid agonist [d-ala2]-methionine-enkephalin (DAME) reversibly decreased ω-conotoxin-GVIA-sensitive Ba2+ currents (N-type currents). Agonist-mediated inhibition of N-type currents could be transiently relieved by strong unphysiological depolarizing prepulses to +80 mV (facilitation). Significant facilitation was also achieved by conditioning the cell with a train of 15 APWs, which roughly mimicked physiological action potentials (1- to 6-ms-long depolarizations to +30 mV from a holding potential of –40 mV). The APW-induced facilitation depended on both conditioning pulse frequency and duration. Summation of the disinhibition produced by each APW was possible because reinhibition following repolarization to –40 mV was a much slower process (τ=88 ms) than the onset of facilitation at +80 mV (τ=7 ms). These results provide evidence that N-type Ca2+ channel facilitation may be a physiologically relevant process, and suggest that neuronal firing may relieve agonist-induced inhibition of N-type currents to an extent depending on both the shape of action potentials and the frequency of firing.

    Notes: Abstract Acetylcholine (Ach) reversibly reduces the high voltage-activated (HVA) calcium (Ca) current in hippocampal neurons. Pretreatment of the cells with pertussis toxin (PTX) abolishes the Ach effect, suggesting that PTX-sensitive GTP-binding regulatory proteins (G-proteins) are involved in the signal transduction mechanism that links Ach receptor activation to inhibition of Ca channel activity. This effect is mimicked by intracellular application of the nonhydrolyzable GTP analog GTPγS. Intracellular application of purified G-proteins restored the response to Ach in PTX-treated cells. Furthermore, Ach inhibits the Ca current independently of the presence of cyclic AMP and of the protein kinase C inhibitor H-7 and neither does the Ach effect on the Ca current seem to be correlated to a transient increase in intracellular Ca. Our results suggest that activation of the α-subunit of the PTX-sensitive G-protein could directly modulate the HVA Ca channel without involving second messenger systems.

    Notes: Abstract  Ca2+ channel modulation by the μ opioid agonist [d-Ala2, N-Me-Phe4, Gly5-ol]-enkephalin (DAGO) and the δ opiate agonists [d-Pen2, d-Pen5]-enkephalin (DPDPE) and [d-Ala2, d-Leu5]-enkephalin (DADLE) in cultured human neuroblastoma SH-SY5Y cells was investigated using the whole-cell variant of the patch-clamp technique. In SH-SY5Y cells, differentiated in vitro with retinoic acid, all agonists reversibly decreased high-voltage-activated, ω-conotoxin-sensitive Ba2+ currents in a concentration-dependent way. Inhibition was maximal with a 1 μM concentration of opiate agonists (76% with DAGO and 63% with δ agonists, when measured at 0 mV) and was characterized by a clear slow down of Ba2+ current activation at low test potentials. Both inhibition and slow down of activation were attenuated at more positive potentials, and could be partially relieved by strong conditioning depolarizations. Current suppression operated by both μ and δ agonists was prevented by pre-treatment of the cells with pertussis toxin. No sign of additivity was observed when δ agonists were applied to cells that were maximally activated by DAGO, suggesting that a common mechanism, involving the same type of modulating molecule, is responsible for Ca2+ channel inhibition promoted by activation of μ and δ opioid receptors in SH-SY5Y cells.

    Notes: Summary Multiblock-(A-B)n-copolymers containing polyester segments together with perfluoropolyether (PFPE) segments have been prepared by polymerizing dimethyl terephthalate (DMT), or an equimolar mixture of DMT and dimethyl isophthalate (DMI), with ethylene glycol (EG) in the presence of different telechelic perfluoropolyethers in various amounts (5–30 wt%), using Tl(OBu)4 as the catalyst. Fomblin ZDEAL (a-COOCH3 terminated PFPE), Fomblin ZDOL (a PFPE having-CH2OH terminal groups), and Fomblin ZDOLTX (a PFPE having-CH2O(CH2CH2O)yH terminal groups) were used as telechelic PFPEs. Fomblin ZDOLTX gave the best results: the highest yield in block copolymer (less sensitive to hydrolysis with respect to block copolymers prepared from the other PFPEs), a longer average length of polyester segments, and a relatively low fraction of PFPE lost by distillation during polymerization.

    Notes: Abstract Nitrogen fertilization in orchards of Emilia-Romagna Region, (Italy) was based in the past on excessive, not split, applications often supplied late in winter; the NUE (Nitrogen Use Efficiency) was therefore low and the risk of nitrate leaching was high. This paper summarizes the studies conducted in the last 10 years at the Department of Horticulture and Forestry of the University of Bologna aimed to develop a more rational use of nitrogen in orchards and vineyards. Root escavation of mature trees revealed that the use of localized irrigation (drip or microjet) causes a concentration of roots in the area wetted by the emitters. In such a situation, band applications of N to the tree row may allow a reduction of amounts of N fertilizer, while widespread applications, especially if the orchard soil is tilled, lead to an accumulation of nitrates in the alley. Results of several field trials where increasing N rates were applied indicate that the kind of response to N supply depends on the presence in soil of natural sources of nitrogen. This fact clearly stresses the necessity of evaluate the N status of an orchard before N fertilization. Rapid estimation of leaf chlorophyll by portable instruments is a promising index of leaf N concentration, only provided that calibration is made for each cultivar. A method, currently under testing in orchards and vineyards of Emilia-Romagna, is proposed here to adjust N fertilizer rates to the demand of the crop and to the level of available N in soil as determined in soil or soil solution samples.
